    Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Use of ozone in production chain of high moisture Mozzarella cheese

    This work aimed to evaluate ozone effectiveness in reducing viable spoilage bacteria load throughout high moisture (HM) Mozzarella cheese-making process. At first, Mozzarella cheese samples were packaged with ozonated water (2mg/L), stored at low temperature and monitored during shelf life. In a following phase cheese samples were put, before packaging, in direct contact with ozonated water at ozone concentrations of 2, 5 and 10mg/L for 60min. Then, gaseous ozone at concentrations of 10, 20 and 30mg/m3 for different times was tested. In these experiments ozone was not effective in surface microbiological decontamination of cheeses. In all cases, there was no increase in the formation of primary and secondary lipid oxidation products.Finally, we treated only the cooling water, artificially contaminated with Pseudomonas spp. strains, obtaining a reduction of about four Log cfu/mL in the microbial load. HM Mozzarella cheeses cooled with pre-treated water were characterized by a low microbial load and an increased shelf life.We concluded that ozone can be successfully applied in Mozzarella cheese processing only to decontaminate water contaminated by potential spoilage bacteria before that it comes into contact with the product. Industrial relevance: HM Mozzarella cheese is a very popular fresh product. Microbiological contamination that occurs after the stretching step when HM Mozzarella cheese comes into contact with cooling water and preserving liquid heavily affects its shelf-life. For this reason, it was decided to investigate whether ozone applications can be extended to HM Mozzarella cheese-making process, as already occurs in other food industry sectors. This study demonstrates that water acting as carrier of spoilage microorganisms can be successfully treated with ozone in order to reduce microbial cheese contamination in each processing step after stretching and shaping. © 2013 Elsevier Ltd

    Effects of ozone processing on chemical, structural and functional properties of whey protein isolate

    High concentration of gaseous ozone was used to treat whey protein isolate (WPI) powder for different times ranging from 30 to 480. min. The aim of the present work was to study the changes in protein structure and evaluate their consequences on selected functional properties. Surface hydrophobicity, free sulphhydryl groups, turbidity, FTIR, SDS-PAGE, HPLC analysis are performed to evaluate the chemical and structural effects of ozone. The effect of ozonation on the solubility and the foaming properties of proteins are also determined. Results show a reduction of free sulphhydryl groups and an increase of surface hydrophobicity, indicating a self-rearrangement in the protein structure following ozonation. Thus, ozonation allows creation of a more flexible structure without formation of a strong network of disulphide bond or aggregations, which is corroborated by the turbidity analysis and SDS-PAGE.     Cystic fibrosis (CF) transmembrane regulator protein (CFTR) gene is undoubtedly the main genetic factor involved in the modulation of CF phenotype. However, other factors such as human defensins and the genes encoding for these antimicrobial peptides have been hypothesized as possible modifiers influencing airways infection in CF patients, but their role in the pathogenesis of lung disease is still debated. Since DEFB1 gene encoding for human beta-defensin 1 displays features such as antimicrobial or chemotactic activity playing a role in inflammation, it has been considered as a possible candidate CF modifier gene. We analysed three single nucleotide polymorphisms (SNPs) in the 5'-untranslated region of the DEFB1 gene (namely g-52G>A, g-44C>G and g-20G>A) in a group of 62 CF patients from North Eastern Italy, and in 130 healthy controls, with the aim of verifying the possible association of these functional SNPs with the pulmonary phenotype of CF patients. DEFB1 SNPs have been genotyped by using Taqman allele-specific fluorescent probes and a real-time PCR platform. No significant differences were found for allele, genotype and haplotype frequencies of DEFB1 g-52G>A, g-44C>G and g-20G>A SNPs in CF patients stratified for Pseudomonas aeruginosa infection, as well as in patients with a severe and mild clinical phenotype or in patients stratified for CFTR genotypes. DEFB1 allele, genotype and haplotype frequencies of CF patients globally considered were similar to those of healthy controls. Our findings are discordant with respect to another recent study performed on CF patients coming from Southern Italy, probably due to different ethnicity of the patients

    Jersey Homesteads (later the Borough of Roosevelt) was established in the 1930s as an agro-industrial cooperative community. It was established specifically for urban Jewish garment workers, many of whom had emigrated from Europe. President John F. Kennedy sent a telegram to the citizens of Roosevelt, New Jersey, apologizing for not being able to attend the memorial dedication in honor of former President Franklin Delano Roosevelt. (Jersey Homesteads became Roosevelt in 1945 in honor of the president.) President Kennedy expressed his gratitude to the people of Roosevelt for constructing the memorial, and commented that it will serve as a constant reminder of Roosevelt's good works
