The role of educative thought in the life and work of Antonio Gramsci
Many philosophers have propounded a vision of an improved society, what distinguishes Antonio Gramsci is his continuous effort to make it happen by understanding the process in order to put into practice. Gramsci's conviction about the importance of educative development came from both theory and experience. While there has been considerable examination of Gramsci's work in relation to the Prison Notebooks, this study will seek to address a lacuna in Gramsci scholarship. Using Gramsci's philological method, I analyse Gramsci's pre-prison activity; his pre-prison articles and letters, which, together with his letters from prison, formed part of his educative mission. This educative process was necessary, in order to construct a new party which would develop a collective will, collaboratively, with the masses.In this study therefore, I explore the contexts and formative experiences of the first part of his life together with the intellectual sources from which Gramsci developed his later theories, making central hitherto underemphasised connections between them which informed his writing and ideas. I intend to illustrate that Gramsci's underlying purpose in his writing, and political activity, was not only practical, on how to create a new socialist ruling class, but also educative in forming the mindset and values of his comrades. So that in addition to outlining his vision of a new order, he implicitly guided or explicitly explained the processes by which the necessary changes in social relations and moral climate could be made in order to achieve it. Each person had to engage with the values of the new order so that each could contribute to the construction of a new robust state. It was essential to build a hegemony at the most profound level, one which was dependent on collective understandings and a collective will

RNAi-Based Strategies for Cyclooxygenase-2 Inhibition in Cancer
Cyclooxygenase-2 (COX-2) enzyme has been involved in the tumorigenesis and in the progression of colorectal cancer (CRC). The use of traditional nonsteroidal anti-inflammatory drugs (NSAIDs) or selective COX-2 inhibitors has been proposed for the prevention and the treatment of this relevant neoplastic disease. In the light of an innovative alternative to these pharmacological approaches, we review here the possible strategies to achieve a strong and selective inhibition of COX-2 enzyme by using the mechanism of RNA Interference (RNAi) targeted against its mRNA. Anti-COX-2 siRNA molecules (siCOX-2) can be generated in CRC cells from short hairpin RNA (shRNA) precursors, delivered in vitro by a retroviral expression system, and induce a significant and stable silencing of overexpressed COX-2 in human colon cancer cells. As a safer alternative to viral approach, nonpathogenic bacteria (E. coli) can be engineered to invade eukaryotic cells and to generate siCOX-2 molecules in cancer cells. Moreover, the involvement of miRNAs in COX-2 posttranscriptional regulation opens up the possibility to exploit an endogenous silencing mechanism to knockdown overexpressed COX-2. Thus, these recent strategies disclose new challenging perspectives for the development of clinically compatible siRNA or miRNA capable of selectively inhibiting COX-2 enzyme.</jats:p

Antonio Rosminis Principles of moral science: translation of an Italian philosophical work to Portuguese
Este trabalho se compõe de duas partes. Primeiramente, um estudo introdutório sobre o autor, Antonio Rosmini, sobre sua obra filosófica e, mais especificamente, sobre o livro Principi della scienza morale, que, na segunda parte, traduzimos para o português. Na introdução, expomos brevemente o sistema filosófico rosminiano e o contextualizamos no âmbito histórico-filosófico. Além disso, apresentamos o percurso por nós desenvolvido para obter uma tradução filologicamente correta do texto original, incluindo um breve glossário dos termos filosóficos empregados pelo autor.This work is composed of two parts. Firstly, there is an introductoty study on the author, Antonio Rosmini, on his phylosophical system and, more specifically, on the book Principi della scienza morale, which we have translated to Portuguese, in the second part. In the introduction, we expose briefly the rosminian phylosofical system and we place it in its historical-phylosofical context. Furthermore, we show the path weve followed to reach a phylologically correct translation from the original text, including a brief glossary of phylosophical terms employed by the author
Selective inhibition of prostacyclin synthase activity by rofecoxib
The development of cyclooxygenase-2 (COX-2) selective inhibitors prompted studies aimed at treating chronic inflammatory diseases and cancer by using this new generation of drugs.Yet, several recent reports pointed out that long-term treatment of patients with COX-2 selective inhibitors (especially rofecoxib) caused severe cardiovascular complicances. The aim of this study was to ascertain whether, in addition to inhibiting COX-2, rofecoxib may also affect prostacyclin (PGI2) level by inhibiting PGI2 forming enzyme (prostacyclin synthase, PGIS). In order to evaluate if selective (celecoxib, rofecoxib) and non-selective (aspirin, naproxen) anti-inflammatory compounds could decrease PGI2 production in endothelial cells by inhibiting PGIS, we analyzed the effect of anti-inflammatory compounds on the enzyme activity by ELISA assay after addition of exogenous substrate, on PGIS protein levels by Western blotting and on its subcellular distribution by confocal microscopy. We also analyzed the effect of rofecoxib on PGIS activity in bovine aortic microsomal fractions enriched in PGIS. This study demonstrates an inhibitory effect of rofecoxib on PGIS activity in human umbilical vein endothelial (HUVE) cells and in PGIS-enriched bovine aortic microsomal fractions, which is not observed by using other anti-inflammatory compounds. The inhibitory effect of rofecoxib is associated neither to a decrease of PGIS protein levels nor to an impairment of the enzyme intracellular localization. The results of this study may explain the absence of a clear relationship between COX-2 selectivity and cardiovascular side effects. Moreover, in the light of these results we propose that novel selective COX-2 inhibitors should be tested on PGI2 synthase activity inhibition
Su una (presunta) lettera di Luigi Credaro ad Antonio Labriola
Il saggio rappresenta un ulteriore approfondimento dei rapporti fra Antonio Labriola e Luigi Credaro. Il movente è rappresentato dal V e ultimo volume del Carteggio del filosofo cassinate (Napoli, Bibliopolis, 2006) e dall’interpretazione che il curatore, Stefano Miccolis (noto studioso labrioliano, purtroppo prematuramente scomparso nel 2009) espone nel testo di una minuta di Credaro, conservata nel Fondo intitolato al Valtellinese all’Archivio Centrale dello Stato (b. n. 1, fasc. 5, Corrispondenza privata 1902), nella quale lo stesso Credaro ringrazia il destinatario, non precisato, per il ruolo decisivo recitato nell’approvazione da parte del Consiglio della Facoltà di Filosofia e Lettere della “Sapienza” romana della sua richiesta di trasferimento dall’Università di Pavia sulla cattedra, già labrioliana, di Pedagogia (27 maggio 1902). L’individuazione del destinatario in Antonio Labriola, proposta da Miccolis, è contestata dall’autore, rinvenendo nella chiusura della minuta, nella quale Credaro esprime, con la sua “riconoscenza”, “la viva soddisfazione di diventare Collega di Lei, che mi fu maestro illuminato e affettuoso”, una chiara indicazione (confermata dal raffronto con la conclusione di L’insegnamento universitario della pedagogia, la prima Prelezione del valtellinese alla Regia Università di Roma, letta il 15 gennaio 1903) che la lettera era in realtà indirizzata a Giacomo Barzellotti (1844-1917), storico della filosofia di matrice neokantiana-spiritualistica, del quale Credaro aveva seguito da studente il corso di Filosofia morale all’Università di Pavia. Tale disamina critica è funzionale, per contrasto, al rafforzamento della tesi già in precedenza formulata dall’autore che esclude l’esistenza di significativi e approfonditi rapporti fra Labriola e Credaro antecedenti alla chiamata a Roma del Valtellinese – circostanza nella quale si sarebbe realizzata, in sostanza, una coincidenza e concordanza di interessi – e che l’avrebbero in qualche modo ascritta a una ben precisa, motivata volontà del Cassinate, magari da inserire in un ben preciso, complessivo suo “disegno” politico-accademico; ma il saggio non si limita a tale serrata pars destruens e insiste, nel prosieguo, con numerosi spunti, sull’ulteriore tesi dell’instaurarsi di un significativo rapporto ideale fra i due studiosi successivo all’approdo romano di Credaro, nel senso, per l’esattezza, di un influsso del Labriola herbartiano, e del suo concetto della libertà morale, sul Valtellinese, semplicemente adombrato nel contributo del 2006.In the 5th, last volume of Carteggio by Antonio Labriola (Napoli, Bibliopolis, 2006) the editor and author, Stefano Miccolis (well known Labriola researcher, untimely died in 2009)reproduce contains the text of a handwritten draft that he also delivered at the Central State Archive, (b. n. 1, file. 5, Corrispondenza privata 1902), in which Credaro thanks the recipient of the letter, unknown, for his efforts and for having ensured approval of Credaro's transfer application to La Sapienza, occurred in May 27, 2002). The recipient of this letter is, some argue, Antonio Labriola. The author objects to this interpretation and points out that at the end of the letter there is a clear indication (confirmed by comparing it to the conclusion of Teaching pedagogy at the University, the first Pre-lesson provided by Credaro at the Rome University, on January 15, 1903) that the letter was indeed addressed to Giacomo Barzellotti (1844-1917), a historian of philosophy with a Neokantian-Spiritualist approach, of which Credaro had already followed the Moral Philosophy course in Pavia. This analysis is important, with a contrast approach, to provide further support to the thesis the author had already expressed, that is that there had not been any deep and significant exchange between Credaro and Labriola before the former was transferred to Rome; however the essay does not stop with this hectic pars destruens but rather insists on the establishing of a significant intellectual and academic relationship (or rather, an influence of the Herbartian Labriola and of his concept of moral freedom, on Credaro) after the latter's move to Rome

RNA interference as a key to knockdown overexpressed cycloxygenase 2 gene in tumour cells
Silencing those genes that are overexpressed in cancer and contribute to the survival and progression of tumour cells is the aim of several researches. Cyclooxygenase-2 (COX-2) is one of the most intensively studied genes since it is overexpressed in most tumours, mainly in colon cancer. The use of specific COX-2 inhibitors to treat colon cancer has generated great enthusiasm. Yet, the side effects of some inhibitors emerging during long-term treatment have caused much concern. Genes silencing by RNA interference (RNAi) has led to new directions in the field of experimental oncology. In this study, we detected sequences directed against COX-2 mRNA, that potently downregulate COX-2 gene expression and inhibit phorbol 12-myristate 13-acetate-induced angiogenesis in vitro in a specific, nontoxic manner. Moreover, we found that the insertion of a specific cassette carrying anti-COX-2 short hairpin RNA sequence into a viral vector (pSUPER.retro) greatly increased silencing potency in a colon cancer cell line (HT29) without activating any interferon response. Phenotypically, COX-2 deficient HT29 cells showed a significant impairment of their in vitro malignant behaviour. Thus, the retroviral approach enhancing COX-2 knockdown, mediated by RNAi, proved to be an useful tool to better understand the role of COX-2 in colon cancer.
