HIV-1-derived lentiviral vectors directly activate plasmacytoid dendritic cells, which in turn induce the maturation of myeloid dendritic cells
Lentiviral vectors (LV) can induce type I interferon (IFN I) production from murine plasmacytoid dendritic cells (pDC), but not myeloid (my)DC. Here, we investigated whether this mechanism is conserved in human DC. MyDC and pDC were isolated from peripheral blood and transduced with increasing vector concentrations. Compared with in vitro differentiated monocyte-derived DC, the transduction efficiency of peripheral blood DC was low (ranging from <1% to 45%), with pDC showing the lowest susceptibility to LV transduction. Phenotype and function of myDC were not directly modified by LV transduction; by contrast, pDC produced significant levels of IFN-α and tumor necrosis factor-α. pDC activation was dependent on functional vector particles and was mediated by Toll-like receptor 7/9 triggering. Coculture of myDC with pDC in the presence of LV resulted in myDC activation, with CD86 up-regulation and interleukin-6 secretion. These findings demonstrate that the induction of transgene-specific immunity is triggered by an innate immune response with pDC activation and consequent myDC maturation, a response that closely resembles the one induced by functional viruses. This information is important to design strategies aimed at using LV in humans for gene therapy, where adverse immune responses must be avoided, or for cancer immunotherapy, where inducing immunity is the goal.Maura Rossetti, Silvia Gregori, Ehud Hauben, Brian D. Modelling individual preferences for environmental policy drivers: Empirical evidence of Italian lifestyle changes using a latent class approach
Degraded air quality severely affects the health of citizens worldwide. The design of effective policies requires exploring public preferences for environmental and air quality policy instruments. Within the EC-FP7 SEFIRA project, using a choice experiment that stresses the trade-offs between attributes, this study investigates public preferences for environmental policy drivers in Italy. The main objective is to investigate the role played by selected policy drivers in determining policy preferences, complemented by elasticity and willingness to pay estimations. Preference heterogeneity and the role of socio-economic and attitudinal variables are explored with a latent class model over 2400 respondents sampled across Italy. The results allow identifying the different role played by the policy drivers across the classes. It emerged that most of the respondents (43%) are particularly sensitive to the cost components (cost sensitive respondents). The remaining respondents instead show an important sensitivity towards personal engagement in term of changes in the mobility and eating habits (lifestyle-change sensitive respondents). However, while 29% of them perceive these habits’ changes as negatively impacting on the personal utility, the other 28% of respondents translate the potential changes in the habitual behaviour of driving and eating as environmental and health benefits. Based on the modelling results, potential policies are simulated reporting respondents’ reaction to selected scenarios. It shows the crucial role played by reduction of premature deaths due to atmospheric pollution and measure cost.

Meet the Inbetweeners : Exploring the Quality and Stregth in the Mafia Capitale network in Rome
This chapter looks at the network of Mafia Capitale, in particular focusing on the quality of ties among the World Above, the World Below and the in-betweeners. It focuses on the characteristics of a modern mafia group; this could open up new debates around organised crime groups’ strategies of infiltration in the public sector, including outside of Italy. The chapter discusses the traditional literature on social network analysis, organised crime and criminal networks. It explores the types of relationships among the main players of the Mafia Capitale network by performing a content and discourse analysis of intercepted communications as transcribed in the main judicial documents presented by the Antimafia prosecutors in Rome. The chapter provides an exploration of the Mafia Capitale network as presented by the authorities in the two years following the arrests. It describes the state of the trial and its outcome and explains the literature on network analysis relevant to a qualitative analysis of terrorist group

η-η′ mixing determination from semileptonic D+ (s) and B+ meson decays

(s) → η(′)ℓ+νℓ and B+ → η(′)ℓ+νℓ decays. A controlled parametrization of the participating hadronic form factor, D(s) → η(′) and B → η(′), in combination with experimental measurements, has allowed us to determine ϕ = 40.6(4.8)◦ and ϕ = 42.3(3.7)◦ from the analyses of differential decay width distributions and ratios of branching ratios, respectively. Our results are in good agreement with a recent determination from the Lattice QCD Extended Twisted Mass Collaboration, ϕ = 39.3(2.0)◦, and larger than the naive estimate we calculate from Chiral
Perturbation Theory at leading order ϕ = 33.5◦
The case for global regulation of carbon capture and storage and artificial intelligence for climate change
This article investigates the trends in the global law-making process as it relates to carbon capture and storage (CCS) and associated technology. It detects regulatory deficiencies and makes a case favouring robust global regulation of such technology in realizing climate aspirations. CCS is recognized as an indispensable ally in the worldwide enterprise that seeks to limit carbon dioxide emissions. This article probes the relatively new and developing area of international regulation of the process of CCS and related technology, in particular Artificial Intelligence (AI), a dimension that promises noteworthy results on the climate change front. It presents a case for the broader deployment of the CCS technology - including more advanced AI options - to enhance in a more radical fashion the prospect of meeting global climate goals and thus stabilising global temperature (that is, achieving net zero CO2 emissions globally in the early 2050s), identifies the need for new and revised sets of rules to govern the development, design and deployment of such technology, and argues in favour of the applicability of existing general sustainable development principles
