Results of the 2023 annual glaciological survey conducted on Italian glaciers are here presented. About two-hundred volunteers, assisted by numerous collaborators, operated last year in the three Alpine sectors (Piemonte - Valle d’Aosta, Lombardy and Triveneto) and in the Apennines (Calderone Glacier, Gran Sasso Group) observing two hundred and nine glaciers. One hundred and thirty glaciers were observed in the Piemonte - Valle d’Aosta sector while frontal variation was measured at the snout of fifty-nine glaciers. Twenty-nine and fifty glaciers were visited in the Lombardy Sector and in the Triveneto Sector, respectively (twenty-two and forty-three of which were measured, respectively). Mass balance measurements were conducted on nineteen Italian glaciers during the 2022-2023 hydrological year: four glaciers in the Western Alps (Piemonte - Valle d’Aosta sector), fourteen in the Eastern Alps (three in the Lombardy Sector, the other glaciers in the Triveneto Sector) and one in the Central Apennines

    Bacterial L-asparaginases are amidohydrolases (EC 3.5.1.1) capable of deaminating L-asparagine and, with reduced efficiency, L-glutamine. Interest in the study of L-asparaginases is driven by their use as biodrugs for the treatment of acute lymphoblastic leukemia. Here, we report for the first time the description of the molecular structure of type II asparaginase from Escherichia coli in complex with its secondary product, L-glutamate. To obtain high-quality crystals, we took advantage of the N24S variant, which has structural and functional features similar to the wild-type enzyme, but improved stability, and which yields more ordered crystals. Analysis of the structure of the N24S-L–glutamate complex (N24S–GLU) and comparison with its apo and L-aspartate-bound form confirmed that the enzyme-reduced catalytic efficiency in the presence of L-glutamine is due to L-glutamine misfitting into the enzyme-binding pocket, which causes a local change in the catalytic center geometry. Moreover, a tight interaction between the two protomers that form the enzyme active site limits the capability of L-glutamine to fit into (and to exit from) the binding pocket of E. coli L-asparaginase, explaining why the enzyme has lower glutaminolytic activity compared to other enzymes of the same family, in particular the Erwinia chrysanthemi one

    HAP1 loss in L-asparaginase resistance

    In this issue of Blood, Lee et al identify huntingtin associated protein 1 (HAP1) loss as a new marker of L-asparaginase resistance in acute lymphoblastic leukemia (ALL) and provide evidence for the pathway involved. They discovered that HAP1 is essential for the formation of the ternary complex with huntingtin (Htt) and inositol 1,4,5-triphosphate receptor (InsP3R) and that its loss impairs the L-asparaginase– mediated increase of cytosolic Ca21 needed for triggering apoptosis (see figure). Their data were confirmed by specific knockdown of HAP1 in SEM cells and by measurement of both endoplasmic reticulum–released Ca21 and external Ca21 influx.
