Because of the increasing interest to test it in pre-clinical and clinical researches for assessing its health benefits, we here evaluate the interactions between TQ and human serum albumin (HSA), a possible carrier of this drug in vivo. Binding to HSA was studied using different spectroscopic techniques. Fourier transform infrared (FT-IR) and circular dichroism (CD) spectroscopies suggest that the association between TQ and HSA does not affect the secondary structure of HSA. Using fluorescence spectroscopy, one mole of TQ was found to bind one mole of HSA with a binding constant of 2.39 ± 0.2 104 M−1. At 25 °C (pH 7.4), van’t Hoff’s enthalpy and entropy that accompany the binding were found to be −10.24 kJ/mol−1 and 45 J/mol−1 K−1 respectively. The thermodynamic analysis of the TQ-HSA complex formation shows that the binding process is enthalpy driven and spontaneous, and that hydrophobic interactions are the predominant intermolecular forces stabilizing the complex. Furthermore, displacement experiments using warfarin and ibuprofen indicate that TQ could bind to site I of HSA, which is also in agreement with the results of the molecular modeling study

    L’ambito d’indagine di questo lavoro attiene al diritto penale dell’informatica. Si propone infatti un’analisi ragionata delle teorie e prassi sviluppatesi, in tempi recenti, con riferimento alla tutela dei diritti della persona, alla luce delle profonde innovazioni che ha portato con sé l’aumento esponenziale di tecnologia e automazione nel vivere quotidiano di ciascuno di noi. Entro tale perimetro, si inizia approfondendo in linea teorica i principali beni giuridici ascrivibili all’essere umano nel mondo digitale, poi affrontando diacronicamente sia l’evoluzione dell’informatica che quella del diritto penale che la governa (Capitolo Primo). Fissati in questo senso sia gli obiettivi che le linee d’indagine, si passa allora al dettagliato esame dei c.d. “reati informatici in senso stretto”, cioè delle previsioni che il Legislatore ha introdotto nell’ordinamento a fronte del dilagante fenomeno dei crimini in materia tecnologica, dando conto della loro applicazione giurisprudenziale lungo l’ultimo decennio (Capitolo Secondo). Con la medesima cadenza strutturale sono in seguito passati in rassegna anche i c.d. “reati informatici in senso ampio”, categoria che raggruppa numerose fattispecie tradizionali, divenute rilevanti in ambito tecnologico sia per l’espansione naturale dei beni giuridici ivi tutelati, sia a causa della sostanziale inerzia in cui è caduto il Legislatore, non proponendone una rivisitazione alla luce delle novità e delle criticità presentate dal ciberspazio (Capitolo Terzo). In conclusione, si propone al lettore una nuova e diversa visione del tema “reato informatico”, partendo da un concetto – quello di Io digitale – costruito con finalità sistematiche: si può allora valutare in quest’ottica l’adeguatezza degli strumenti di cui dispone, oggi, il diritto penale, nonché l’uso che ne viene fatto. Al termine dell’analisi, si propongono al lettore alcuni profili di possibile adeguamento della normativa codicistica, nel rispetto dei principi fondanti della materia penale (Capitolo Quarto).This work aims at evaluating the criminal-law approach to the digital age. Between the analysis of the theoretical constructs provided by the best Italian doctrine and the jurisprudential application of the law, we try to understand if everyone’s fundamental rights are protected by today’s complex criminal system, in light of the profound innovations that tecnology has brought to the society. First of all, we take on a sweeping journey of the basic legal interests that anyone is entitled to in the digital world (personal identity, honour, privacy and freedom), along with a historic view of the evolution both of computer science and computer crimes. On these basis, we proceed to examine both the “strictly cyber-related” provisions, or laws that have been enacted specifically for computer-related crimes, and the “wider cyber-related” rules, that have seen themselves applied to computer-related crimes, although they never received an update to better comply with the so-called digital age. The research conduct us to depict a clearer view of how today’s computer-related provisions oversee and protect people in the technological world: looking through a different point of view (the so-called “Digital Self”) it emerges that the criminal system should update itself as soon as possibile, by systematically enhancing its set of rules as well as by providing the judge with more specific provisions. This approach is ultimately the only option to adequately protect anyone’s Digital Self, without violating the fundamental principles of criminal law

    Gelonin from the Indian plant Gelonium multiflorum belongs to the type I ribosome-inactivating proteins (RIPs). Like other members of RIPs, this toxin glycoprotein inhibits protein synthesis of eukaryotic cells; hence, it is largely used in the construction of immunotoxins composed of cell-targeted antibodies. Lysosomal degradation is one of the main issues in targeted tumor therapies, especially for type I RIP-based toxins, as they lack the translocation domains. The result is an attenuated cytosolic delivery and a decrease of the antitumor efficacy of these plant-derived toxins; therefore, strategies to permit their release from endosomal vesicles or modifications of the toxins to make them resistant to degradation are necessary to improve their efficacy. Using infrared spectroscopy, we thoroughly analyzed both the secondary structure and the thermal unfolding of gelonin. Moreover, by the combination of two-dimensional correlation spectroscopy and phase diagram method, it was possible to deduce the sequence of events during the unfolding, confirming the typical characteristic of the RIP members to denature in two steps, as a sequential loss of tertiary and secondary structure was detected at 58 °C and at 65 °C, respectively.     The structure of thioredoxin from Alicyclobacillus acidocaldarius (previously named Bacillus acidocaldarius ) (BacTrx) and from Escherichia coli ( E. coli Trx) was studied by Fourier-transform IR spectroscopy. Two mutants of BacTrx [Lys(18)-->Gly (K18G) and Arg(82)-->Glu (R82E)] were also analysed. The data revealed similar secondary structures in all proteins, but BacTrx and its mutants showed a more compact structure than E. coli Trx. In BacTrx and its mutants, the compactness was p(2)H-dependent. All proteins revealed the existence of a molten globule-like state. At p(2)H 5.8, the temperature at which this state was detected was higher in BacTrx and decreased in the different proteins in the following order: BacTrx>R82E>K18G> E. coli Trx. At neutral or basic p(2)H, the molten globule-like state was detected at the same temperature in both BacTrx and R82E, whereas it was found at the same temperature in all p(2)Hs tested for E. coli Trx. The thermal stability of the proteins was in the following order at all p(2)Hs tested: BacTrx>R82E>K18G> E. coli Trx, and was lower for each protein at p(2)H 8.4 than at neutral or acidic p(2)Hs. The formation of protein aggregates, brought about by thermal denaturation, were observed for BacTrx and K18G at all p(2)Hs tested, whereas they were present in R82E and E. coli Trx samples only at p(2)H 5.8. The results indicated that a single mutation might affect the structural properties of a protein, including its propensity to aggregate at high temperatures. The data also indicated a possible application of Fourier-transform IR spectroscopy for assessing molten globule-like states in small proteins

    Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship
