    Monitoring the homemade fermentation of readymade malt extract using the SCiO NIR sensor: A convergence of technology and tradition

    Miniaturized near-infrared (NIR) instruments are launched on the market to satisfy the need of both researchers and consumers for quick ways of analysing stuff, especially in the food field. Their portability and ease of operation are at the centre of the “do-it-yourself” idea behind providing virtually anyone with the ability of making analytical measurements on their own. In this study, we highlighted the convergence of technology and tradition in two seemingly disparate domains: spectroscopy and homebrewing. Homebrewing is in fact a leisure activity which conceptually shares the do-it-yourself approach: the consumer becomes the producer of his/her own beer. Hence, in our study, we investigated the analytical possibilities provided by a handheld NIR spectrometer to monitor the homemade fermentation process of a commercial malt extract, over the course of one week. The spectroscopic data, acquired using the SCiO sensor (Consumer Physics), were analysed via Principal Component Analysis (PCA) to investigate temporal trends and relationships with brewing parameters. Our findings reveal that discernible trends, reflective of brewing stage progression and temperature variations, can be captured and unveiled with simple data analysis approaches. At the same time, the detection of scattering effects that can be attributed to bubble formation on the spectrometer's acquisition window confirm the need for robust and reasoned experimental procedures, but also for proper data preprocessing methods. This rather simple and basic analytical approach could provide the homebrewer the possibility to qualitatively monitor the advancement of the brewing process

    La principale trasformazione del caffè prima della commercializzazione è rappresentata dalla tostatura, processo che influenza pesantemente le caratteristiche organolettiche della bevanda. A livello industriale, per soddisfare le aspettative dei consumatori legate ad una qualità costante, il processo richiede un controllo molto attento. Il monitoraggio del colore del caffè durante la tostatura potrebbe costituire uno strumento molto importante per valutare in tempo reale le performance del processo e garantirne la riproducibilità. In questo lavoro viene descritta l'applicazione di un sistema on-line per il monitoraggio del processo di tostatura del caffè, eseguito su un tostino da laboratorio. Il controllo di processo è realizzato utilizzando una sonda nel vicino infrarosso (Bruker Optics, Germania) posta all'interno del tostino (Probat, Germania). E' stata esaminata l'influenza sul colore di diverse variabili sperimentali: la varietà di caffè (Arabica, Robusta e miscele 50/50 p/p), la temperatura (sono stati considerati 4 diversi valori), e l'umidità iniziale dei chicchi; sono stati acquisiti un totale di 800 spettri durante 65 esperimenti di tostatura. Per ottenere il modello di predizione sul colore di tostatura viene utilizzata la Partial Least Square; sono stati utilizzati i parametri Hotelling T2 e i residui Q per valutare la presenza di campioni outliers. Il modello ottenuto presenta una buona capacità predittiva per la determinazione del colore di tostatura con un RMSECV pari all'1,5 U.A. (in Fig. 1 è riportato il grafico del valore predetto vs. reale del colore). Questo studio conferma l'applicabilità della spettroscopia NIR per il monitoraggio on-line del processo di tostatura del caffè. Dopo un'ottimizzazione del metodo si prospetta l'implementazione di questa tecnologia su scala industriale per automatizzare il controllo dei parametri del process

    Exploring NIR spectroscopy data: A practical chemometric tutorial for analyzing freeze-dried pharmaceutical formulations

    Chemometrics tools are of fundamental importance for data analysis in the pharmaceutical field, especially with the increasingly strong assertion of the Process Analytical Technologies (PAT). In fact, analytical technologies such as Near-Infrared or Raman spectroscopies generate a lot of data, the spectra, that must be analyzed in a proper way. Typically, it is quite difficult to deeply understand the information hidden within the raw data. Therefore, careful, and efficient data exploration is needed to highlight the chemical and physical features of the analyzed samples. Here, a tutorial on all the fundamental steps and concepts needed to perform a proper data analysis based on a case-study of different freeze-dried formulations in the pharmaceutical field is proposed. The data analysis pipeline begins with the dataset explanation, to better point out the main known differences and similarities among the investigated formulations. After the first step of data preprocessing, Principal Component Analysis (PCA), Partial Least Squares (PLS) for regression, and Partial Least Squares-Discriminant Analysis (PLS-DA) for classification are presented and applied to show how to obtain deep comprehension of the real-case NIR dataset at hand. The experimental results demonstrate that trends related to increasing levels of sucrose and/or arginine, as well as distinct clusters related to the sample type and to the operator who conducted the analysis can be found and modelled in the example data. The tutorial aims at providing clear practical steps to conduct a robust data analysis, starting from the extraction and organization of the raw data, up to building more advanced predictive models (regression and classification). At each step some key questions are asked and answered to stimulate critical thinking in the reader. Also, commented MATLAB scripts are provided together with the real-case example NIR data, so that anyone could reproduce the whole data analysis in the tutorial, and try first hand to work with the data

La valutazione del colore, cioè l'imbrunimento del chicco, è utilizzato in modo efficace per definire il grado di tostatura come indicatore delle performance del processo e questa misura può essere effettuata utilizzando la spettroscopia NIR. Lo scopo di questo lavoro è quello di valutare l'applicabilità della spettroscopia nel vicino infrarosso per monitorare on-line il colore dei chicchi di caffè durante il processo di tostatura. Il controllo di processo è realizzato utilizzando una sonda nel vicino infrarosso (emissione head, Bruker Optics, Germania) posta all'interno di un tostino da laboratorio (Probat, Germania). Sono stati acquisiti un totale di 800 spettri, durante 65 esperimenti di tostatura, pianificati mediante un experimental design che ha tenuto conto dell'influenza sul colore di diverse parametri sperimentali. Tra queste variabili: la varietà di caffè (Arabica, Robusta e miscele), la temperatura di processo (155°C, 160°C, 170°C e 180°C). Per ottenere il modello di predizione sul colore di tostatura viene utilizzata la PLS; sono stati utilizzati i parametri Hotelling T2 e i residui Q per valutare la presenza di campioni outliers. Il modello ottenuto presenta una buona capacità predittiva per il colore di tostatura con un RMSECV pari all'1,5 U.A.. Questo studio conferma l'applicabilità della spettroscopia NIR per il monitoraggio on-line del processo di tostatura del caffè.
