    Politics of transition: Maguindanaon leaders and the Autonomous Region in Muslim Mindanao (ARMM

    This study will cover the political processes in the Province of Maguindanao on the Autonomous Region for Muslim Mindanao (ARMM), from the ratification of the 1987 Constitution on February 2, 1987 to its inauguration (ARMM) on November 6, 1990. This period will be referred to as the politics of transition. The study intends to analyze Maguindanaon leaders as major players in the politics of transition. The author will use documentary analysis. A historical overview of the problem will be given. The Records of the 1986 Constitutional Commission and the House of Representatives providing for its establishment will likewise be used in addition to newspaper accounts. The leaders involved will all be interviewed. This study as a whole will give insights on the Maguindanaon leadership and the role of three Maguindanaon leaders: Michael O. Mastura, Zacaria A. Candao, and Guimid P. Matalam, in the creation of the ARMM

    Establishment of axenic culture and induction of organogenesis in kapur bukit (dryobalanops beccarii dyer)

    Dryobalanops beccarii Dyer (Kapur Bukit) has been recommended by Sarawak Forest Department as one of the four timber species for forest plantation in Sarawak (Thai, 2002) and clonal materials are preferred for this purpose. However, the production of large quantity of planting stock through conventional propagation has not been very successful as Kapur Bukit posses recalcitrant seed and is irregular in flowering and thus fruiting. The in vitro technique therefore has been considered as an alternative for mass production of clonal material. Nevertheless, previous studies had shown that, it was difficult to establish aseptic and viable explants in culture because of browning and contamination. In 2009, Ang has found that phenolic released from petiolule explants were negligible. Roots also were successfully induced from friable callus that derived from this explants. To improve the percentage of axenic and viable explants in culture, further research was done by modifying the disinfection protocol that previously developed. Organogenesis of Kapur Bukit also was carried out by using various types and concentration of cytokinins, either alone or in combination with auxin to induce shoot formation. From this study, it was found that organogenesis of root was successfully induced at concentration 0.5, 1.0, 1.5, 2.0 mg/L of 2, 4-D. While green callus was observed in 4 mg/L BAP, 2 mg/L BAP+ 1 mg/L Kin and 3 BAP+ 0.5 mg/L Kin supplemented WPM medium. However, no sign of shoot formation was observed as phenolic that released caused green callus died after 3 months cultured. Seems browning problem is more critical than microbial contamination

    Heterologous expression of recombinant alcohol dehydrogenase gene from sago palm in bacterial and plant system

    Alcohol dehydrogenase (ADH) is an enzyme involved in pathways that respond to various environmental stresses such as osmotic, wound and anaerobic condition. In this study, the recombinant Metroxylon sagu Adh1 cDNA (r-msAdh1) was cloned into an expression vector; pET-41a(+) and expressed in Escherichia coli (E. coli) strain BL21 (DE3). SDS-PAGE analysis of the E. coli lysate revealed that large amount of expressed protein formed insoluble aggregation, non-active protein. However, soluble r-msAdh1 protein was successfully produced at low-growth temperature and showed catalytic activity when spectrophotometrically assayed. Second part of this study involves the in-planta transformation of r-msAdh1 cDNA into tomato seeds using Agrobacterium tumefaciens (A. tumefaciens) strain LBA4404. The result showed that, out of 15 putative transformed plants (T0) were analysed, five putative transgenic plant lines were determined to carry r-msAdh1 cDNA in their genome. Further analysis also showed that r-msAdh1 cDNA have passed into all T1 transgenic lines and expression analysis at transcript level confirmed the presence of r-msAdh1 in transgenic tomato genome

    CD4+ T cell subsets in adult allergic rhinitis patients attending Hospital Universiti Sains Malaysia

    Memory T cells exert effector function or generate effector cells in response to antigen. The proportions of CD4+ T cell subsets especially memory cells in allergic rhinitis (AR) patients sensitized to common allergens of house dust mites (HDMs) and shellfish have not been extensively studied. This study aimed to compare the mean percentages and absolute counts of CD4+ memory T cell subsets between: (i) non-allergic controls and AR patients; (ii) mild AR patients and moderate-severe AR patients. In addition, sensitization to common allergens, symptom severity scores, and mean absolute counts of leukocyte subsets were also determined. Fifty non-allergic controls and 100 AR patients diagnosed by physicians were recruited in this study. However, only 33 non-allergic controls were included in the analyses as others were excluded due to sensitization to the common allergens (Dermatophagoides farinae (Der f), crab and shrimp) as measured by plasma specific IgE tests. Stratified analyses were done based on two different definitions of AR patients , i.e. (i) non IgE-mediated AR patients; and (ii) IgE-mediated AR patients. Flow cytometry was used to determine the percentage of CD4+ naïve (TN; CD45RA+ CCR7+), central memory (TCM; CD45RA- CCR7+), effector memory (TEM; CD45RA- CCR7-) and terminally differentiated effector memory (TEMRA; CD45RA+ CCR7-) T cells from the peripheral blood. The absolute counts of CD4+ T cell subsets were obtained by dual platform methods from flow cytometer and hematology analyzer. It was observed that AR patients sensitized to common allergens (Dermatophagoides pteronyssinus, Der f, crab and shrimp) measured were predominantly sensitized to HDMs as compared to shellfish allergens. Moderatesevere AR patients had higher nasal and non-nasal symptom scores and reduced quality of life as compared to mild AR patients. Furthermore, the eosinophil count was significantly higher in IgE-mediated AR patients as compared to non-allergic controls. There were no significant differences in the mean percentages and absolute counts of CD4+ T cell subsets between non-allergic controls and IgE-mediated AR patients. However, significant reduction in the mean percentage (p = 0.0287) and absolute count (p = 0.0298) of CD4+ TEMRA cells were found in IgE-mediated moderate-severe AR patients as compared to IgE-mediated mild AR patients and 14/25 (56.0%) IgE-mediated moderate-severe AR patients had persistent symptoms. In conclusion, the mean percentage and absolute count of CD4+ CD45RA+ CCR7- TEMRA cells were siginificantly reduced in IgE-mediated moderate-severe AR patients as compared to IgE-mediated mild AR patients in our population of AR patients predominantly sensitized to HDMs

    Expression of Recombinant Alcohol Dehydrogenase in Escherichia coli Strain BL21 (DE3) and In Planta Agrobacterium Transformation of Tomato Seeds

    Alcohol dehydrogenase is an enzyme that is involved in various roles in plant such as in plant development, growth and plant responses to abiotic and biotic stresses. A recombinant alcohol dehydrogenase 1 (Adh1) cDNA (r-msAdh1) from Metroxylon sagu has been previously isolated, containing 20 nucleotides derived from Elaeis guineensis at the 5’-end, with a molecular weight of 1.14 kb. The objective of this study is to determine the function of r-msAdh1 via analyses in prokaryotic and eukaryotic hosts. For expression in prokaryotic system, pET-41a(+) with a 8x His tag at the C terminal was used for r-msAdh1 protein purification and expression was achieved using IPTG for four to six hours in Escherichia coli strain BL21 (DE3) incubated at low temperature. The induced BL21 strain produced a small amount of soluble r-msAdh1 protein while large amount was present as insoluble aggregates. Subsequently, the r-msAdh1 cDNA was transformed into tomato seeds (Solanum lycopersicum cv. MT1) via Agrobacterium-mediated in planta transformation. The integration of r-msAdh1 cDNA and the selectable marker were detected in transformed seedlings, T0, using polymerase chain reaction technique. The transformation efficiency was determined to be 33% for r-msAdh1 cDNA and 46% for the selectable marker. For stability analysis of the transgene, eleven T1 generation randomly selected from the transgenic T0 were analyzed for the presence of the cDNA, and all seedlings were found to contain the full length of r-msAdh1 cDNA. However, out of eleven T1 transgenic lines produced, only four seedlings were used for expression analysis using the reverse transcriptase PCR (RT-PCR). Two transgenic lines, T19 and T111, were determined to contain r-msAdh1 cDNA and verified by nucleotide sequencing. Although only a small number of T1 transgenic seedlings was obtained, this study shows that tomato seeds could be used as a target tissue for Agrobacterium-mediated in planta transformation primarily because the protocol is easy, rapid and cheaper compared to tissue culture-based method

The author considered three approaches to the analysis of the concept of love philosophical, linguo-cognitive, linguocultural approaches. The commonality of these three approaches is that the concept of love is viewed not only as a basic element of the personality language, but as an expression of the cultural characteristics of the people. It is concluded that the concept of love occupies a leading position among the concepts of happiness, hope, faith and goodness. Not only emotional, but also cognitive and volitional aspects are necessarily present in one love

In Ardalan and its capital, Sanandaj, as a center for the development of culture and education, Islamic and historical, in addition to a strong base, a large group of historians and intellectuals was found that significantly influenced Mahsharaf-Khanom's thought and consciousness. Mahashraf-Khanom, by recognition of many the researchers , was the only female historian in the Middle East in the 19th century. She was the only woman in the period who, along with the author of poems, practiced Islamic studies with great ability and had writings and compositions. In the beginning of the 19th century, the Iranian authorities, in particular, influenced the Ardalanian Kurds and changed their Sunni sect to Shia al-Jaafari. When they felt resistance, they imposed their authority and took control of the government directly in the emirate. They briefly removed it from existence. The Emirate of Ardalan, as part of the South - East Kurdistan, in addition to it was formed in areas, which later became under the control of the Emirate of Baban for more than two centuries, and became the source of disputes and conflicts between them and it came to the occupation of the land and cities, but despite the oppression of the Occupiers of Kurdistan, especially at the end of the 18-th century and the beginning of the 19-th century, there was a strong convergence and diverse integration among the population of the both Emirates and even the ruling class. Finally, when the dyanasty of Ardalan went to fall, Mahsharaf-Khanom and a few thousand Ardalanis moved to the capital of the Emirate of Baban, where they were received with great hospitality

    Determining Facebook as a Platform of Expressing Politics among University Students

    This study investigates the Facebook as a platform of expressing politics. The main objective of this study is to examine the students demographic; gender and academic performance of Facebook users regard to different variations of Facebook addiction, Facebook motives, and unwillingness to communicate face-to-face. Next objective is to define the level of unwillingness to communicate faceto-face, to determine and explain the relationship between Facebook motives and Facebook addiction, and to examine the relationship between Facebook addictions, Facebook motives, and Unwillingness to communicate face-to-face. Survey research design; questionnaire was applied. Data was analyzed by using SPSS 22 for both descriptive and inferential statistics that include with t-Test, Multiple regression analysis, Pearson correlation, and the pre-test of the study to fulfill the research objectives. The implication of this study is to give benefits to practical contribution and theoretical contribution that associated with this field. Future research needs to replicate this study in other situation and possibly in other areas for different view

    An exploratory study on the special education and early intervention programme for autistic children

    The objective of this paper is to explore the special education and the intervention programme for autistic children run by two Non-Governmental Organizations and identify how their programmes help autistic children improve learning disability. The methodology of this study is qualitative using a single case study. The research will be an exploratory study with a filed study on two different organizations which are currently active in helping children with learning disabilities. Semi-structured interviews and informal conversations are used to obtain the data. Findings showed that both centres used a naturalistic curriculum model where activities carried out are mostly dealing with real life activities, while topics and skills are focused on sensory skills and children are taught to use their senses and attention span. Teaching methods used are a mixture of several latest teaching techniques currently being applied worldwide for autistic children. In the early intervention programme, the teaching and learning techniques are based on the individual child’s ability. Both centres admitted that some parents play a supportive role while others play non-supportive roles. More knowledge of the learning process and learning development of autistic children as well as the teaching and early intervention strategies is needed to provide better understanding and guidance for teachers and parents or caregivers to deal with the needs of these children
