    Pseudogenes in metazoa: Origin and features

    The complete genome sequences with their annotations are a considerable resource in biology, particularly in understanding the global structure of the genetic material at the molecular level. The reason why some eukaryotic genomes contain large quantities of apparently unnecessary DNA, namely pseudogenes, while others seem to invest in more efficient thinning processes or are equipped with protection systems against parasitic elements still remains a mystery. Several genome-wide surveys have been undertaken to identify pseudogenes in the completely sequenced genome, bringing to light some differences both in their amount and distribution. Since pseudogenes are important resources in evolutionary and comparative genomics — as ‘molecular fossils’ — in this paper, a survey on the origins, features, abundance and localisation of the different pseudogenes is reported. As an example of genes producing processed pseudogenes, some experimental data obtained in the authors’ laboratories from the study of a nuclear gene coding for the mitochondrial transcription factor A (mtTFA), a key regulator of mitochondrial biogenesis, are also reported

    Variations on the Author

    “Variations on the Author” discusses two of Eduardo Coutinho’s recent films (Um Dia na Vida, from 2010, and Últimas Conversas, posthumously released in 2015) and their contribution to the general question of documentary authorship. The director’s filmography is characterized by a consistent yet self-effacing form of authorial self-inscription: Coutinho often features as an interviewer that rather than express opinions propels discourses; an interviewer that is good at listening. This mode of self-inscription characterizes him as an author who is not expressive but who is nonetheless markedly present on the screen. In Um Dia na Vida, however, Coutinho is completely absent form the image, while Últimas Conversas, on the contrary, includes a confessional prologue that moves the director from the margins to the center of his films. This article examines the ways in which these works stand out in the filmography of a director who offers new insights into the notion of cinematic authorship

    Quasi-dynamic modelling of shared mooring lines for floating wind turbines

    The idea of shared mooring systems has been introduced in the offshore wind farm industry as an effort to lower the overall installation costs for floating offshore wind farms. The reason for building such plants is that using floating turbines allows them to move to deeper waters, reducing visual and environmental impact and allowing footprint reduction, with the downside being an increase in the total installation costs.Because shared mooring lines create couplings between neighbouring turbines, the use of shared mooring lines adds complexity to the design and modelling of offshore floating wind farms. So far, these systems have been investigated and analysed using either quasi-static or fully dynamic models. With the goal of using a method that can be more accurate in terms of dynamic behaviour of the mooring lines than the quasi-static one and less time-consuming than the fully dynamic one by using a system with less degrees of freedom, this project seeks to further explore the idea of shared mooring lines within a quasi-dynamic model. A quasi dynamic system is not as complex as the dynamic model but more rigorous than the quasi-static one because it includes terms related to the drag and inertia of the mooring lines. Inertial loads on mooring lines can alter the tension of the mooring line at the fairlead, while drag forces on mooring lines will dampen platform motions, especially for slowly varying motions. These are the two reasons why it is critical to include these effects in the model. In this case inertial terms have not been considered, focusing mainly on including drag terms for both the anchor and shared lines together with the geometric and elastic stiffnesses. To begin with, the quasi-dynamic model has been applied to a two-turbine system, constrained to move only horizontally in surge as a model assumption. The model has been implemented in Matlab as a two-degree-of-freedom system. The ultimate goal of this research is to see if this approach can be used instead of conducting a thorough dynamic analysis. As a result, in addition to the Matlab model, an equivalent system in SIMA has been built to perform a full dynamic analysis and allow comparison of the two models' findings.Based on the model results, it appears possible to obtain good accuracy for both motion amplitudes and tension values in a shorter amount of time than a full-dynamic analysis.     Metazoan OXPHOS gene families: Evolutionary forces at the level of mitochondrial and nuclear genomes

    AbstractMitochondrial and nuclear DNAs contribute to encode the whole mitochondrial protein complement. The two genomes possess highly divergent features and properties, but the forces influencing their evolution, even if different, require strong coordination. The gene content of mitochondrial genome in all Metazoa is in a frozen state with only few exceptions and thus mitochondrial genome plasticity especially concerns some molecular features, i.e. base composition, codon usage, evolutionary rates. In contrast the high plasticity of nuclear genomes is particularly evident at the macroscopic level, since its redundancy represents the main feature able to introduce genetic material for evolutionary innovations. In this context, genes involved in oxidative phosphorylation (OXPHOS) represent a classical example of the different evolutionary behaviour of mitochondrial and nuclear genomes. The simple DNA sequence of Cytochrome c oxidase I (encoded by the mitochondrial genome) seems to be able to distinguish intra- and inter-species relations between organisms (DNA Barcode). Some OXPHOS subunits (cytochrome c, subunit c of ATP synthase and MLRQ) are encoded by several nuclear duplicated genes which still represent the trace of an ancient segmental/genome duplication event at the origin of vertebrates

    La información de esta miniguía se basa en los trabajos de Felipe Solís.Durante el periodo Postclásico Tardío (12OO a 1521 d.C.) se llevaron a cabo en la Cuenca de México grandes movimientos culturales y migratorios que originaron uno de los más importantes desarrollos del México antiguo: el de los mexicas. El estudio de los materiales arqueológicos recuperados en las primeras exploraciones -escultura y cerámica- , así como de las características arquitectónicas del edificio que Brasero ceremonial, parte superior del basamento se conserva, no han permitido conocer la extensión del sitio, el número de edificios que conformaban el centro, ni cuáles eran las zonas de habitación. Sin embargo, se sabe que su economía estuvo ligada a los depósitos lacustres de agua dulce o salada y a la agricultura. Se explotaron la fauna, la flora, la sal y el tequesquite, este último se utilizaba en el proceso de cocción del maíz. Se ha descubierto que Acatitlan está cultural y políticamente ligado a los mexicas, aunque no se le menciona en los textos indígenas de la época ni en las fuentes históricas de la conquista o de la Colonia, por lo cual se supone que fue abandonado antes del arribo de los españoles.</p

She performs and exhibits her work widely at national and international venues. She has an MFA from the University of Chile in Santiago, and she did her postgraduate work at the Slade School of Fine Arts, University College, London. The recipient of many honors, she received The Pennies from Heaven Award, 2002, The Anonymous Was a Woman Award, l999, The Lila Wallace-Reader’s Digest Arts International Award in l992, The Fund for Poetry Award in l995-96 and The Human Rights Award from the Fund for Free Expression in New York in l985.     The RHNumtS compilation: Features and bioinformatics approaches to locate and quantify Human NumtS

    Abstract Background To a greater or lesser extent, eukaryotic nuclear genomes contain fragments of their mitochondrial genome counterpart, deriving from the random insertion of damaged mtDNA fragments. NumtS (Nuclear mt Sequences) are not equally abundant in all species, and are redundant and polymorphic in terms of copy number. In population and clinical genetics, it is important to have a complete overview of NumtS quantity and location. Searching PubMed for NumtS or Mitochondrial pseudo-genes yields hundreds of papers reporting Human NumtS compilations produced by in silico or wet-lab approaches. A comparison of published compilations clearly shows significant discrepancies among data, due both to unwise application of Bioinformatics methods and to a not yet correctly assembled nuclear genome. To optimize quantification and location of NumtS, we produced a consensus compilation of Human NumtS by applying various bioinformatics approaches. Results Location and quantification of NumtS may be achieved by applying database similarity searching methods: we have applied various methods such as Blastn, MegaBlast and BLAT, changing both parameters and database; the results were compared, further analysed and checked against the already published compilations, thus producing the Reference Human Numt Sequences (RHNumtS) compilation. The resulting NumtS total 190. Conclusion The RHNumtS compilation represents a highly reliable reference basis, which may allow designing a lab protocol to test the actual existence of each NumtS. Here we report preliminary results based on PCR amplification and sequencing on 41 NumtS selected from RHNumtS among those with lower score. In parallel, we are currently designing the RHNumtS database structure for implementation in the HmtDB resource. In the future, the same database will host NumtS compilations from other organisms, but these will be generated only when the nuclear genome of a specific organism has reached a high-quality level of assembly.</p
