Blaufuss, F. Halzen, C. Kopper (Changed to add one missing author, no other changes from initial version.)The recent observation by the IceCube neutrino observatory of an astrophysical flux of neutrinos represents the "first light" in the nascent field of neutrino astronomy. The observed diffuse neutrino flux seems to suggest a much larger level of hadronic activity in the non-thermal universe than previously thought and suggests a rich discovery potential for a larger neutrino observatory. This document presents a vision for an substantial expansion of the current IceCube detector, IceCube-Gen2, including the aim of instrumenting a volume of clear glacial ice at the South Pole to deliver substantial increases in the astrophysical neutrino sample for all flavors. A detector of this size would have a rich physics program with the goal to resolve the sources of these astrophysical neutrinos, discover GZK neutrinos, and be a leading observatory in future multi-messenger astronomy programs

Examination of parasites as a treatment for inflammatory bowel disease using murine models
ABSTRACTEXAMINATION OF PARASITES AS A TREATMENT FOR INFLAMMATORY BOWEL DISEASE USING MURINE MODELSByJamie Jennifer Kopper Inflammatory Bowel Disease (IBD) is a serious, life-threatening chronic inflammatory disease of the gastro-intestinal tract that has been reported to affect 1.1 million adults in the US. Patients with IBD often become refractory to traditional treatments which include antibiotics, immune modulators and corticosteroids. IBD can result in a life-threatening condition and lead to the necessitation of surgical interventions such as for bowel resections. In 2003 Dr. Joel Weinstock proposed using Trichuris suis as a treatment for inflammatory bowel disease in patients who were otherwise refractory to standard treatments and subsequently published the results of two small-scale clinical trials. Here, he reported that T. suis resulted in amelioration of clinical signs of disease in many patients and no adverse side effects. Despite these positive results, the mechanism(s) by which T. suis improved patient outcomes remains unknown. In this work we set forth to address possible mechanisms, and to investigate the possibility of adverse side effects due to T. muris infections in a murine model. We addressed the hypothesis that infections with T. muris alters the colonic microbiota in mice, which may serve as one mechanism to explain the improvement observed in the human patients treated with this worm. We found that C57 BL/6 IL-10-/- mice infected with T. muris had sex and dose dependent morbidity and mortality. Additionally, these infections resulted in severe peritonitis with or without gastrointestinal perforations in some mice. In a subsequent experiment C57 BL/6 IL-10-/- mice infected with T. muris were concurrently treated with prednisolone or metronidazole, neither of which resulted in a significant decrease in mortality when compared to the untreated infected mice. Finally we infected C3H/HeJ and C3Bir IL-10-/- mice, which are predisposed to developing a spontaneous colitis, with T. muris to test our hypothesis that infections with T. muris would change the colonic microbiota. Using Terminal Restriction Fragment Length Polymorphisms (TRFLPs) we determined that mice infected with T. muris had significantly different proximal colon microbial microbiota when compared to uninfected mice. To further assess the effects of colitis without T. muris infections on proximal colon microbiota we obtained samples from C3Bir IL-10-/- mice within our breeding colony who developed spontaneous colitis and compared them to C3Bir IL-10-/- mice without colitis. From this study we found that overall there were no differences, as assessed using TRFLP, between the two groups. From the entirety of this work we conclude that T. muris has the possibility of causing adverse side effects in some individuals, which could include gastrointestinal perforations and peritonitis, particularly those who are deficient in IL-10. Additionally, we concluded that T. muris does result in changes in the structure of the proximal colon microbiota, which may be one mechanism through which it worked as a treatment for IBD in human patients.Thesis (Ph. Accuracy of ultrasound-based (BAT) prostate-repositioning: A three-dimensional on-line fiducial-based assessment with cone-beam computed tomography
Purpose: To assess the accuracy of ultrasound-based repositioning (BAT) before prostate radiation with fiducial-based three-dimensional matching with cone-beam computed tomography (CBCT). Patients and Methods: Fifty-four positionings in 8 patients with I-125 seeds/intraprostatic calcifications as fiducials were evaluated. Patients were initially positioned according to skin marks and after this according to bony structures based on CBCT. Prostate position correction was then performed with BAT. Residual error after repositioning based on skin marks, bony anatomy, and BAT was estimated by a second CBCT based on user-independent automatic fiducial registration. Results: Overall mean value (MV +/- SD) residual error after BAT based on fiducial registration by CBCT was 0.7 +/- 1.7 mm in x (group systematic error [M] = 0.5 mm; SD of systematic error [Sigma] = 0.8 mm; SD of random error [sigma] 1.4 mm), 0.9 +/- 3.3 mm in y (M = 0.5 mm, Sigma = 2.2 mm, sigma = 2.8 mm), and - 1.7 +/- 3.4 turn in z (M = - 1.7 mm, Sigma = 2.3 mm, sigma = 3.0 mm) directions, whereas residual error relative to positioning based on skin marks was 2.1 +/- 4.6 mm in x (M = 2.6 mm, Sigma = 3.3 mm, sigma = 3.9 mm), -4.8 +/- 8.5 mm in y (M = -4.4 mm, Sigma = 3.7 mm, sigma = 6.7 mm), and -5.2 +/- 3.6 mm in z (M = -4.8 mm, Sigma = 1.7 mm, sigma = 3.5mm) directions and relative to positioning based on bony anatomy was 0 +/- 1.8 mm in x (M = 0.2 mm, Sigma = 0.9 mm, sigma = 1.1 mm), -3.5 +/- 6.8 mm in y (M = -3.0 mm, Sigma = 1.8 mm, sigma = 3.7 mm), and -1.9 +/- 5.2 mm in z (M = -2.0 mm, Sigma = 1.3 mm, sigma = 4.0 mm) directions. Conclusions: BAT improved the daily repositioning accuracy over skin marks or even bony anatomy. The results obtained with BAT are within the precision of extracranial stereotactic procedures and represent values that can be achieved with several users with different education levels. If sonographic visibility is insufficient, CBCT or kV/MV portal imaging with implanted fiducials are recommended. (C) 2008 Elsevier Inc
The missing link: an examination of skin clothing production of north central plains Native Americans
Deer hides are one of the most commonly used materials for clothing production among the prehistoric Northern Great Plains peoples. Richard Michael Gramly?s 1977 article, Deerskins and Hunting Territories: Competition for a Scarce Resource of the Northeastern Woodlands, assumed an estimate of 3.5 hides per person per year. A figure widely cited in anthropological literature, yet provides no supporting data. This investigation seeks to find that data as applied to the Plains Indians. Measurements were taken of known clothing articles to determine surface area, and these were compared with the surface areas of average sizes of small, medium, and large hides. Comparing the surface areas allowed for an accurate estimation of the number of hides needed by both a man and woman for an appropriate climate wardrobe. Such information can be vital in interpreting prehistoric and historic hunting patterns, with significant social and ecological implications
