This article is distributed under the terms of the Creative Commons Attribution License, which permits unrestricted use and redistribution provided that the original author and source are credited.When the nucleolus disassembles during open mitosis, many nucleolar proteins and RNAs associate with chromosomes, establishing a perichromosomal compartment coating the chromosome periphery. At present nothing is known about the function of this poorly characterised compartment. In this study, we report that the nucleolar protein Ki-67 is required for the assembly of the perichromosomal compartment in human cells. Ki-67 is a cell-cycle regulated protein phosphatase 1-binding protein that is involved in phospho-regulation of the nucleolar protein B23/nucleophosmin. Following siRNA depletion of Ki-67, NIFK, B23, nucleolin, and four novel chromosome periphery proteins all fail to associate with the periphery of human chromosomes. Correlative light and electron microscopy (CLEM) images suggest a near-complete loss of the entire perichromosomal compartment. Mitotic chromosome condensation and intrinsic structure appear normal in the absence of the perichromosomal compartment but significant differences in nucleolar reassembly and nuclear organisation are observed in post-mitotic cells

    Phenomenographic perspectives on the learning experience and process in higher education physics

    Presented at the Higher Education Close-Up conference (HECU4), Cape Town, June 2008The focus of phenomenographic research has been the experience of learning (Marton & Säljö, 1976a; Marton & Säljö, 1976b; Säljö, 1979; Marton et al., 1984/1997; Booth, 1997; Marton & Booth, 1997; Pang, 2003). Drawing on our recent research into the process of learning in higher education physics contexts, we present a discussion of the experience and process of learning, and perspectives from which it can be analysed and understood that emerge from the phenomenographic tradition. First we will relate our empirical work, then elaborate on it as an example of a study of learning. Then we will enter into a reflection on phenomenography as a research approach for the 21st century, in the delimited field of researching learning and teaching practices in higher education, and make an argument that phenomenography has a role to play in the transformative processes demanded by a changing society

    The research presented in this thesis rests on the premise that the administrative and legal systems of France have a critical bearing on the way that decisions on applications for permissions to build are taken, and the nature of the decisions themselves. In the knowledge that the French system of law offered a legalistic, regulatory franiework for planning policy and policy implementation, four specific questions are posed: firstly about the relationship of plans to development control decisions; secondly about the effects of the system on applicants; thirdly about the possibilities for third parties to be involved in, and seek redress from, development control decisions and fourthly about the effects of the decentralisation of development control powers that has taken place since 1983. These questions are then located within a broader discussion of discretion, accountability and the management of uncertainty. The theoretical discussion of the first chapter paves the way for a more detailed presentation of the nature and origins of French local administration and French planning law and procedure which in turn lead to a case study of the 55 communes of the Urban Community of Lyon and eight studies of development control applications which are explored through an examination of the case file documents and interviews with participants. Two sets of conclusions are drawn from the study. The first set concerns the effects of a legalised system on the making and implementation of planning policy. The first conclusion is that the legalistic approach of the French planning system appears to create serious difficulties for finding an appropriate expression for policy. In part the problem is shown to be as much a question of ethos as of what is really possible under the law, amid some examples of practice in Lyon show how flexibility is still possible even within a legalised system. The second conclusion is that once the rules are departed from, the system offers no alternative means of testing policy in its specific application, although the use of non-statutory consultation meetings in Lyon has gone some way to meeting the problem. The third is that the pattern of zoning and regulations does not appear to help the maintenance of a planning strategy. The fourth is that a legalised system does not promote certainty for either administrators or applicants. The fifth is that a legalised system does not permit third parties to participate in the decision-making and ensures that objections are seen mainly as being about property values. The second set of conclusions has to do with the question of the power to decide and the accountability of decision-makers. The first is that the legalised system, while offering potential for agency discretion, nevertheless appears to favour officer discretion which on the evidence of the case studies is rife. While offering mayors the possibility of tactical power, it appears to reduce the accountability for decisions taken. Moreover, the control of the legality of decisions is dependent equally upon the discretion of the prefect. The second is that the pattern of crossregulation within the French system of local government has ensured the continuity of dependencies between the principal actors in the planning system. The final conclusion is that decentralisation has had relatively little effect on the balance of power. In the Lyon conurbation, COURLY would appear to be the principal beneficiary of the new powers, which would suggest that more power will be concentrated in future at the local level, but that the power will not be any more susceptible to control by the electorate

    Oggi è evidente la complessità del ruolo dell’insegnante, chiamato a governare complessi sistemi formativi orientati all’inclusione. Il percorso sviluppato entro il Corso di Pedagogia e Didattica per l’Inclusione, Corso di Studi in Scienze della Formazione Primaria, Università di Padova, potrebbe essere considerato un buon processo (Trinchero, 2017) che si focalizza sulle qualità rilevanti che un professionista dell’inclusione dovrebbe possedere (obiettivo perseguito attraverso la didattica frontale) e su come queste necessitino di essere messe insieme nella pratica quotidiana dei docenti (didattica laboratoriale). Lo sfondo teorico, costituito dall’Index for inclusion (Booth e Ainscow, 2014) e dal Sistema di Attività (Engestrom, 1987), ha guidato la costruzione dell’itinerario didattico (co-teaching practice), le attività svolte (Inclusario, Manifesto per l’inclusione, Didattiche inclusive, giornata seminariale), e le narrazioni promosse (grande domanda e portfolio dello studente)

    The construction of Karen Karnak: The multi-author-function

    This thesis is situated within the comparatively recent developments of Web 2.0 and the emergence of interactive WikiMedia, and explores the mode of authorship within a Read/Write culture compared to that of a Read/Only tradition. The hypothesis of this study is that the role of the audience has become merged with the author, and as such, represents new functions and attributes, distinct from a more conventional concept of authorship, in which the roles of audience and author are more separate. Read/Write and participatory culture, as defined by this study, is focused on collaboration, and includes the influences of D.I.Y. culture, Open-Source practices and the production of text by multiple authors. Multi-authorship presents a re-thinking of several concepts which support the notion of the individual author, since the focus of multi-authorship is not on attribution and ownership of a finished text, but on the continued malleability of a text. Modes of multi-authorship, demonstrated in the use of the pseudonyms Alan Smithee and Karen Eliot, represent declarative authors whose names signify multiple origins, whilst concurrently indicating a distinct body of work. The function of these names form an important context to this study, since primary research involves the construction of an experimental mode of multi-authorship utilising WikiMedia technology and the interaction of thirty nine participants, who are invited to create a body of work under the collective pseudonym Karen Karnak. The data generated by this experiment is analysed using aspects of Michel Foucault's author-function to identify and determine power structures inherent in the WikiMedia context. The interplay of power structures, including concepts such as identity, ownership and the body of work, affect the resulting mode of authorship and contribute to the construction of Karen Karnak, suggesting further areas of research into the emerging multi-author

    Fluctuation, invisibility, fatigue – the barriers to maintaining employment with systemic lupus erythematosus: results of an online survey

    © The Author(s) 2018. Objectives: Systemic lupus erythematosus (SLE) is associated with high levels of workplace disability and unemployment. The objective of this study was to understand the reasons for this and to describe the barriers and facilitators of employment identified by people with SLE to develop appropriate solutions. Unemployment, as well as unsuitable work, has adverse health outcomes. Methods: Adults with SLE completed a UK-specific online survey, through the LUPUS UK website, designed to find out more about the difficulties and successes that people with SLE have in maintaining employment. The survey was predominantly qualitative, to understand participants’ employment experiences to generate possible solutions. Results: Three hundred and ninety-three people gave detailed responses to the survey within eight weeks. Every respondent reported a detrimental effect of SLE on their ability to work: 40.45% had left employment because of it. The themes of concern to respondents were unambiguous: (i) the difficulties of working (and career damage) with SLE, (ii) fear and anxiety overshadowing work/family life, (iii) the greater potential to remain in some employment or stay in full employment when modifications of work pattern and support from management and colleagues were available. SLE-related fatigue, its invisibility and fluctuating nature were felt to be the main barriers to maintaining employment. Numerous respondents could work only part-time and anxiety was high regarding their future ability to continue working. Many had taken substantial pay reductions and refused offered promotions to preserve their health. Distress due to loss of work and the benefits it brings were reported by every respondent who had left work. Conclusion: SLE presents specific difficulties for maintaining employment – fatigue, fluctuation and invisibility – not addressed by current anti-discrimination legislation or currently available ‘reasonable adjustments’. This study demonstrates that (i) employment is an important area of concern for people with SLE, (ii) SLE has significant detrimental effects on individuals’ ability to participate and progress in employment, (iii) legislators and employers need information about SLE as invisibility and fluctuation cause hidden problems, and (iv) more data is needed to inform workplace adjustments if individual distress and societal loss of skills are to be addressed
