Xylosandrus crassiusculus (Motschulsky) (Coleoptera: Curculionidae) and Its Fungal Symbiont Ambrosiella roeperi Associated with Arecanut Kernel Decay in Karnataka, India
Xylosandrus crassiusculus (Coleoptera: Curculionidae: Scolytinae) is reported causing damage to areca palm plantations (Areca catechu L.—Arecaceae) in Karnataka (India). In particular, X. crassiusculus has been observed attacking and successfully reproducing on areca nuts; besides the new host plant record, the data provided here represent the first documented case of spermatophagy for this xyleborine beetle. All infestation symptoms of this polyphagous pest were documented and illustrated. The identity of the scolytid, besides morphologically, was confirmed by its DNA barcoding. Eggs, larvae and pupae were found within the galleries of infested kernels. All galleries of the infested kernels were characterized by the presence of whitish to greyish fungal growth. The fungus was identified as Ambrosiella roeperi, a known symbiont of Xylosandrus crassiusculus. Incidence of this symbiotic insect-fungus complex in the economic part of arecanut, i.e., the kernel, is of serious concern. In a climate change scenario, this beetle with fungal symbionts may pose a serious threat to arecanut production in India and elsewhere
An overview of genetic and molecular factors responsible for recurrent pregnancy loss
Recurrent pregnancy loss usually results from disorders that cause intrauterine fetal damage, such as maternal or paternal chromosomal abnormalities. About 15 to 20 percent of all recognized pregnancies end in a first-trimester spontaneous abortion. Parents who are carriers of structural abnormalities have a higher risk of miscarriage because of the aberrations in genetic information may not segregate properly into the reproductive cells. This may be due to translocations, inversions, deletions, and duplications causing pregnancy loss. It is believed that between 3 and 5 percent of recurrent miscarriages are due to genetic factors, about 7 percent are caused by chromosome defects, 15 percent to hormonal defects, and 10 to 15 percent to anatomical defects. The most common cause of early pregnancy losses are chromosomal abnormalities that occur by chance, except in the case of parental chromosomal rearrangements and are not under any controllable influences. This review focuses on the genetic and molecular abnormalities that may contribute to this clinical problem and delineates strategies for genetic evaluation and clinical management in subsequent pregnancie
Impact of spermiogram and sperm function test in idiopathic recurrent pregnancy loss
Aims: To evaluate the role of sperm factors in idiopathic recurrent pregnancy loss cases. Methods: We recruited 100 male subjects where their female partners experienced 2 or more idiopathic pregnancy losses. Recurrent pregnancy loss (RPL) males were grouped into two groups RPL1 (individuals having 2 abortion) and RPL2 (individuals having more than 2 abortions). Fifty volunteers who had fathered child/children prior to the study without the history of recurrent pregnancy loss and unassisted pregnancies were considered as control group. We grouped RPL males into two categories age below 35 years and above 35 years to test whether age plays any role on RPL. Routine semen parameters and sperm function test were performed for all the subjects. Statistical analysis was performed using Independent- Samples T test. Results: Insignificant differences were observed in seminal volume, pH, motility and sperm count but vitality test scores were significantly lesser in both RPL groups when compared to control group. For all three sperm function test both the RPL group showed lesser scores when compare to control. RPL2 group showed more abnormalities when compared to RPL1 group. We did not identify any significant difference for any of the parameters between two age group of RPL males. Conclusion: Our results recommend the screening of both partners simultaneous in RPL cases for the better diagnosis and treatment

The traditional Ayurvedic literature includes a thorough grasp of effective treatment to a majority of Modern-day health issues. Idiopathic Thrombocytopenic purpura (ITP) is one of these issues that is strengthening its roots in society. ITP is an Autoimmune disorder characterized by a low platelet count and mucocutaneous bleeding. ITP is defined as a Hematologic disorder, characterized by isolated thrombocytopenia without a clinically apparent cause. The major causes of accelerated platelet destruction include Immune thrombocytopenia, Decreased bone marrow production and increased splenic sequestration. Bleeding disorders are explained in classics under the heading of Raktapitta. ITP can be correlated with Tiryaka Raktapitta. It is Pitta pradhana tridoshaja vyadhi and it manifests as subcutaneous bleeding. In this study, a 21 year old female patient had complaints of Epistaxis, purpura, petechia, bruises since 6 months and she was treated with ayurvedic medicines. After treatment significant improvement was observed in patient’s condition, Haematological parameteres i.e, Platelet count was stable

Role of phenylthiocarbamide as a genetic marker in predicting the predisposition of disease traits in humans
The main objective of this study is to find out the genetic variation and predisposition of overweight/obese, smoking/alcoholism and thyroid disease traits among tasters and non-tasters in Mysore population, South India. Bitter-taste perception for phenylthiocarbamide (PTC) is a classically variable trait both within and between human populations. Many studies have reported that in world population, approximately 30% of them are PTC non-tasters and 70% are tasters. This investigation was conducted during the year 2009-2010 involving a total 1352 study subjects and divided into three different groups, considering the age ranging from 13 to 50 years. Phenylthiocarbamide taste sensitivity was measured by administering a freshly prepared 0.025% of phenylthiocarbamide solution using the Harris and Kalmus method with a slight modification and the results were recorded. In the first group of 100 obese/overweight children, 28% are taster and 72% are non-taster and among 100 control group 67% are tasters and 43% are non-tasters. In second group, out of 1152 individuals 710 (61.63%) are tasters and 442 (38.37%) are non-tasters including both males and females. In the third group, out of each 100 thyroid patients and the control group, tasters are significantly more frequent (61.41%) than the non-tasters (38.58%) in the control group, but a higher proportion of non-tasters are recorded among individuals with thyroid problems (68%) compared to tasters (32%). There is a significant higher incidence of PTC tasters than non-tasters among general population in this study. As phenotypic variation in PTC sensitivity is genetic in origin, this may represent a surrogate risk factor for the development of multifactorial disease and disorders

Beauveria bassiana is an entomopathogenic fungus that causes the white muscadine disease in insects. The majority of entomopathogenic fungi are soil and insect borne, 15 soil samples were collected from seven different locations during 2021, from January to December. Similarly, during 2022, March to December, 15 fungus-infected insect specimens were collected from five different locations hence soil and insect samples from various ecosystems were collected. As a result, 30 B. bassiana isolates from 11 different geographical areas were identified using morphological characteristics and multilocus sequence data in this investigation. The taxonomical positions of the isolates were determined using morphological characteristics and phylogenetic inferences based on three loci (Internal Transcribed Sequence, Elongation Factor-1α, and B. bassiana chitinase 1). In phylogenetic analysis of B. bassiana, the Maximum Likelihood analytical method produced distinct tree topology when compared to Neighbor-joining and minimum evolution. Three isolates viz., Bb3, Bb7 and Bb20 were found closely linked with reference isolate (KTU-24) and other showed the higher population diversity among them. The genetic distances of 30 B. bassiana isolates revealed that 15 were not closely related (D varied from 0.003 to 0.036). The pathogenicity of B. bassiana isolates from various hosts along with one commercial formulation (Beveroz) was assessed against Tetranychus truncatus under in vitro conditions by a completely randomized design (CRD) experiment. The same experiment was repeated thrice to confirm the pathogenicity of B. bassiana against T. truncatus. Later, the collected T. truncatus mortality data was converted into corrected mortality by using the Abbott formula and the values were examined using analysis of variance (ANOVA) in SPSS 23.0 software. Duncan’s Multiple Comparison Test was also done to compare the percentage mortality rates among the 30 B. bassiana isolates. The recorded results showed that the Bb6, Bb15 and Bb12 isolates caused significantly higher mortality of T. truncatus, i.e., 97.73, 96.73 and 94.50% respectively, than the other isolates. This study showed the relativeness among the B. bassiana isolates and establishes their bio-efficacy against T. truncatus, which further can be used for commercialization as bio-pesticide.</p
