Insight on trans-plasma membrane behavior of virus-infected plant cells
Little is known about the ion fluxes generated during plant-virus interactions, despite significant losses caused by viruses to agricultural crops. Changes in average ion currents were identifying an early event in the signal transduction pathway related to virus/host interaction. While significant decrease in the average inward currents, mainly due to Ca2+ moving into the cell was observed, the role of potassium may be significant. Host specific K+ efflux with a concomitant decrease in the intracellular K+ was observed in tobacco plants during the early minutes after infection, suggesting many hypothesis about the role of potassium in host-virus interaction. In the last years, trans-plasma membrane potential was evaluated for some viruses, observing as effect on membrane was different in relation to virus infection and host. Procedimento per l'inserimento di microchip a radiofrequenze (RFID) in innesti - Talea di vite
Sono state individuate due specifiche metodologie di inserimento dei microchip identificativi (TAG) a radiofrequenze RFID (Radio Frequency IDentification) in vite. Le procedure adottate sono state studiate per annullare o ridurre al minimo i potenziali effetti traumatici imputabili all’introduzione permanente di una componente elettronica in piante di vite. Questa azione è essenziale per la realizzazione di un sistema integrato di tracciabilità nella filiera vivaistico viticola. Dalla lettura, infatti, del TAG effettuata successivamente al suo inserimento in pianta è possibile rilevare un numero abbinato ad una scheda consultabile direttamente su PC palmare (PDA), contenente alcune informazioni chiave per una prima identificazione della pianta. Successivamente, per consultare una scheda tecnica dettagliata relativa alla medesima pianta, è sufficiente collegarsi presso uno specifico sito web, ed accedere ad una banca dati con il semplice inserimento del numero letto con il palmare
Radio frequency identification technology for improving traceability in the grapevine nursery sector
Radio frequency identification (RFID) systems seems to be the new challenge for developing traceability programs in agriculture, as widely illustrate in other fields of applications, i.e. logistic, animal identification or environmental monitoring. In grapevine nursery sector, this technology could be useful for increasing consumer fidelity, i.e. thanks to the possibility to check the “history” of grapevines used for producing a wine, besides assist the logistic e production phase of farming process. In our trial we are evaluating the effects on growth due to the implementation of microchips into grapevine, and developing an effective procedure for online managing of information associated to chips.
The trial was carried out from 2007 in Vivai New Plants, Cenaia (Pisa, Italy) considering five different clones supplied by Associazione Toscana Costitutori Viticoli (TOS.CO.VIT., San Piero a Grado, Pisa, Italy; www.toscovit.it). Clones (Sangiovese I-SS-F9-A5-48, Prugnolo gentile I-Bruscello, Colorino I-US-FI-PI-10, Trebbiano toscano I-S. Lucia 12 and Vernaccia di S. Gimignano I-VP6) were grafted on rootstock 1103P, inserting the microchips (Transponder Glass TAG) by two different procedures actually under monitoring. The first one concern a TAG inserting into plantlet medulla, while the second one involved a specific machine for providing the insertion under bark.
Each TAG inserted into plantlet is identified by a number readable by a Palm-PC, linked to a simplified identification file. To consult a complete grapevine identification file, attached by technical information, the users (farmers, consumers) can access to a website, actually under construction, and enter into an online database, using the identification number supplied by Palm-PC
In vivo inhibition of trans-plasma membrane electron transport by antiviral drugs in grapevine
Electrophysiological techniques were applied to investigate the action of antiviral drugs during trans-plasma events in in vivo grapevine cells infected by GLRaV-1 and GLRaV-3. Carbon fiber microelectrodes and redox-sensitive dyes were used to measure trans-plasma membrane electron transport (t-PMET) activity in healthy and infected samples treated with ribavirin, tiazofurin and oseltamivir. Each drug caused a reduction in oxidation current (expressed as Δ[Fe(2+)]) in healthy samples, indicating t-PMET inhibition. In almost all infected samples, the effect of drugs on t-PMET activity was significantly lower, suggesting that higher content of NADH in infected plants can interfere with t-PMET inhibition caused by drugs. Moreover, virus-infected samples exhibited elevated t-PMET activity compared to healthy samples. Analogous effects were observed by dye tests. Considering the effects of drugs on trans-plasma membrane potential, tests showed the activity of a proton pump during drug treatments with no significant difference with regard to health status
