Fungal lipids.Biosynthesis and signalling during plant-pathogen interaction
Lipids occur in fungi as major constituents of the membrane systems and minor component in the cell wall; they can store energy in the lipid bodies and, in some cases, they can act as intra-extracellular signals. Fungi contain a various set of lipids, including fatty acids, oxylipins, sphingolipids, phospholipids, glycolipids, and sterols. Current studies in lipids suggest their additional role in cell signalling; for instance, host-pathogen exchange lipid signals at the interface during their interaction. This review aims examining those fungal lipid classes involved in the pathogenic interaction with the host plants. The lipid signals may trigger host immune response as well as functioning as virulence factors altering the lipid homeostasis of the host cells
β-Glucan synthase induction in mushrooms grown on olive mill wastewaters
Beta-1-3-Glucan synthase activity and its induction by olive mill wastewaters (OMW) was studied in ten fungal strains (Auricularia auricula-judae, Lentinula edodes, Pleurotus eryngii, Stropharia aeruginosa, Agrocybe aegerita, P. pulmonarius, Armillaria mellea, P. ferulae, P. ostreatus, P. nebrodensis). A microtiter-based enzymatic assay on -1-3-glucan synthase activity was carried out on all mycelia growth both on the control medium and on OMW. Among the fungi assayed, L. edodes -1-3-glucan synthase was highly enhanced in OMW. The main components of OMW, i.e. phenols and lipids, were added separately to the control medium, to highlight the mechanism of L. edodes -1-3-glucan synthase induction. A Southern blot analysis and PCR with degenerated primers were carried out to detect the presence of fks1-like genes in these Basidiomycetes. The sequences obtained from the ten Basidiomycota were remarkably similar to fks1 from Filobasidiella neoformans. Spectrofluorimetric and RT-PCR analyses of -1-3-glucan synthase were performed on the mycelia of L. edodes. How environmental stresses affect aspergillus flavus metabolism.
Aspergillus flavus is a saprophytic fungus responsible for world-wide spread harvest and post-harvest infections on cultivations, mainly on cereal grains and legumes. A. flavus is one of the main producers of aflatoxins, the most dangerous carcinogenic metabolites in nature. In order to effectively address the economic and sanitary consequences of A.flavus contamination, a detailed and extensive knowledge of the pathogen metabolism and of the environmental conditions, that trigger the different biological processes, is of paramount importance. We investigated how some biological processes (Starvation, Oxidant and Hypoxia conditions) affected A. flavus mycelial growth, conidiogenesis and aflatoxin synthesis

Il testo si presenta diviso in 6 sezioni, in cui le prime 3 approfondiscono
rispettivamente aspetti molecolari e cellulari di virus e viroidi (sezione 1), fitoplasmi
e batteri (sezione 2), oomiceti e funghi (sezione 3), mentre la quarta
sezione è interamente dedicata alle interazioni molecolari pianta-patogeno
e ambiente, con un focus particolare sulle principali reazioni difensive della
pianta, locali e sistemiche, sia ai patogeni che ai principali stress abiotici.
Nella quinta sezione si affrontano i temi più legati alla difesa sostenibile ed
allo sviluppo di innovazione nel controllo delle fitopatie (es. genome editing),
mentre la sesta riporta un’ampia rassegna delle metodologie più recenti per la
diagnosi delle principali fitopatie.
Il libro è un testo unico nel suo genere, composto da 37 capitoli, ciascuno
dei quali è stato concepito e scritto dai principali esperti dello specifico
ambito trattato, e cerca di rappresentare al meglio tutti gli aspetti della ricerca
