Natural Variation of the Circadian Clock in Neurospora
Most living organisms on earth experience daily and expected changes from the rotation of the earth. For an organism, the ability to predict and prepare for incoming stresses or resources is a very important skill for survival. This cellular process of measuring daily time of the day is collectively called the circadian clock. Because of its fundamental role in survival in nature, there is a great interest in studying the natural variation of the circadian clock. However, characterizing the genetic and molecular mechanisms underlying natural variation of circadian clocks remains a challenging task. In this chapter, we will summarize the progress in studying natural variation of the circadian clock in the successful eukaryotic model Neurospora, which led to discovering many design principles of the molecular mechanisms of the eukaryotic circadian clock. Despite the success of the system in revealing the molecular mechanisms of the circadian clock, Neurospora has not been utilized to extensively study natural variation. We will review the challenges that hindered the natural variation studies in Neurospora, and how they were overcome. We will also review the advantages of Neurospora for natural variation studies. Since Neurospora is the model fungal species for circadian study, it represents over 5 million species of fungi on earth. These fungi play important roles in ecosystems on earth, and as such Neurospora could serve as an important model for understanding the ecological role of natural variation in fungal circadian clocks.Advances in Genetics, Vol. 99

The idea of combining discrete-event simulation and system dynamics has been a topic of debate in theoperations research community for over a decade. Untersuchungen zum interzellulären Diskurs von Keratinozyten und T-Zellen in einem Allergen-spezifischen in vitro Modell der Effektorphase der allergischen Kontaktdermatitis und des akuten atopischen Ekzems.
Das atopische Ekzem und die allergische Kontaktdermatitis gehören zu den häufigsten ekzematösen Erkrankungen. In dieser Arbeit konnten wir zeigen, dass Keratinozyten nicht nur Zielzellen dieser Entzündungsreaktion darstellen, sonder aktiv in das Geschehen eingreifen. Keratinozyten modulierten in in vitro Modellen der Ekzemreaktion die Proliferation und Zytokinproduktion in Allergen-spezifischen T-Zellklonen, unabhängig von deren Subtyp, meist im Sinne einer Hemmung. Die modulierende Kapazität der Keratinozyten konnte auf sezernierte Mediatoren zurückgeführt werden. Keratinozyten sind somit zentrale Regulatoren, die im Mikromilieu der Haut den Verlauf von Immunreaktionen entscheidend beeinflussen

Frequency Domain Estimation of Integrated Volatility for Ito Processes in the Presence of Market-Microstructure Noise
This paper proposes a novel multiscale estimator for the integrated volatility of an Ito process in the presence of market microstructure noise (observation error). The multiscale structure of the observed process is represented frequency by frequency, and the concept of the multiscale ratio is introduced to quantify the bias in the realized integrated volatility due to the observation error. The multiscale ratio is estimated from a single sample path, and a frequency-by-frequency bias correction procedure is proposed, which simultaneously reduces variance. We extend the method to include correlated observation errors and provide the implied time-domain form of the estimation procedure. The new method is implemented to estimate the integrated volatility for the Heston and other models, and the improved performance of our method over existing methods is illustrated by simulation studies
Sotatercept safety and effects on hemoglobin, bone, and vascular calcification
Introduction: Patients with end-stage kidney disease (ESKD) exhibit anemia, chronic kidney disease‒mineral bone disorder (CKD-MBD), and cardiovascular disease. The REN-001 and REN-002 phase II, multicenter, randomized studies examined safety, tolerability, and effects of sotatercept, an ActRIIA-IgG1 fusion protein trap, on hemoglobin concentration; REN-001 also explored effects on bone mineral density (BMD) and abdominal aortic vascular calcification.
Methods: Forty-three patients were treated in REN-001 (dose range: sotatercept 0.3‒0.7 mg/kg or placebo subcutaneously [s.c.] for 200 days) and 50 in REN-002 (dose range: 0.1‒0.4 mg/kg i.v. and 0.13‒0.5 mg/kg s.c. for 99 days).
Results: In REN-001, frequency of achieving target hemoglobin response (\u3e10 g/dl [6.21 mmol/l]) with sotatercept was dose-related and greater than placebo (0.3 mg/kg: 33.3%; 0.5 mg/kg: 62.5%; 0.7 mg/kg: 77.8%; 0.7 mg/kg [doses 1 and 2]/0.4 mg/kg [doses 3‒15]: 33.3%; placebo: 27.3%). REN-002 hemoglobin findings were similar (i.v.: 16.7%-57.1%; s.c.: 11.1%‒42.9%). Dose-related achievement of ≥2% increase in femoral neck cortical BMD was seen among only REN-001 patients receiving sotatercept (0.3‒0.7 mg/kg: 20.0%‒57.1%; placebo: 0.0%). Abdominal aortic vascular calcification was slowed in a dose-related manner, with a ≤15% increase in Agatston score achieved by more REN-001 sotatercept versus placebo patients (60%‒100% vs. 16.7%). The most common adverse events during treatment were hypertension, muscle spasm, headache, arteriovenous fistula site complication, and influenza observed in both treatment and placebo groups.
Conclusion: In patients with ESKD, sotatercept exhibited a favorable safety profile and was associated with trends in dose-related slowing of vascular calcification. Less-consistent trends in improved hemoglobin concentration and BMD were observed

<p>Either A549 lung carcinoma cells (A), or Bcap37 breast cancer cells(B) were implanted to nude mice of 6×10<sup>6</sup> tumor cells each, subcutaneously. Tumor-bearing mice were then treated for successively 15 days with rhEndostatin (5 mg/kg/day) intravenously (•), or at the site close to the tumor (▴). PBS was injected as the vehicle control at the site close to the tumor (▪). Results were presented as mean±SD(n = 10). Tumor volume was measured every other day, and the data indicated an enhanced reduction in tumor volume, when administered rhEndostatin s.c. peritumorally.</p
