    This thesis is a modern spelling edition of unedited poems and song lyrics from British Library, Additional Manuscript 15233, and a cultural and sociological study of the collection.The manuscript contains music, poetry and fragments of drama attributed to John Redford, Organist, Almoner and Master of the Choristers at St. Paul's Cathedral c.1534–1547, as well as work by at least six other mid Tudor poets. The manuscript has systematically been cited as a ‘St. Paul’s miscellany’(because the main body of work within it is attributed to Redford), and despite its varied content, has been considered almost solely from the perspective of Early Modern drama. This thesis considers the manuscript anew: as a whole, rather than in parts divided along disciplinary lines, as an example of material culture, and separately from the well-researched centres of St. Paul’s and John Redford.Chapters one and two comprise a study of the physical makeup of the manuscript, demonstrating new evidence for its date, and suggesting that it is the product of an artistic network centred on the London parish church of St. Mary-at-Hill.Chapters three and four comprise new studies of the content of the manuscript. Chapter three examines The Play of Wit and Science, with particular attention to its bibliographical status, and its engagement with contemporary artistic debate in performance. It also demonstrates its importance as a source for two later sixteenth-century plays. Chapter four is a case study of a single poem from the manuscript. This demonstrates the overall significance of MS 15233 as a source for verse and song, uncovers a network of printers involved in the transmission of its contents, and calls into question the long-standing theory that the Elizabethan poet George Gascoigne was a contributor to the manuscript.The final chapter of the thesis comprises a modern spelling edition of the poems and song lyrics from MS 15233 with individual commentaries and textual apparatus. This thesis demonstrates that to examine MS 15233 purely in relation to St. Paul’s Cathedral and John Redford, and from any one perspective, is reductive, and that these approaches have caused evidence to be skewed, and scholars to miss more complex possibilities regarding its compilation and provenance

    2-dimensional magnetization exchange spectroscopy of Anabaena 7120 ferredoxin- Nuclear Overhauser effect and electron self-exchange cross peaks from amino acid residues surrounding the 2Fe-2S-star cluster

    Hyperfine H-1 NMR signals of the 2Fe-2S* vegetative ferredoxin from Anabaena 7120 have been studied by two-dimensional (2D) magnetization exchange spectroscopy. The rapid longitudinal relaxation rates of these signals required the use of very short nuclear Overhauser effect (NOE) mixing times (0.5-20 ms). The resulting pattern of NOE cross-relaxation peaks when combined with previous 1D NOE results [Dugad, L. B., La Mar, G. N., Banci, L., & Bertini, I. (1990) Biochemistry 29, 2263-2271] led to elucidation of the carbon-bound proton spin systems from each of the four cysteines ligated to the 2Fe-2S* cluster in the reduced ferredoxin. Additional NOE cross peaks were observed that provide information about other amino acid residues that interact with the iron-sulfur cluster. NOE cross peaks were assigned tentatively to Leu27, Arg42, and Ala43 on the basis of the X-ray coordinates of oxidized Anabaena 7120 ferredoxin [Rypniewski, W. R., Breiter, D. R., Benning, M. M., Wesenberg, G., Oh, B.-H., Markley, J. L., Rayment, I., & Holden, H. M. (1991) Biochemistry 30, 4126-4131]. Three chemical exchange cross peaks were detected in magnetization exchange spectra of half-reduced ferredoxin and assigned to the H-1-alpha protons of Cys49 and Cys79 [both of whose sulfur atoms are ligated to Fe(III)] and Arg42 (whose amide nitrogen is hydrogen-bonded to one of the inorganic sulfurs of the 2Fe-2S* cluster). The chemical exchange cross peaks provide a means of extending assignments in the spectrum of reduced ferredoxin to assignments in the spectrum of the oxidized protein. Our results suggest that 2D magnetization exchange spectroscopy employing short mixing times will be useful for the assignment and characterization of hyperfine H-1 peaks in a variety of paramagnetic proteins

    We create a new multi-language corpus for author verification based on Wikipedia talkpages, and evaluate the influence that differences in topic and time have on character n-gram author profiles.     Appropriate Similarity Measures for Author Cocitation Analysis

    We provide a number of new insights into the methodological discussion about author cocitation analysis. We first argue that the use of the Pearson correlation for measuring the similarity between authors’ cocitation profiles is not very satisfactory. We then discuss what kind of similarity measures may be used as an alternative to the Pearson correlation. We consider three similarity measures in particular. One is the well-known cosine. The other two similarity measures have not been used before in the bibliometric literature.     Prevalence and aetiology of neuropathic pain in cancer patients: a systematic review

    Pain in cancer patients remains common and is often associated with insufficient prescribing of targeted analgesia. An explanation for undertreatment could be the failure to identify neuropathic pain mechanisms, which require additional prescribing strategies. We wanted to identify the prevalence of neuropathic mechanisms in patients with cancer pain to highlight the need for detailed assessment and to support the development of an international classification system for cancer pain. We searched for studies that included adult and teenage patients (age above 12 years), with active cancer and who reported pain, and in which a clinical assessment of their pain had been made. We found 22 eligible studies that reported on 13,683 patients. Clinical assessment methods varied, and only 14 studies reported confirmatory testing for either sensory abnormality or diagnostic lesion to corroborate a diagnosis of neuropathic pain. We calculated that the prevalence of patients with neuropathic pain (95% confidence interval) varied from a conservative estimate of 19% (9.4% to 28.4%) to a liberal estimate of 39.1% (28.9% to 49.5%) when patients with mixed pain were included. The prevalence of pain with a neuropathic mechanism (95% confidence interval) ranged from a conservative estimate of 18.7% (15.3% to 22.1%) to a liberal estimate of 21.4% (15.2% to 27.6%) of all recorded cancer pains. The proportion of pain caused by cancer treatment was higher in neuropathic pain compared with all types of cancer pain. A standardised approach or taxonomy used for assessing neuropathic pain in patients with cancer is needed to improve treatment outcomes

    Structures of microfilament destabilizing toxins bound to actin provide insight into toxin design and activity

    Marine macrolides that disrupt the actin cytoskeleton are promising candidates for cancer treatment. Here, we present the actinbound x-ray crystal structures of reidispongiolide A and C and sphinxolide B, three marine macrolides found among a recently discovered family of cytotoxic compounds. Their structures allow unequivocal assignment of the absolute configuration for each compound. A comparison of their actin-binding site to macrolides found in the trisoxazole family, as well as the divalent macrolide, swinholide A, reveals the existence of a common binding surface for a defined segment of their macrocyclic ring. This surface is located on a hydrophobic patch adjacent to the cleft separating domains 1 and 3 at the barbed-end of actin. The large area surrounding this surface accommodates a wide variety of conformations and designs observed in the macrocyclic component of barbed-end-targeting macrolides. Conversely, the binding pocket for the macrolide tail, located within the cleft itself, shows very limited variation. Functional characterization of these macrolides by using in vitro actin filament severing and polymerization assays demonstrate the necessity of the N-methyl-vinylformamide moiety at the terminus of the macrolide tail for toxin potency. These analyses also show the importance of stable interactions between the macrocyclic ring and the hydrophobic patch on actin for modifying filament structure and how this stability can be compromised by subtle changes in macrolactone ring composition. By identifying the essential components of these complex natural products that underlie their high actin affinity, we have established a framework for designing new therapeutic agents
