    Microangiography of the Humeral Condyle in Cocker Spaniel and Non-Cocker Spaniel Dogs

    SummaryThe purpose of this study was to evaluate the blood supply to the humeral condyle of dogs and compare the blood supply of Cocker Spaniel dogs to non- Cocker Spaniel dogs. We hypothesize that there is a congenitally altered blood supply of the humeral condyle in some Cocker Spaniels when compared to the blood supply of the humeral condyle in other breeds of dogs, which leads to abnormal ossification between the medial and lateral aspects of the humeral condyle, and predisposes them to fracture and delayed healing.Microangiography was performed on four Cocker Spaniel and four non- Cocker Spaniel dogs of similar size and weight. Qualitative and quantitative evaluation of the microangiographs of the humeral condyles revealed a decreased vascular density in the medial, midcondylar and lateral aspects of the condyle in the Cocker Spaniel dogs compared to the non-Cocker Spaniel dogs. This decreased vascular density may play a role in the increased incidence of humeral condylar fractures in the adult Cocker Spaniel dog without any or only minor trauma, and in the delayed healing seen in association with these fractures.A microradiographic comparison was made between the blood supply of the humeral condyles of Cocker Spaniel and other breeds of dogs. Contrast radiography and surgical excision showed communication of the tract with the left frontal sinus and rostral cerebral dura, respectively.     A large deletion in the GP9 gene in Cocker Spaniel dogs with Bernard-Soulier syndrome

    Inherited bleeding disorders including abnormalities of platelet number and function rarely occur in a variety of dog breeds, but are probably underdiagnosed. Genetically characterized canine forms of platelet disorders provide valuable large animal models for understanding similar platelet disorders in people. Breed-specific disease associated genetic variants in only eight different genes are known to cause intrinsic platelet disorders in dogs. However, the causative genetic variant in many dog breeds has until now remained unknown. Four cases of a mild to severe bleeding disorder in Cocker Spaniel dogs are herein presented. The affected dogs showed a platelet adhesion defect characterized by macrothrombocytopenia with variable platelet counts resembling human Bernard-Soulier syndrome (BSS). Furthermore, the lack of functional GPIb-IX-V was demonstrated by immunocytochemistry. Whole genome sequencing of one affected dog and visual inspection of the candidate genes identified a deletion in the glycoprotein IX platelet (GP9) gene. The GP9 gene encodes a subunit of a platelet surface membrane glycoprotein complex; this functions as a receptor for von Willebrand factor, which initiates the maintenance of hemostasis after injury. Variants in human GP9 are associated with Bernard-Soulier syndrome, type C. The deletion spanned 2460 bp, and included a significant part of the single coding exon of the canine GP9 gene on dog chromosome 20. The variant results in a frameshift and premature stop codon which is predicted to truncate almost two-thirds of the encoded protein. PCR-based genotyping confirmed recessive inheritance. The homozygous variant genotype seen in affected dogs did not occur in 98 control Cocker Spaniels. Thus, it was concluded that the structural variant identified in the GP9 gene was most likely causative for the BSS-phenotype in the dogs examined. These findings provide the first large animal GP9 model for this group of inherited platelet disorders and greatly facilitate the diagnosis and identification of affected and/or normal carriers in Cocker Spaniels

    Association between anal sac gland carcinoma and dog leukocyte antigen-DQB1 in the English Cocker Spaniel

    Anal sac gland carcinomas occur frequently in English Cocker Spaniels and, to a lesser extent, in other spaniel breeds. The disease typically presents in dogs aged 8 years or older and frequently metastasises to the local lymph nodes. The association between anal sac gland carcinoma in English Cocker Spaniels and the major histocompatibility complex class II loci (the dog leukocyte antigen loci DLA-DRB1, -DQA1, -DQB1) was investigated in 42 cases and 75 controls. Based on a corrected error rate of 0.017 for each test, the allele distribution in DLA-DRB1 showed no significant difference between cases and controls (Pvalue = 0.019), while a significant difference was obtained for DLA-DQA1 and -DQB1 alleles (P values are 0.010 and 3.3 × 10-5). The DLA-DQB1*00701 allele was the most common in both cases and controls, but it had a higher frequency among the former (0.89) than in the latter (0.61), while the second most common allele had a higher frequency in the controls (0.23) than in the cases (0.07). Haplotype distributions were also significantly different between the two groups (Pvalue = 1.61 × 10-4). This is the second disease in English Cocker Spaniels for which the most common DLA-DQB1 allele in the breed has been shown to have a higher frequency in cases than controls, while the second most common allele in the breed (*02001) has a significantly higher frequency in the controls, compared with the cases. © 2010 John Wiley &amp; Sons A/S

    Circular bioeconomy potential and challenges within an African context: From theory to practice

    A circular bioeconomy has the potential to minimize the environmental impacts of biowaste while simultaneously generating value-added bioproducts and bioenergy. Currently, most countries of the African Union lack well-defined policies, requisite infrastructure, and expertise for biowaste valorisation, thus limiting the potential development of the region. Against this background, it is necessary to deploy circular bioeconomy principles based on the awareness of the biocapacity of territories through the nexus of biowaste management and life cycle thinking. In the present study, a preliminary assessment of waste management practices in a tourist hotel in Victoria Falls in Zimbabwe is explored. The hotel produces about 3.26 tons per month of biowaste, which is often improperly disposed in non-engineered waste dumps. Furthermore, the disposal options for 1 tonne of biowaste are explored using City of Harare (CoH) as a case study. The preliminary results show composting as the most environmentally favourable option (9.6 kg CO2 eq), followed by anaerobic digestion (56.4 kg CO2 eq), and finally, biowaste incineration (140 kg CO2 eq). Anaerobic digestion and composting remain the most viable biowaste disposal alternatives in Africa, due to limited expenses and expertise for construction, operation, and maintenance. However, both technologies remain under-utilized, hence, a significant portion of the source-separated biowaste is still disposed of in waste dumps and this reflects the lack of supportive institutional, regulatory and policy frameworks. Overall, these early results point to the potential to develop a circular bioeconomy in Africa, while calling for shared responsibilities among the state, market, and civil society actors to develop and adopt appropriate institutional, regulatory, policy and funding models

    Experiences of a proactive IR(ME)R inspection in radiotherapy.

    The Ionizing Radiation (Medical Exposure) Regulations 2000, IR(ME)R, apply to the safety of the patient referred for a medical exposure to ionizing radiation. In Scotland, the Scottish Executive (Department of Health) is responsible for carrying out inspections of compliance with these regulations. IR(ME)R specifically addresses issues concerned with Employer's duties, responsibilities of the Practitioner, Operator and Referrer, justification of individual medical exposures for diagnosis and treatment, optimization of all procedures, clinical audit and adequate training of all duty holders. A proactive IR(ME)R inspection of the Clinical Oncology Department, Raigmore Hospital, Inverness, was carried out in November 2001 by inspectors based at the Department of Health, London, and seconded by the Scottish Executive, Department of Health. The aim of the inspection was to assess the degree of compliance with the regulations. In this case study the experiences of a proactive inspection are described in detail and some of the important elements of implementing IR(ME)R in a department that operates an ISO 9000-2000 Quality Management System addressed. The identification of IR(ME)R Duty Holders' responsibilities is one important aspect which may be inadequately described by the existing Quality Management System documentation. Other key elements of the inspection include the methods of authorizing the justification, the importance of the treatment prescription sheets in the demonstration of compliance with IR(ME)R, patient identification and pregnancy questions and dose recording procedures. The integration of the standard operating procedures as described in Schedule 1 of the regulations is also important. Where the existing Quality Management System documentation is written to include the IR(ME)R requirements of duty holder's responsibilities and the allocation of all the important tasks, then there is no need to re-badge these documents for IR(ME)R purposes. IR(ME)R encourages departments to focus on the safety of the patient and to document good practice. In order to comply, departments will have to show evidence of optimization of their procedures and must address the clinical governance issues associated with delivery of treatment

    Panniculitis associated with pancreatitis in a cocker spaniel

    A 13-year-old, male cocker spaniel presented with a history of inappetence, depression and reluctance to stand. The dog had multiple, ulcerated skin lesions which were diagnosed as panniculitis by histopathology. A diagnosis of pancreatitis was made on the basis of markedly elevated serum lipase concentrations, abdominal ultrasonography which showed an abnormal lobulated area of hypoechoic tissue in the body and right lobe of the pancreas, and a fine needle biopsy from this area which revealed large numbers of degenerate neutrophils. After treatment with antibiotics and prednisolone, the dog made a full clinical recovery and was free of clinical signs for four months. The dog was euthanased five months later and postmortem examination revealed chronic, active pancreatitis and a pancreatic adenoma. This is the first report of antemortem diagnosis of pancreatitis and panniculitis in a dog
