The novel NOX inhibitor 2‐acetylphenothiazine impairs collagen‐dependent thrombus formation in a GPVI‐dependent manner
Background and Purpose NADPH oxidases (NOXs) contribute to platelet activation by a largely unknown mechanism. Here, we studied the effect of the novel NOX inhibitor 2-acetylphenothiazine (2-APT) on human platelet functional responses and intracellular signaling pathways. Experimental Approach The generation of superoxide ions was assessed by single cell imaging on adhering platelets using dihydroethidium (DHE), while other reactive oxygen species (ROS) were detected with 5-(and-6)-carboxy-2',7'-dichlorodihydrofluorescein diacetate (CM-H2-DCFDA). Whole blood thrombus formation, washed platelet aggregation, integrin aIIb beta 3 inside-out signalling, Syk phosphorylation and PKC activation were analysed to understand the functional consequences of NOX inhibition by 2-APT in platelets. Key Results Superoxide ion generation stimulated by platelet adhesion on collagen and fibrinogen was significantly inhibited by 2-APT in concentration-dependent manner (IC50 = 306?nM and 227?nM, respectively), whereas cumulative ROS accumulation was not affected by this pharmacological agent. 2-APT also abolished collagen-dependent whole blood thrombus formation and washed platelet aggregation in response to collagen but not thrombin. The activation of integrin aIIb beta 3 and PKC in response to the GPVI-specific agonist collagen-related peptide (CRP) was significantly reduced, whereas the same responses to thrombin were not significantly affected by 2-APT. Finally, Syk activation in response to collagen but not thrombin was inhibited by 2-APT. Conclusions and Implications Taken together, our results suggest that 2-APT attenuates GPVI-specific signalling and is a novel inhibitor of collagen-induced platelet responses. Therefore, NOXs could represent a novel target for the discovery of anti-thrombotic drugs

Statistical soil characterization of Italian sites for reliability analyses
The paper is a review of the most commonly used techniques and methods for the
estimation of inherent variability and uncertainties concerning soil physical and mechanical properties
estimation. The methods illustrated in the following as, among others, intraclass correlation coefficient
method, the Vivatrat procedure, the Cusum method and the Bartlett modified method are described
theoretically and applied to three types of Italian soils in order to give useful suggestions for common
geotechnical design activity. Furthermore reliability approach is also presented and some classical methods
are dealt with here as FORM (first order reliability method) and SORM (second order reliability
method). Finally an example of the reliability analysis application to a pile design is presented and
discussed. Accordingly the evaluation of the reliability index of the global safety factor is carried out
in order to show that the variation the safety factor can suffer on its level of reliability by taking into
account the variability structure of physical and mechanical soil properties
Volleyball at University of Pula
Odbojka je nastala krajem 19. stoljeća kada ju je Amerikanac William G. Morgan izmislio. Njegova ideja je bila ta odbojka bude igra koja bi svojom jeftinom i jednostavnom opremom bila pristupačna svim uzrastima bez obzira na spol. 25. lipnja 1946. godine nastaje Hrvatski odbojkaški savez koji i dan danas organizira sva natjecanja u istoimenom sportu. Odbojka dobiva veći razvoj tek nakon Drugog svjetskog rata, onda kada su stvoreni svi potrebni uvjeti za njen masovan i kvalitetan razvoj. Danas je odbojka među popularnijim sportovima u svijetu. Sve više sportaša se uključuje u taj sport, a u zadnje vrijeme sve je češća prisutna volja i želja za učenjem tog sporta kod rekreativaca. U ovom radu spominje se mogućnost bavljenja odbojkom i u predškolskoj dobi što može znatno pridonositi razvoju svakog djeteta u ranom djetinjstvu i kasnije u životu. Studenti Sveučilišta Jurja Dobrile dali su svoj doprinos aktivnim učešćem u raznim sportovima i aktivnostima na Sveučilištu i ostvarili brojne kvalitetne rezultate.Volleyball originated in the late 19th century when American William G. Morgan invented it. His idea was that volleyball would be a game that would be accessible to all ages, regardless of gender, with its cheap and simple equipment. 25.th June 1946., the Croatian Volleyball Association was formed, and today it organizes all competitions in the sport of the same name. Volleyball only got more development after the Second World War, when all the necessary conditions for its massive and quality development were created. Today, volleyball is among the most popular sports in the world. More and more athletes are getting involved in this sport, and lately there is an increasing willingness and desire to learn the sport from recreational athletes. Students of the University of Juraj Dobrila contributed to their active participation in various sports and activities at the University, achieving numerous quality results, which is the theme of this thesis
Autocrine amplification of integrin alpha IIb beta 3 activation and platelet adhesive responses by deoxyribose-1-phosphate
Using direct injection mass spectrometry (DIMS) we discovered that deoxyribose-1-phosphate (dRP) is released by platelets upon activation. Interestingly, the addition of exogenous dRP to human platelets significantly increased platelet aggregation and integrin αIIbβ3 activation in response to thrombin. In parallel, genetically modified platelets with double genetic deletion of thymidine phosphorylase and uridine phosphorylase were characterised by reduced release of dRP, impaired aggregation and decreased integrin αIIbβ3 activation in response to thrombin. In vitro platelet adhesion onto fibrinogen and collagen under physiological flow conditions was potentiated by treatment of human platelets with exogenous dRP and impaired in transgenic platelets with reduced dRP release. Human and mouse platelets responded to dRP treatment with a sizeable increase in reactive oxygen species (ROS) generation and the pre-treament with the antioxidant apocynin abolished the effect of dRP on aggregation and integrin activation. Experiments directly assessing the activation of the small G protein Rap1b and protein kinase C suggested that dRP increases the basal levels of activity of these two pivotal platelet-activating pathways in a redox-dependent manner. Taken together, we present evidence that dRP is a novel autocrine amplifier of platelet activity, which acts on platelet redox levels and modulates integrin αIIbβ3.</p
