    Harnessing Marine Renewable Energy from Poole Harbour: Case Study

    Global warming and its impact on our environment, society, economies and security is one of the fundamental concerns of our time. In response, the United Kingdom government has put in place a legally binding target of an 80% reduction in greenhouse gas emissions on 1990 levels by 2050. The United Kingdom will need to achieve a tenfold expansion of energy supply from renewable sources by 2020 to meet its share of the European Union renewable energy target. The marine and coastal environment’s renewable energy potential in Britain is high. It is estimated that it has 50% of the tidal energy, 35% of wave and 40% of wind resources in the European Union. Use of geothermal resources using heat pump technology is the least evolved sector, but in 2010 contributed to 0.7 TWh of energy and it is believed that non domestic heat pumps could contribute up to 22 TWh by 2020. In the Southwest of England, Poole Harbour has been recognised as a potential, highly predictable source of tidal and heat energy. Local groups are embarking on a feasibility study for harnessing this energy for the benefit of the community. The purpose of this article is to examine the potential conflict of interest between the laudable aims of promoting the use of renewable energy and of safeguarding ecosystems and their biodiversity. Using Poole Harbour as a case study, it will consider the environmental and economic costs and benefits of a Community Renewable Energy project (the Poole Tidal Energy Partnership) in the context of an area subject to a number of statutory and non-statutory designations to protect nationally and internationally important habitats and species. The paper identifies key environmental legislation, including spatial planning law and policy, which will facilitate exploring whether there is potential for reconciling what may be perceived as competing objectives for sustainable development

Physiological Reports, 5(5), 10. doi:10.14814/phy2.13180In contrast to cardiomyocytes, store overload-induced calcium ion (Ca2+) release (SOICR) is not considered to constitute a primary Ca2+ releasing system from the sarcoplasmic reticulum (SR) in skeletal muscle myocytes. In the latter, voltage-induced Ca2+ release (VICR) is regarded as the dominant mechanism facilitating contractions. Any role of the SOICR in the regulation of cytoplasmic Ca2+ concentration ([Ca2+](i)) and its dynamics in skeletal muscle in vivo remains poorly understood. By means of in vivo single fiber Ca2+ microinjections combined with bioimaging techniques, we tested the hypothesis that the [Ca2+](i) dynamics following Ca2+ injection would be amplified and fiber contraction facilitated by SOICR. The circulation-intact spinotrapezius muscle of adult male Wistar rats (n = 34) was exteriorized and loaded with Fura-2 AM to monitor [Ca2+](i) dynamics. Groups of rats underwent the following treatments: (1) 0.02, 0.2, and 2.0 mmol/L Ca2+ injections, (2) 2.0 mmol/L Ca2+ with inhibition of ryanodine receptors (RyR) by dantrolene sodium (DAN), and (3) 2.0 mmol/L Ca2+ with inhibition of SR Ca2+ ATPase (SERCA) by cyclopiazonic acid (CPA). A quantity of 0.02 mmol/L Ca2+ injection yielded no detectable response, whereas peak evoked [Ca2+](i) increased 9.9 +/- 1.8% above baseline for 0.2 mmol/L and 23.8 c 4.3% (P < 0.05) for 2.0 mmol/L Ca2+ injections. The peak [Ca2+](i) in response to 2.0 mmol/L Ca2+ injection was largely abolished by DAN and CPA (-85.8%, -71.0%, respectively, both P < 0.05 vs. unblocked) supporting dependence of the [Ca2+](i) dynamics on Ca2+ released by SOICR rather than injected Ca2+ itself. Thus, this investigation demonstrates the presence of a robust SR-evoked SOICR operant in skeletal muscle in vivo

Orchestrated by selective breeding for physiology and biomechanics, expressed as speed, the millennia-old symphony of man and beast reaches its climax. At nearly 73 kilometers per hour (45 mph) over half a ton of flesh and bone dwarfs its limpet-like jockey as, eyes wild and nostrils flaring, their necks stretch for glory. Beneath each resplendent livery-adorned, latherin-splattered coat hides a monstrous heart trilling at 4 beats per second, and each minute, driving over 400 L (105 gallons) of oxygen-rich blood from lungs to muscles. Matching breath to stride frequency, those lungs will inhale 16 L (4 gallons) of air each stride moving >1,000 L/min in and out of each nostril - and yet failing. Engorged with blood and stretched to breaking point, those lungs can no longer redden the arterial blood but leave it dusky and cyanotic. Their exquisitely thin blood-gas barrier, a mere 10.5 mu m thick (1/50,000 of an inch), ruptures, and red cells invade the lungs. After the race is won and lost, long after the frenetic crowd has quieted and gone, that blood will clog and inflame the airways. For a few horses, those who bleed extensively, it will overflow their lungs and spray from their nostrils incarnadining the walls of their stall: a horrifically poignant canvas that strikes at horse racing's very core. That exercise-induced pulmonary hemorrhage (EIPH) occurs is a medical and physiological reality. That every reasonable exigency is not taken to reduce/prevent it would be a travesty. This review is not intended to provide an exhaustive coverage of EIPH for which the reader is referred to recent reviews, rather, after a brief reminder of its physiologic and pathologic bases, focus is brought on the latest developments in EIPH discovery as this informs state-of-the-art knowledge, the implementation of that knowledge and recommendations for future research and treatment

    Rapid method for proline determination in grape juice and wine

    Proline is typically the most abundant amino acid present in grape juice and wine. The amount present is influenced by viticultural and winemaking factors and can be of diagnostic importance. A method for rapid routine quantitation of proline would therefore be of benefit for wine researchers and the industry in general. Colorimetric determination utilizing isatin as a derivatizing agent has previously been applied to plant extracts, biological fluids, and protein hydrolysates. In the current study, this method has been successfully adapted to grape juice and wine and proved to be sensitive to milligram per liter amounts of proline. At sugar concentrations above 60 g/L, interference from the isatin-proline reaction was observed, such that proline concentrations were considerably underestimated in grape juice and dessert wine. However, the method was robust for the analysis of fermentation samples and table wines. Results were within ±10% agreement with data generated from typical HPLC-based analyses. The isatin method is therefore considered suitable for the routine analysis required to support research into the utilization or release of proline by yeast during fermentation.Danfeng Long, Kerry L.
