No full textBrief project description:As part of this project, human cybrid cell lines carrying a mixture of neutral mtDNA haplotypes differing in multiple nucleotide positions but not harboring pathogenic variants were generated. The aim of this study was to investigate the presence of heteroplasmy selection and intermolecular mtDNA recombination in humans in vitro.To generate cybrids containing a mixture of two mtDNA haplotypes, human 143B.TK- cells devoid of mtDNA (ρ0) were fused with enucleated cells serving as mitochondrial donors (cytoplasts). Six combinations of four mtDNA haplotypes (A, B, C, and D) were used, varying in genetic distance measured by the number of nucleotide differences. The nucleotide sequences of the studied mtDNA haplotypes were deposited in the GenBank database under accession numbers PQ468425 (A), PQ468426 (B), PQ468427 (C), and PQ468428 (D).Individual cell clones were screened for the presence and level of mtDNA heteroplasmy. mtDNA heteroplasmy was monitored over successive passages during cell culture to assess the occurrence of selection, its direction, and rate. Preliminary assessment was performed using Sanger sequencing of a selected mtDNA region (a fragment of the non-coding control region characterized by the highest variability). Detailed analysis of heteroplasmic cybrid lines was subsequently performed using high-throughput whole mtDNA sequencing using a method developed and implemented in our group's research on the MiSeq platform (Illumina) (https://doi.org/10.1016/j.exer.2018.10.004). The occurrence of intermolecular recombination in mtDNA in human cell culture in vitro was assessed by direct sequencing of native mtDNA molecules using Nanopore sequencing technology.The dataset consists of the following zip archives:Sanger_data.zip – DNA traces/chromatograms from Sanger sequencing (*.ab1) of selected mtDNA region for individual cybrid cell clones at different passages. The files can be opened using Sequencher software or any other similar bioinformatics software for Sanger data analysis.Illumina_FASTQ_files.zip – raw sequencing data as FASTQ files (*.fastq.gz) from next-generation sequencing (NGS) of mtDNA from heteroplasmic cybrid cell lines at different cell passage points. The files can be opened using CLC Genomics Workbench software, any other similar bioinformatics software, or open-source bioinformatics tools for NGS data analysis.Illumina_BAM_files.zip – mapped reads as BAM files and their corresponding index files (*.bam and *.bam.bai) generated from the bioinformatics analysis and mapping of mtDNA NGS data to the human mtDNA reference genome, rCRS (NC_012920). The files can be opened using CLC Genomics Workbench software, any other similar bioinformatics software, or open-source bioinformatics tools for NGS data analysis.Illumina_variant_lists.zip – Lists of mtDNA variants (SNVs and indels) identified from the bioinformatics analysis of mtDNA NGS data (*.xlsx). The files can be opened with MS Excel or any other spreadsheet software.ONT_data.zip – raw sequencing data as FASTQ files (*.fastq.gz) of long reads from direct sequencing of native mtDNA molecules using Oxford Nanopore Technology (ONT). No full textThe article outlines Polish literature dedicated to the phenomenon of flamenco. An author not only presents the state of research on flamenco in Poland, but also refers readers both to the earliest and to the most popular works on flamenco functioning abroad. While analyzing Polish literature Piotrowska asks a question about general attitude of Polish authors towards the issue of ‘Gypsyness’ of flamenco and presents how Polish writers treat Gypsy roots of flamenco. The author concludes that in Polish literature – comprising a few articles and books – Gypsy connotations of flamenco are usually duly underlined. At the same time, Piotrowska states that Polish literature on flamenco is dominated by practical approach focusing on performantive practices (dance, instrumental play), rather than on historic or aesthetic perspective. The author stresses the absence of Polish translations of important books on flamenco, however she also acknowledges the role Polish Internet sites play in promoting the Gypsy aspect of flamenco phenomenon.Natalia Gancar

No full textIn Black and White Agnieszka Piotrowska presents a unique insight into the contemporary arts scene in Zimbabwe – an area that has received very limited coverage in research and the media. The book combines theory with literature, film, politics and culture and takes a psychosocial and psychoanalytic perspective to achieve a truly interdisciplinary analysis. Piotrowska focuses in particular on the Harare International Festival of the Arts (HIFA) as well as the cinema, featuring the work of Rumbi Katedza and Joe Njagu. Her personal experience of time spent in Harare, working in collaborative relationships with Zimbabwean artists and filmmakers, informs the book throughout. It features examples of their creative work on the ground and examines the impact it has had on the community and the local media. Piotrowska uses her experiences to analyse concepts of trauma and post-colonialism in Zimbabwe and interrogates her position as a stranger there, questioning patriarchal notions of belonging and authority. Black and White also presents a different perspective on convergences in the work of Doris Lessing and iconic Zimbabwean writer Dambudzo Marechera, and how it might be relevant to contemporary race relations. Black and White will be intriguing reading for psychoanalysts, psychotherapists and psychotherapeutically engaged scholars, film makers, academics and students of post-colonial studies, film studies, cultural studies, psychosocial studies and applied philosophy

No full textReformulation and Acquisition of Linguistic Complexity proposes a new answer to the question of the appropriation or acquisition of a mother tongue – a complex object, one that is both stable and perpetually evolving. This answer is based on the reformulating principle that children spontaneously apply; a principle that is illustrated here with children retelling the same story. These children are all 6, 8 or 10 years old and speak French, Italian, Croatian or Polish as a first language. This book demonstrates that the acquisition of any mother tongue is explained by the application of various reformulation procedures between source predications and reformulated predications. These procedures are comparable from one language to another, and different from one age group to another. This book also studies certain complex phenomena at the lexical and syntactical levels, and analyzes how children, depending on their age, treat these phenomena. Finally, we show that the acquisition of a mother tongue is a fundamentally linguistic activity
