Cytochrome oxidase subunit VI of Trypanosoma brucei is imported without a cleaved presequence and is developmentally regulated at both RNA and protein levels
No full textMitochondrial respiration in the African trypanosome undergoes dramatic developmental stage regulation. This requires co-ordinated control of components encoded by both the nuclear genome and the kinetoplast, the unusual mitochondrial genome of these parasites. As a model for understanding the co-ordination of these genomes, we have examined the regulation and mitochondrial import of a nuclear-encoded component of the cytochrome oxidase complex, cytochrome oxidase subunit VI (COXVI). By generating transgenic trypanosomes expressing intact or mutant forms of this protein, we demonstrate that COXVI is not imported using a conventional cleaved presequence and show that sequences at the N-terminus of the protein are necessary for correct mitochondrial sorting. Analyses of endogenous and transgenic COXVI mRNA and protein expression in parasites undergoing developmental stage differentiation demonstrates a temporal order of control involving regulation in the abundance of, first, mRNA and then protein. This represents the first dissection of the regulation and import of a nuclear-encoded protein into the cytochrome oxidase complex in these organisms, which were among the earliest eukaryotes to possess a mitochondrion

Cyclopentyl-adenosine decreases caspase-3 activity and LDH release following simulated ischemia in cerebellar granule neurons
No full textActivation of adenosine A1 receptors in vivo or in vitro, prior to simulated ischemic insults have been demonstrated to be neuroprotective. Preconditioning with A1 receptor agonists, both in vivo and in vitro, has been shown to induce neuroprotective effects against necrosis even when administered 24–72 hours before ischemic insult. This thesis examined the neuroprotective effects of the selective A1 receptor agonist, N6-cyclopentyl-adenosine (CPA), against both apoptotic and necrotic cell death following simulated ischemia in cultured rat cerebellar granule neurons. Using antibodies directed towards neuronal nuclear protein and glial fibrillary acid protein, the purity of cerebellar granule cell cultures was found to be 98.6 ± 0.4%. This indicates that the results from these experiments were likely mediated by effects of the drugs and simulated ischemia on neurons and not astrocytes. To determine if the neuroprotection induced by CPA treatment was mediated by activation of A1 receptors, a concentration response curve for DPCPX was generated in the presence and absence of 1μM CPA. No full textThis paper reports on the development of an RF IV waveform based stress test procedure. DC and low-voltage RF characterisation was carried out before and after high power RF stress. RF waveform measurements showed that the exact change in the RF load line induced during RF degradation cannot be directly inferred from the DC or low power RF measurement. The RF degradation takes the form of a knee-walkout, a small pinch-off shift consistent with charge trapping and defect generation, and in addition gate leakage occurs once the RF voltage exceeds a critical voltage. The work is raw, intensely physical, and honest. We all know the experience of getting ourselves stuck in behaviors or relationships or feelings that we just can’t break out of. This work takes that idea and turns it into a dance. You can feel our energy and our frustration as we fight against the pathways we find ourselves on.
