    No full text

Constraint-based modelling of mixed microbial populations: Application to polyhydroxyalkanoates production

Dissertação para obtenção do Grau de Doutor em Engenharia Química e BioquímicaThe combined use of mixed microbial cultures (MMC) and fermented feedstock as substrate may significantly decrease polyhydroxyalkanoates (PHA) production costs and make them more competitive in relation to conventional petroleum-based polymers. However, there still exists a lack of knowledge at metabolic level that limits the development of strategies to make this process more effective. In this thesis, system biology computational tools were developed and applied to PHA production by MMC from fermented sugar cane molasses, rich in volatile fatty acids (VFA). Firstly, a metabolic network able to describe the uptake of complex mixtures of VFA and PHA production was defined. This metabolic network was applied to metabolic flux analysis (MFA) to describe substrate uptake and PHA production fluxes over the enrichment time of a culture submitted to the feast and famine regimen. Then, the minimization of the tricarboxylic acid cycle (TCA) fluxes was identified as the key metabolic objective of a MMC subjected to this regimen by flux balance analysis (FBA). This model enabled to predict, with an acceptable accuracy, the PHA fluxes and biopolymer composition. Subsequently, data gathered from microautoradiography-fluorescence in situ hybridization (MAR-FISH) was used to develop a segregated FBA model able to predict the flux distribution for the three populations identified in the enriched culture. These results were slightly better than those obtained by the non-segregated FBA and were consistent with MFA results. Finally, a dynamic metabolic model was proposed based on the previous models and on a regulatory factor for VFA uptake and PHA production. This model allowed to identify the dynamics of the process and regulatory factor as well as to validate the previous results. Globally, this thesis enabled to demonstrate the potential of using computational tools to understand and optimize PHA production by MMC

Wyobrażenia historii w fi lmach Wojciecha Jerzego Hasa, Jana Jakuba Kolskiego, Filipa Bajona i Anny Jadowskiej Łódź 2013. According to the reviewer, the book is a bold attempt to combine film analysis with social issues. Korczarowska-Różycka proposed her own perspective of film reading on the intersection of close film analysis, sociological attitude and historical reflections about collective memory. She sees movie as a document of some socially established historical points of view or images, but also as a dynamic force, that is transforming and recreating these images. Her selection of directors and applied methodological basis allow her to avoid clichéd reflections on genre of historical cinema, but also lay her work open to the charge of certain arbitrariness. The author tries to not only present Korczarowska-Różycka’s book, but also bring out its theoretical background and place it in the broader context of memory studies.Translated by Aleksandra IdczakTHREE LOOKS.

    No full text
The primary objective of this essay is to reflect on the connection between public and private scopes represented in Augusto Abelaira’s (1926-2003) work. Abelaira’s literary production was published along the second half of 20th century and consists mostly of novels in which dialogue is a privileged device, rather than action, in the creation and definition of its characters, emphasizing their debates and ideas as well as their relations. The author conceives his work as a «one book», of which every novel only stands for a part: in this framework, the focus on both erotic life and historical condition of their characters is always an essential part of the diegetic universe. Both individual and collective levels constitute a scenario in which the subject tries to react to the paralysis of Portuguese political, social, and cultural life during Salazar’s dictatorship: a reality that they perceive as stagnating and made of bolor (mould), quoting the title of one of the most iconic novels by Abelaira. The characters’ disenchantment about love and the possibility of reaching their fulfilment mirrors their incapability in understanding the reality that surrounds them and in transforming it. This situation generates a permanent tension between an ideal of plenitude and contingence, a conflict without any solution. The erotic universe and the utopic ideals are, then, the starting point for a reflection on the possibilities and value of human action in a reality that denies the subject a concretization of their ambitions

The objective of bachelors thesis is to explain trends of economical development based on the Phillips curve analysis with data from USA and Latvia. Finding relations in trend changes can give a good base to further study of the Phillips curve. Also finding experimental solutions in alternative analysis related to the Phillips curve. As a result of analytical development research of USA economy in 1950. – 2008. year period and development of Latvia in 2001. – 2008. year period there were made following conclusions: •Tradeoff in the Phillips curve exists at all times however it is not always seen in the trend from actual data of unemployment rate and inflation. Reason is that shape of Phillips curve can be modeled by using only demand driven inflation and position is determined by supply driven inflation. •Methodology used by author excludes latter flaw because instead of actual Phillips curve analysis - author analyses direction of changes in coordinates that represent unemployment rate and inflation at the given period. There is a clear relation between movement in the Phillips curve and the economic cycle. There is a force of development in the economy which appears in the expansion period followed by turning point finding which can help forecasting crisis. Then followed by either types of recession – stagflation or stagnation. •Author presents alternative methodology for the economical analysis. Inflation and unemployment rate correlation method which can reveal approximate structure of inflation structure. Analysis of indexes present in the trend of the Phillips curve reveal how these indexes correlate with average GDP growth and amplitude of inflation. Also the Phillips curve is used to determine forecasts of the unemplyment rate using existing inflation forecasts. In the USA forecasted unemplyment rate is in between 8.5% and 10%. In Latvia this forecast is 27%. Keywords: Phillips curve, unemployment rate, demand inflation, supply inflation, economical cycle

    No full text
Within the intercultural activity that the Portuguese Crown developed in the 16th-18th centuries, the reports on the New Worlds occupied an important space, supported by a relevant manuscript production in Portuguese, Latin and ‘exotic’ languages (Zwartjes, 2011, p.1). Non-native figures also contributed to the circulation of this knowledge; they considered the Lusitan vernacular a comprehensive, influential and respected linguistic environment at the crossroads of colonial empires. In this study, we will deal with manuscript 536, «Noticias do Reyno do Malabár», without signature and date, and preserved in the National Library of Lisbon: we will put it in dialogue with other archive sources, by analyzing its contents in the light of the socio-cultural and linguistic panorama in which it was probably written. We will also reflect on the explicit and implicit positions adopted by the author – an indigenous priest from Malabar (India), a student of the Collegio Urbano de Propaganda Fide in Rome – concerning the information conveyed in the work, and we will finally highlight the role of the Portuguese language at the time of the decline of its prestige in the colonial network. That the testimony of our interest offers, both for the data itself and for the way they are collected and presented, is a clear adherence to a cultural, religious and linguistic policy focused on the construction of a copious collection of texts, of different typologies; texts concerning the knowledge of non-European lands and peoples – although filtered by the Eurocentric perspective – which constitute the foundations for the action of evangelization and the maintenance of the possessions

These clonal shifts were primarily explained by an increased antimicrobial resistance profile. Here we explore genomic and biochemical differences among these and more recent clones, which could further explain the diversity and evolution of this species.info:eu-repo/semantics/publishedVersio
