    Influence of crystal hydration on the mechanical properties of sodium naproxen

    The aim of this work is to establish a correlation between water uptake by anhydrous sodium naproxen (ASN) at two different relative humidities and modifications in tableting and densification behaviour under hydration. Water uptake was evaluated at different relative humidities. Models for the hydration kinetics of ASN at 55% and 86%. corresponding to the formation of the dihydrated and tetrahydrated forms, respectively, were evaluated assuming Eyring's dependence on temperature. Tabletability, compressibility, compactibility, and densification behaviour were determined using an instrumented single punch tablet machine. Kinetic data are consistent with a model where water molecules enter the crystal preferentially along hydrophilic tunnels existing in the crystal structure and corresponding to the propionate side chain. Water inclusion perturbs the crystallographic structure, causing slight structural changes according to the amount and associated to an increase in entropy. The interposition of water molecules between sodium naproxen molecules weakens intermolecular bonds, and these sites can behave like sliding planes under compression. Such structural changes may explain the improved compression behaviour and modified densification propensity mechanism. Kinetic data describing the water hydration mechanism of ASN explain in an original way the improved tableting and densification properties under hydration. (C) 2008 Elsevier B.V.     Characterization and compaction behaviour of nimesulide crystal forms.

    Nimesulide is a typical nonsteroidal anti-inflammatory drug (NSAID), widely used in solid oral formulations. By crystallizing nimesulide from an ethanol solution a crystalline form was obtained, different from the reference sample, as confirmed by X-ray powder diffraction (XRPD), Differential Scanning Calorimetry (DSC) and solid cross polarization-magic angle spinning (C-13-CPMAS) NMR. Moreover, when crystallized from dioxane nimesulide forms a solvate. The solvate was characterized by XRPD, IR-spectrometry, DSC, thermo-gravimetric analysis (TGA) and by C-13-CPMAS NMR. In particular, through this technique, the presence of several conformational isomers was demonstrated. In addition to the physico-chemical characterization, the technological properties of nimesulide, namely densification and tableting, were evaluated. Contrarily to the other forms that are affected by capping phenomena at increasing compression pressures, the form obtained by desolvation of dioxane solvate has positive effect on tableting properties, increasing both compressibility and tabletability of nimesulide.     2018 - 2019La tesi di dottorato consta di cinque capitoli e due appendici, e propone l’edizione critica della versione latina di Burgundio of Pisa del trattato galenico De causis pulsuum (XII s.). Dopo una presentazione generale sul testo greco e sull’impatto della scienza galenica del polso in Occidente nel XII secolo, seguono un breve profilo del traduttore e un’introduzione alla sua versione del De causis pulsuum, limitata ai soli libri III-IV (commento lemmatico al De pulsibus ad tirones). Vengono indagati il metodo di traduzione e il rapporto di interdipendenza tra le traduzioni di De causis pulsuum e De pulsibus ad tirones, per giungere a una proposta di datazione nella cronologia relativa delle traduzioni burgundiane. Ho poi potuto confermare la dipendenza della traduzione latina dal testo del ms. Laur. plut. 74.18, la cui descrizione comprende l’analisi delle note di Burgundio e di “B”, collaboratore dello scriba Ioannikios. A seguire, una breve descrizione dei venticinque testimoni latini si accompagna all’analisi dei loro rapporti stemmatici e allo stemma codicum. L’elaborato culmina nell’edizione critica, che si presenta in sinossi con la trascrizione del ms. Laur. plut. 74.18 (in apparato le varianti relative all’edizione di Kühn). Ci sono inoltre due appendici: la prima include le differenze tra la traduzione e il testo del manoscritto greco modello e l’apparato latino esaustivo; la seconda si compone di un census dei testimoni e in un testo di lavoro della traduzione dell’Ad tirones in sinossi con la trascrizione del manoscritto greco modello, il ms. Laur. plut. 75.5. La dissertazione si conclude con le corrispondenze greco-latine e latino-greche di De causis pulsuum e De pulsibus ad tirones. [a cura dell'Autore]This dissertation consists of five chapters and two appendices and provides the critical edition of the Latin version of Galen’s treatise De causis pulsuum by Burgundio of Pisa (XII c.). After a general overview on the Greek text and on the impact of the galenic science of pulse in the West during the XII c., it follows the translator’s profile and an introduction to his version, limited just to the books III-IV (lemmatique commentary about the De pulsibus ad tirones). A reflection on the translator’s method and on the links of interdependence between his versions of De causis and Ad tirones leads to a dating proposal into the relative chronology of his work. Then, I was able to confirm the dependence of the Latin version from the Greek text of the ms. Laur. plut. 74.18, whose description is enriched with the analysis of the notes by Burgundio and “B”, colleague of the scribe Ioannikios. Thereafter, it comes a brief description of the twenty-five Latin witnesses and the analysis of the stemma of the manuscript tradition. The dissertation culminates in the critic edition, in which the Latin text is in synopsis with the transcription of the ms. Laur. plut. 74.18 (in apparatus the variants relative to Kühn’s edition). There are also two appendices: the first includes the differences between the translation and the Greek exemplar and the exhaustive apparatus of the Latin text; the second provides a census of the witnesses and a working text of the translation of the Ad tirones in synopsis with the transcription of the Greek exemplar, the ms. Laur. plut. 75.5. The dissertation ends with Greek-Latin and Latin-Greek correspondances of the De causis pulsuum and the De pulsibus ad tirones. [edited by Author]XXXII cicl
