Der Bauchspeicheldrüsenkrebs gehört zu den gefährlichsten Krebsarten. Die Überlebensrate liegt bei nur 5%. Nach Einschätzung der Weltgesundheitsorganisation (WHO) werden bis 2030 weltweit 21 Millionen Menschen an diesem Krebs erkranken. Eine Entdeckung am Institut Toxikologie und Genetik (ITG) macht jetzt erstmals Hoffnung auf eine wirksame Therapie. Dort hat man festgestellt, dass eine Segmentierung des bereits in den 90er Jahren am KIT entdeckten Proteins CD44v6 die Bildung der gefürchteten Metastasen hemmen kann. Radio KIT-Reportern Jennifer Warzecha hat mit Prof. Dr. Véronique Orian-Rousseau vom Institut für Toxikologie und Genetik am KIT gesprochen.

Étude de l’organisation et de la dynamique des complexes formés par MET et CD44v6 à la membrane plasmique
Le récepteur tyrosine kinase MET et son ligand, le facteur de croissance des hépatocytes (HGF) contrôlent des processus cellulaires vitaux tels que la survie, la prolifération, la différenciation et la motilité. La signalisation de MET induite par HGF est essentielle au développement embryonnaire et à la régénération des tissus. Un dérèglement de la voie HGF/MET peut entraîner une tumorigénèse et des métastases. MET possède un autre ligand appelé internaline B (InlB), que l'on retrouve sur la bactérie Listeria Monocytogenes. L'activation de MET médiée par HGF et InlB dépend de CD44v6, qui est un membre de la famille des protéines CD44. CD44v6 a deux fonctions : sa partie extracellulaire régule la phosphorylation de MET, alors que son domaine cytoplasmique est impliqué dans l'internalisation de MET et sa signalisation en aval. L'objectif de cette thèse a été de caractériser l'organisation et la dynamique de MET et CD44v6 lors de l'activation de MET à la membrane plasmique. Dans ce contexte, une interaction directe a été observée entre MET et CD44v6 lors de l'induction de HGF et InlB par mesures d’imagerie de fluorescence résolue en temps, combinées à des mesures de transfert d’énergie de Förster. Nous avons montré que HGF et InlB augmentent la mobilité de CD44v6 mais induisent également sa dimérisation. De la même façon que HGF,le traitement des cellules par la méthyl-β-cyclodextrine conduit à une augmentation de la mobilité deCD44v6. En outre, nos données indiquent que les domaines lipidiques ne sont vraisemblablement pas impliqués dans la phosphorylation de MET et celle des cibles en aval. Nous avons proposé un modèle selon lequel CD44v6 diffuserait hors d'un domaine confiné tel que les domaines lipidiques pour se dimériser et se lier au ligand de MET avant de présenter le ligand au récepteur MET dans un domaine liquide désordonné.The receptor tyrosine kinase MET and its ligand the hepatocyte growth factor (HGF) control vital cellular processes such as survival, proliferation, differentiation and motility. HGF-induced MET signaling is critical in embryonic development and tissue regeneration. A dysregulation of HGF/MET pathway can drive tumorigenesis and metastasis. MET has another ligand called internalin B (InlB), found on Listeria Monocytogenes bacteria. HGF and InlB-mediated MET activation is dependent on CD44v6, which is a member of the CD44 protein family. CD44v6 has two functions : its extracellular part regulates the phosphorylation of MET and its cytoplasmic domain is involved in MET internalization and downstream signaling. The aim of this thesis was to characterize the organization and dynamics of MET and CD44v6 during MET activation at the plasma membrane. In this context, a direct interaction was observed between MET and CD44v6 upon induction by HGF or InlB by Förster resonance energy transfer-fluorescence lifetime imaging microscopy. HGF and InlB increased the mobility of CD44v6 but also induced its dimerization. In the same way as HGF, the treatment of cells with methyl-β-cyclodextrin leads to an increase in the mobility of CD44v6. Furthermore, our data indicate that lipid domains are unlikely to be involved in the phosphorylation of MET and that of downstream targets. We proposed a model whereby CD44v6 diffuses out of a confined domain such as lipid domains to dimerize and bind to the MET ligand before presenting the ligand to the MET receptor in a disordered-liquid domain
