No full textLa trasferta nelle e dalle principali sedi universitarie (soprattutto Bologna e Parigi) fornisce sovente materia alla narrativa breve, a partire dal “Novellino”, e mette in luce un diffuso sentimento di più o meno latente ostilità nei confronti degli aspiranti o certificati intellettuali, gli “scolari”. Lo dimostrano in particolare due scenette famigliari tratteggiate da Sacchetti; mentre la novella XLV di Masuccio, che narra le alterne vicende di un “nobilissimo legista” castigliano in transito verso Bologna, offre collateralmente una conferma delle implicazioni di status connesse al soggiorno nelle città universitarie. La seconda parte del contributo è dedicata alla famosissima novella della vedova e dello “scolare” (“Decameron” VIII 7), anche per indagare incongruenze tematiche e stilistiche ed escursioni di genere.The journey to and from the main universities (especially Bologna and Paris) often provides material for short fiction, starting with the “Novellino”, and highlights a widespread feeling of latent hostility towards aspiring or certified intellectuals, the “scolari”. Session based access control in geographically replicated Internet services
No full textPerformance critical services over Internet often rely on geographically distributed architectures of replicated servers. Content Delivery Networks (CDN) are a typical example where service is based on a distributed architecture of replica servers to guarantee resource availability and proximity to final users. In such distributed systems, network links are not dedicated, and may be subject to external traffic. This brings up the need to develop access control policies that adapt to network load changing conditions. Further, Internet services are mainly session based, thus an access control support must take into account a proper differentiation of requests and perform session based decisions while considering the dynamic availability of resources due to external traffic. In this paper we introduce a distributed architecture with access control capabilities at session aware access points. We consider two types of services characterized by different patterns of resource consumption and priorities. We formulate a Markov Modulated Poisson Decision Process for access control that captures the heterogeneity of multimedia services and the variable availability of resources due to external traffic. The proposed model is optimized by means of stochastic analysis, showing the impact of external traffic on service quality. The structural properties of the optimal solutions are studied and considered as the basis for the formulation of heuristics. The performance of the proposed heuristics is studied by means of simulations, showing that in some typical scenario they perform close to the optimum. (c) 2006 Elsevier B.V. All rights reserved

Integration and disentanglement of single-cell and spatial transcriptomics in health and disease
Get PDFSingle-cell and spatial transcriptomics have transformed our understanding of cellular diversity in complex biological systems, enabling detailed exploration of tissue heterogeneity in health and disease. This research focuses on advancing analysis methods for single-cell and spatial transcriptomics, specifically applied to kidney biology and Autosomal Dominant Polycystic Kidney Disease (ADPKD), a genetic disorder marked by progressive cyst formation leading to kidney failure. We developed a comprehensive Mouse Kidney Atlas by integrating multiple single-cell RNA sequencing datasets, creating a hierarchical reference of kidney cell populations. Applying spatial transcriptomics to healthy and ADPKD mouse kidneys, we enhanced cell-type resolution, identifying increases in fibroblasts, injury-repair associated cells, and immune cells. Notably, we discovered failed-repair proximal tubule cells near cyst formations, exclusive to diseased kidneys, and characterized ligand-receptor interactions in cyst microenvironments. Addressing integration challenges, we introduced spVIPES, a computational method distinguishing shared from condition-specific signals in single-cell data, improving integration across conditions and species. Pilot studies optimizing single-nuclei RNA sequencing provided initial insights into early-stage ADPKD cellular and molecular changes. These methodological advancements significantly enhance our understanding of the cellular mechanisms underlying ADPKD.LUMC / Geneeskund
