The positive and negative selection of B cells into the pre-immune repertoire
The mechanisms of positive and negative selection of B cells and the ontogenetic switch between early and late tissue are not fully understood. The selection of mouse B1 B cells, immune components that secrete antibodies against self-antigens and common pathogens, and have a role in housekeeping and homeostasis, is known to be strongly influenced by self-antigen and BCR signalling in early ontogeny.
The objective of this thesis is to elucidate the role of GRB2, an adaptor protein, in the positive and negative selection of B cells. Furthermore, it examines the potential role of GRB2 in preserving the ontogenetic switch between early and late hematopoietic environments. Additionally, this work aims to explore the inherent differences from fetal liver (FL) and bone marrow (BM) to give rise to B1 B cells, employing single-cell transcriptomics for in-depth analysis.
In this project, addition to a B cell specific GRB2 knock-out mouse model (Grb2f/fMb1Cre+ (KO)), several transgenic mouse models were utilised to investigates GRB2's role in positive selection process of B1 B cells.
It was found that the number of B1b B cells, marginal zone B cells (MZ), and serum IgM levels were increased in the absence of GRB2. The development of B1b B cells was not restricted to early-stage tissue in the absence of GRB2, the effect of which was B cell intrinsic. Regardless of the presence of antigen, GRB2 deficiency generated more B1b B cells and MZ B cells, while intracellular membrane bound self-antigen presentation further increased the number of B1b B cells by ten-fold. Single-cell immune profiling revealed a putative B1/MZ precursor population (CD19hiB220lowIgMhiCD43hiCD21hiIgDlow) in the absence of GRB2 in adult BM, suggesting a shared developmental pathway. This study also suggested GRB2 was involved in a pathway that modulates IgM receptor levels in response to self-antigens, influencing the shift from positive to negative B cell selection, which might contribute to B1b B cell ontogeny.
In conclusion, our findings suggests that GRB2 is an intrinsic negative regulator of B1b B cell development and is required to restrict the generation of these cells in adult mice. The absence of GRB2 enhanced the positive selection by self-antigen while did not alter the requirement of appropriate self-antigen form.
The effect of heat waves on hospital admissions for renal disease in a temperate city of Australia
Published by Oxford University Press on behalf of the International Epidemiological Association © The Author 2008; all rights reserved.Background A rarely investigated consequence of heat exposure is renal dysfunction resulting from dehydration and hyperthermia. Our study aims to quantify the relationship between exposure to extreme high temperatures and renal morbidity in South Australia. Methods Poisson regression accounting for over dispersion, seasonality and long-term trend was used to estimate the effect of heat waves on hospital admissions for renal disease, acute renal failure and renal dialysis over a 12-year period. Selected comorbidities were investigated as possible contributing risk factors. Results Admissions for renal disease and acute renal failure were increased during heat waves compared with non-heat wave periods with an incidence rate ratio of 1.100 [95% confidence intervals (CI) 1.003–1.206] and 1.255 (95% CI 1.037–1.519), respectively. Hospitalizations for dialysis showed no corresponding increase. Comorbid diabetes did not increase the risk of renal admission, however ‘effects of heat and light’ and ‘exposure to excessive natural heat’ (collectively termed effects of heat) were identified as risk factors. Conclusion Our findings suggest that as heat waves become more frequent, the burden of renal morbidity may increase in susceptible individuals as an indirect consequence of global warming.Alana L Hansen, Peng Bi, Philip Ryan, Monika Nitschke, Dino Pisaniello and Graeme Tucke
MONOPOLY OF FORCE ON THE DEFENSIVE: ON CHANGING SOVEREIGNTY IN THE SPHERE OF INTERNAL SECURITY
Autor polazi od teze da je državni monopol sile izložen dekonstrukciji. Na djelu su fragmentacija sigurnosnog interesa, privatizacija javne sigurnosti i izvršni deficiti državnog monopola sile. Pokazuje se da je privatizacija javne sigurnosti najsuptilniji fenomen s golemim dometom. Naime, u okviru ekonomizacije životnih područja koju donosi globalizacija, dolazi do golemog rasta privatnih sigurnosnih službi. Prva posljedica toga je fragmentacija socijalnog prostora tako što se stvaraju privatno organizirane zaštitne zone, a druga je partnerstvo privatnih i policijskih interesa. Autor iz ovog izvodi da država više ne može jamčiti unutarnju sigurnost. Potiskivanju državnog monopola sile pridonosi segregacijska kultura koja je osobito vidljiva u državama Trećeg svijeta gdje često silu ne nadzire država nego različite komunitarne ili komercijalne organizacije. Sličan učinak imaju i nadnacionalni integracijski procesi. Autor zaključuje da je na djelu širenje sigurnosnih partnerstava kojima se država okružuje, što svemu daje jednu postmodernu notu: sustav nekad svemoćnog Levijatana danas je tek malo ispred prirodnog stanja.The author's starting assumption is that the state monopoly of force is being deconstructed through the fragmentation of security interest, the privatization of public security and the executive deficits of the state monopoly of force. It has turned out that the privatization of public security is an extremely subtle phenomenon with a huge scope. Namely, the economization of habitats resulting from globalization leads to the mushrooming of private security services. The first consequence of this is the fragmentation of the social sphere and the creation of the privately organized protective zones; the second is the partnership of the private and the polity interests. The author consequently concludes that the state cannot any longer guarantee internal security. The segregationist culture contributes to the diminishing of the state monopoly of force. This is especially noticeable in the Third World countries where force is often not controlled by the state but by various communitarian or commercial organizations. The supranational integrational processes have a similar effect. The author concludes that at work here is the expansion of security partnerships by which the state surrounds itself; this strikes a rather postmodern note: the system of a once all-powerful Leviathan today is just a notch removed from its natural state
