    Major histocompatibility complex class II transcriptional platform: assembly of nuclear factor Y and regulatory factor X (RFX) on DNA requires RFX5 dimers

    Major histocompatibility complex class II (MHC-II) genes are regulated in a B-cell-specific and gamma interferon-inducible manner. Conserved upstream sequences (CUS) in their compact promoters bind nuclear factor Y (NFY) and regulatory factor X (RFX) complexes. These DNA-bound proteins form a platform that attracts the class II transactivator, which initiates and elongates MHC-II transcription. In this report, we analyzed the complex assembly of these DNA-bound proteins. First, we found that NFY can interact with RFX in cells. In particular, NFYA and NFYC bound RFXANK/B in vitro. Next, RFX5 formed dimers in vivo and in vitro. Within a leucine-rich stretch N-terminal to the DNA-binding domain in RFX5, the leucine at position 66 was found to be critical for this self-association. Mutant RFX5 proteins that could not form dimers also did not support the formation of higher-order DNA-protein complexes on CUS in vitro or MHC-II transcription in vivo. We conclude that the MHC-II transcriptional platform begins to assemble off CUS and then binds DNA via multiple, spatially constrained interactions. These findings offer one explanation of why in the Bare Lymphocyte Syndrome, which is a congenital severe combined immunodeficiency, MHC-II promoters are bare when any subunit of RFX is mutated or missing

    MHC class II (MHCII) determinants play a crucial role in the immune response by presenting antigenic peptides to T cells. Their expression is controlled from compact promoters at the transcriptional level. Pre-assembled regulatory factor X (RFX) and nuclear factor Y (NFY) complexes form a platform on DNA. The class II transactivator (CIITA) can then be recruited through multiple protein±protein interactions. In this report, we de®ned domains of CIITA that are responsible for its interactions with these DNA-bound factors. Furthermore, using DNA-af®nity precipitation, we demonstrated that although CIITA binds at least ®ve activators, RFX5, RFXAP, RFXANK/B, NFYB and NFYC, its assembly on the promoter requires the addition of nuclear extracts. We conclude that not only does the platform bind DNA via multiple, spatially constrained nteractions, but that it can recruit only modi®ed and/or complexed CIITA to MHCII promoters

    Construction of sanitary and stormwater sewer system in the Brda area

    V diplomskem delu je predstavljena zasnova in izvedba ločenega kanalizacijskega sistema za naselje Brda, ki vključuje gradnjo komunalne in meteorne kanalizacije. Namen projekta je odprava pomanjkljivosti obstoječega neurejenega stanja, kjer komunalne odpadne vode še vedno odtekajo v greznice, meteorna voda pa se odvaja površinsko ali neposredno v teren. Nova komunalna kanalizacija bo priključena na obstoječi kanalizacijski sistem, povezan s Centralno čistilno napravo Koper, kar bo omogočilo ukinitev greznic in zagotovilo varno ter okolju prijazno odvajanje odpadnih voda. Meteorna kanalizacija bo prevzemala odvodnjavanje streh objektov in odvajala padavinsko vodo neposredno v vodotok Vrgalovec, s čimer se zmanjšuje možnost zastajanja vode in poškodb površin. Analiza je pokazala, da trenutna rešitev ne vključuje cestnih površin, zato se kot izboljšava predlaga razširitev sistema na te odseke, kar bi še dodatno zmanjšalo vplive večjih padavinskih dogodkov. Predlagani sistem bo pomembno prispeval k izboljšanju komunalne infrastrukture, kakovosti bivanja in prostorskemu razvoju območja Brda.The thesis presents the design and implementation of a separate sewerage system for the settlement of Brda, which includes the construction of sanitary and stormwater sewer networks. The aim of the project is to address the shortcomings of the existing unregulated system, where sanitary wastewater still flows into septic tanks and stormwater is discharged overland or directly into the ground. The new sanitary sewer will be connected to the existing sewerage network linked to the Central Wastewater Treatment Plant Koper, enabling the elimination of septic tanks and ensuring safe and environmentally friendly wastewater disposal. The stormwater sewer will collect rainwater from building roofs and discharge it directly into the Vrgalovec stream, thereby reducing the risk of water stagnation and surface damage. The analysis showed that the current solution does not include road surfacestherefore, as an improvement, the extension of the system to these sections is proposed, which would further reduce the impacts of major rainfall events. The proposed system will significantly contribute to the improvement of communal infrastructure, quality of life, and spatial development in the Brda area

    Na primeru izgradnje komunalne opremljenosti naselja SE-3 smo prikazali smiselno zaporedje faz projekta. Podlaga za kvalitetno presojo investicije so opisi komunalne infrastrukture občinskega podrobnega prostorskega načrta, odloka o občinskem podrobnem prostorskem načrtu in projekta za izvedbo. Nekaj več pozornosti smo namenili projektni dokumentaciji za izvedbo fekalne in meteorne kanalizacije, saj sta poleg ceste, to elementa, ki najbolj vplivata na stroške. Opisane so aktivnosti, ki so potrebne, da pripeljemo projekt od ideje do izgradnje ter prevzema zgrajene infrastrukture v upravljanje. V skladu s pogoji in potrebami Občine Muta je prikazana upravičenost investicije.With the example of the construction of public utilities in the complex SE-3, we have shown the logical sequence of phases of the project. The basis for the quality assessment of the investment are the description of municipal infrastructure detailed municipal spatial plan, the Ordinance on the detailed municipal spatial plan and project for implementation. Some more attention was paid to the project documentation for the implementation of faecal and storm sewer, because this are, in addition to the road, the elements that are affecting costs the most. It describes the activities required to bring a project from idea to construction and realisation. In accordance with the conditions and needs of the Municipality of Muta is shown the viability of investments

    Z diplomskim delom spodbujamo in ozaveščamo uporabo deževnice s pomočjo zbiralnika vode. Opisali smo, kakšno podnebje imamo v Sloveniji,kako to vpliva na dež ter kako deževnica pravzaprav nastane, kakšna je njena kakovost in uporaba. Poglobili smo se v preteklost in podajamo rezultate razvoja zbiralnikov skozi čas, od najenostavnejših naravnih zbiralnikov v vrtačah (kalov, ki so tudi podrobno obrazloženi) do sodobnih plastičnih zbiralnikov. Ker je zbiralnik sam po sebi le prazen prostor, nismo pozabili na dotok vode, njeno zajemanje in zaščito pred vstopom nepotrebnih delcev. Namen naloge je preveriti, ali je deževnico mogoče uporabiti tudi v sodobnem času kot kvaliteten vodni vir. Na primeru rešitve za individualno hišo spremljamo adaptacijo obstoječega zbiralnika deževnice v edini vir pitne vode v individualni stanovanjski hiši. Opisali bomo dela od začetka gradnje hiše, preko potrebnih priklopov zbiralnika, do danes, z obrazložitvijo, kaj je potrebno popraviti oziroma zamenjati, da bo zbiralnik služil oskrbi z neoporečno pitno vodo.Through dissertation, we encourage and raise awareness of the use of rainwater with the help of a rainwater collector. We have described what kind of climate we have in Slovenia and how it affects the rain, how rainwater occurs, its quality and its use. We looked in the past practices and development of rainwater collectors through time, from the simplest natural reservoirs in sinkholes (pits, which are also explained in detail) to modern plastic reservoirs. Since the rainwater collector is an empty shell in itself, we have not forgotten the flow of water, its capture and protection against the entry of unnecessary particles. The purpose of the dissertation is to check whether rainwater can also be used in modern times as a quality water source of clean water. Solution is offered through the example of an individual house, where we monitor the adaptation of the existing rainwater collector into the only source of drinking water in an individual residential house. We will describe the works from the beginning of the construction of the house, through the necessary attachment of the collector, to this day, when we explain what needs to be corrected or to replaced so that the tank will serve as a supply of clean drinking water

    To diplomsko delo obravnava magistralne cevovode, ki so opisani skozi zgodovino, njihove splošne karakteristike, značilnosti in primere na obalno kraškem območju in delno v hrvaški Istri. Projektiranje omenjenih cevovodov je redek izziv projektanta, zato so pa tudi možne napake, ki lahko nastanejo kljub poznavanju vodovodnih naprav in hidravlike. Diplomska naloga vsebuje nekaj zanimivih podatkov, ki so se nabrali z dolgoletnim delom avtorja na Rižanskem vodovodu Koper in so tu predstavljeni. Razumevanje dobrih in slabih lastnosti delovanja obstoječih naprav pripomore k dopolnjevanju znanja na tem področju gradbeništva.This bachelor\u27s dissertation deals with the history of magistral pipelines , their general characteristics and properties as well as with the description of some examples built in the carsic region and in the croatian Istria. The design of magistral pipelines is a rare challenge in everyday\u27s life and therefore mistakes in their design can occur, no matter how experienced the projecting engineer may be. In the bachelor\u27s dissertation some interesting information collected by the author through his years of work at "Rižanski vodovod Koper" is reported. The author believes that a thorough understanding of good and bad aspects in the functioning of existing devices can help to improve the knowledge in this field of civil engineering

    Integrated Design in Case of Digital Fabricated Buildings

    AbstractThe importance of the detailed BIM for buildings like the free-form building in our case cannot be emphasized enough in case of automated and especially additive manufacturing. Integral design of all building accessories like internal heating, ventilation, and air conditioning (HVAC), water supply, and drainage network installations is essential when applying additive manufacturing. In such a building it is impossible to build the ventilation network with traditional vertical and horizontal canals. Important benefit of digitally fabricated building can be internal topological optimization of building elements like walls and slabs for thermal insulation and structural capabilities. Application of large-scale additive manufacturing systems in the AECO industry is in early research phase. Future research directions are further parameterization of the interoperability demand function, BIM maturity, automation of workflow models, and new approaches for engineering of embedded building elements

    International audienceThe regulatory factor X (RFX) complex, which contains RFXANK(B), RFXAP, and RFX5, binds to X and S boxes in major histocompatibility complex class II (MHC II) promoters. In the bare lymphocyte syndrome (BLS), which is a human severe combined immunodeficiency, MHC II promoters are neither occupied nor transcribed. Thus, the absence of any one subunit prevents the formation of the RFX complex. Nevertheless, except for a weak binding between RFX5 and RFXAP, no other interactions between RFX proteins have been described. In this study, we demonstrate that RFXANK(B) binds to RFXAP to form a scaffold for the assembly of the RFX complex, which then binds to DNA. Moreover, mutant RFXANK(B) and RFXAP proteins from complementation groups B and D of BLS, respectively, cannot support this interaction. Our data elucidate an intriguing medical situation, where a genetic disease targets two different surfaces that are required for the nucleation of a multisubunit DNA-protein complex
