Analyzing protein intermediate interactions in living E. coli cells using site-specific photo-crosslinking combined with chemical crosslinking
Information on protein-protein interactions is crucial in understanding protein-mediated cellular processes; however, analyzing transient and unstable interactions in living cells is challenging. Here, we present a protocol capturing the interaction between an assembly intermediate form of a bacterial outer membrane protein and β-barrel assembly machinery complex components. We describe steps for expression of a protein target, chemical crosslinking combined with in vivo photo-crosslinking and crosslinking detection procedures including immunoblotting. This protocol can be adapted to analyze interprotein interactions in other processes. For complete details on the use and execution of this protocol, please refer to Miyazaki et al. (2021).1
Exploring Emptiness: An Investigation of MA and MU in My Sonic Composition Practice
The commentary investigates Japanese aesthetics of space, silence and emptiness - ma and mu - that informed my compositional practice during the research period 2012 - 2015. The portfolio comprises text compositions and sound installations in which forms of micro events and sustained events are employed. Throughout, the emphasis is on my personal engagement with, and manifestation of emptiness that concerns a particular model of listening and perception.
Chapter 1 discusses six primary research areas: ma and mu, material, text, form, listening and perception. Firstly, I introduce ma and mu by examining noh culture and Zeami's teaching of senu hima (where there is no-action) in the context of my personal approaches to music. The following subjects are then used to contextualise my PhD practice by means of examples from various composers and visual artists. Here, these particular and enigmatic concepts are explored through Japanese art as well as Western contemporary works by Alvin Lucier, Eliane Radigue and those of the Wandelweiser collective.
Part 2 provides contextual commentaries on selected compositions from the portfolio that mostly articulate my aesthetics in relation to the topics covered in Chapter 1. koso koso addresses my methodologies to investigate the essence of senu hima, followed by treow that discusses my approach to materials and the importance of space. I move on to grade two and grade two extended in order to examine text scores, and then, look into Espèces d'espaces 03 and 04 as examples of musical forms that I employ.
Finally, listening and perception are investigated through the compositions gnome and con.de.structuring. Throughout, I describe how my works explore emptiness as a result of my particular emphasis on listening over composing

INTRAVENOUS "MINOCYCLINE" IN THE TREATMENT OF CHRONIC URINARY TRACT INFECTION AND GENITAL INFECTION
Twenty patients of urinary tract infection (16 cases of chronic cystitis, 3 of chronic pyelonephritis, and 1 chronic prostatitis) were treated by intravenous drip of Minocycline hydrochloride 100 mg twice daily for four or five days. The efficacy of the treatment was judged on the basis of clinical examination, bacteriological test, and improvement of subjective symptoms. Eighty percent of the cases were favorable: 8 excellent, 8 good, 4 null. No manifestation of side effects was encountered except the cases in which S-GOT or S•GPT was slightly elevated

Effect of Pellet Feed Prepared from Shochu Distiller's By-product on Growth and Meat Quality of Beef Cattle (Preliminary Experiment)
本プロジェクトでは、焼酎粕(ソバ焼酎粕濃縮液および同脱水ケーキの等重量混合物)を主原料とし、大麦ヌカおよびビートパルプの混合物(60:40)を副原料として2軸型エクストルーダーにより製造した焼酎粕ペレット飼料(主副両原料割合,60:40)を用いて、肉用牛による飼養試験を行うことを目的とした。また、本プロジェクトは、これまでその多くを海洋投棄してきた焼酎粕を飼料の製造に再利用することにより地球環境の浄化に資することも背景にある大きな目的の一つである。
飼養試験では、焼酎粕ペレット飼料が肉用牛(黒毛和種)の成長(増体量)、飼料要求率および肉質に及ぼす影響を検討するため、対照区の飼料として宮崎県内で広く給与されている「宮崎しもふり特号」を給与し、また、試験飼料として「宮崎しもふり特号」の15% (乾物当たり)を、可消化養分総量および粗タンパク質が同じになるように勘案しながら、焼酎粕ペレット飼料で置換した飼料を試験飼料として給与して、10~13ケ月齢(平均月齢11.6カ月)から478日間(約16カ月間)にわたって、ほぼ1月毎に試験牛の体重および飼料摂取量を測定した。試験期間中、どの動物にも異常は認められなかった。その結果、301日目(平均月齢22カ月)までは、試験飼料の肉用牛(黒毛和種)成長促進効果(飼料効率)は、「宮崎しもふり特号」と同程度と考えられたが、23カ月齢以降は、試験飼料の飼料効率は対照区よりも30%以上も高くなることが示された。
格付けの結果は、同じ血統の牛(試験区901および対照区902)では、両者とも歩留等級A、肉質等級4とランクされた。血統が異なる牛では、対照区が試験区よりもややよい結果になったが、この点はさらなる検討が必要である。肉質は、両者に大きな差は見られなかったが、試験区は、ドリップが少なく、肉食のL値は高かった。ロース芯の総脂質とビタミンEの含量は試験区がやや高く、筋間総脂質はその逆だった。The aims of this project were to establish a technique for preparing pellet feeds for cattle using shochu distiller's by-products and to evaluate the product as a feedstuff for beef cattle. Pellets were made by extrusion using a mixture of buckwheat, shochu distiller's condensed solubles and its cake (both wet) as the main materials, together with smaller amounts of barley bran and beet pulp.
Three pellet feeds among the thirteen prepared showed good palatability, had high protein contents and normal levels of minerals. Very little amounts of vitamins A and D were detected, while vitamins E and B6 were present at almost the same concentration as in corn and grain sorghum, respectively.
Six Japanese black cattle, three animals each in control and experimental groups, received for 16 months a commercial formula feed (Miyazaki Shimofuri Tokugo ) as a control diet and a formula feed replacing 15 % of the dry matter of the 'Miyazaki Shimofuri Tokugo' with the pellet feed as an experimental diet. The experimental diet contained the same levels of TDN and CP as those of the control diet. No differences in body weight gains or feed conversion were detected for the first 10 months; thereafter, up to more than 27 months after birth, animals supplied with experimental diets showed higher daily gains and lower feed conversion (higher feed efficiency) than those observed in control animals. However, there have been almost no differences in meat quality between the control and the experimental cattle as a whole, although α-tocopherol content in rib eye of the experimental cattle was higher than that of the control. CLINICAL EXPERIENCE WITH FOSFOMYCIN IN TREATMENT OF COMPLICATED URINARY TRACT INFECTIONS
Twenty-one patients with complicated urinary tract infections were administered 4 g of Fosfomycin sodium by intravenous drip infusion twice a day for 5 days. The efficacy of Fosfomycin was excellent in 8, moderate in 8 and poor in 5 cases. The overall efficacy rates for 21 cases were 76.2% (16 out of 21 cases). Pathological Changes in Gastric Walls of Wild Boars Infected with Gnathostoma doloresi (1) : Macroscopical findings
宮崎県下に生息するイノシシには極めて高率にドロレス顎口虫が寄生するが,寄生胃15例の肉眼的病変を検索して以下の知見を得た.
1. 顎口虫の主たる寄生部位は胃底である.その粘膜面に寄生孔を形成し,虫体の前半(頭側)を孔内にそう入,後半(尾側)を胃腔へ突出させて寄生する.寄生孔の数は最多例で30コ以上,最少例で3コ,平均11.5コ. 寄生孔に寄生する虫体(すなわち最終寄生態勢の虫体)の数は最多例で52匹,最少例で2匹,平均17匹(寄生孔から脱落して胃内容物に混在する虫体を含む).なお虫体の大きさ(体長)は,大は30mm,小は7mm とすこぶる大小不揃を呈していた.
2. 寄生孔を中心として胃壁(主として粘膜下織)は増厚を呈するが,その程度はとくに著しいものではなく,最厚18mm (うち粘膜下織の厚さは12~13mm)を越えない.また増厚域の範囲も比較的狭い.増厚部の粘膜下織は硬い結合織より成るが,水腫性疎開を呈するものもあった.なお増厚域に虫体(幼若虫・成虫)の穿入・埋在を認めたものがある.
3. 増厚域の粘膜下織(一部は筋層・漿膜)には,各種発育段階の虫体が穿行・遊走したあとの病変(虫体穿行跡の病巣)が極めて高率に検出せられた. この種の病巣を微小巣と虫道巣に区分するが,前者は肉眼で辛うじて認められる程度のごく小さく(針尖大)かつ限界明瞭な結節性の病巣,後者はいくぶん大きく粟粒大・米粒大で暗褐紅色を呈し,境界のやや不明瞭な不斉形の病巣である.なお両者の中間移行型も認められた. このうち微小巣は,寄生孔からかなり離れて胃壁増厚を呈しない部位からも検出せられた.
4. 噴門部に虫体の侵入・寄生を認めたものが供試胃の約半数あった.また噴門部の胃壁(主として粘膜下織)に微小巣・虫道巣を所見したものもはば同数. ただし病変の程度は胃底に比べるとはるかに軽少である.幽門部においても, 6例に虫体の穿入・寄生と胃壁(主として筋層)に虫体穿行跡病巣を所見した.Wild boars (Sus scrofa leucomystax) inhabiting Miyazaki Prefecture are infected with Gnathostoma doloresi at a very high frequency. Observation was made on macroscopical changes in the stomachs infected with this parasite of 15 wild boars collected in the shooting seasons in 1975-1977. The findings obtained are as follows.
1. The fundus was a main locality of infection with G. doloresi. The parasite formed a parasitic hole on its mucosal surface. The cranial half of its body was inserted into the hole, and the caudal half projected into the gastric cavity. The number of parasitic holes per stomach ranged from 3 to more than 30, averaging 11. 5. The number of parasitic worms per stomach ranged from 2 to 52, averaging 17. The body length of the parasite was quite variable, ranging from 7 to 30mm.
2. The gastric wall (mainly the tunica submucosa) became thick around the parasitic hole. Its thickness, however, did not exceed 18mm (the tunica submucosa was 12-13mm in thickness). And the increase in thickness was not seen in so wide an extent. The tunica submucosa was generally composed of hard connective tissue, and some part of it presented edematous dissociation. Immature and young adult worms were found buried in the thickened tunica submucosa in some cases.
3. Lesions caused by the penetration and migration of immature and young adult worms were found at a high frequency in the gastric wall around parasitic holes (mostly in the tunica submucosa and partly in the tunica muscularis and serosa). They were divided into two types of foci. One type was called a micro-focus, which was a well-defined nodular focus so small that it was hardly observed macroscopically. The other type was called a migratory-route focus, which was a dark brownish red irregular shaped focus 3-5mm in diameter, and its demarcation was a little indistinct. Micro-foci were also noticed in some portions of the gastric wall which were considerably far from any parasitic hole and showed no thickening.
4. In 7 cases worms entered the cardiac region by perforation and stayed there, inducing migratory-route and micro-foci in the gastric wall. In 6 cases worms were harbored in the pyloric region, and both types of foci found in its gastric wall.departmental bulletin pape
