    Molecular identification of autochthonous bacterial fibrinolytic protein producers from fermentative food preparations

    154-162The current study investigates screening, isolation and characterization of fibrinolytic enzyme producing autochthonous bacteria from fermented foods. About 21 different fermented food samples were screened for fibrinolytic protease producing bacteria. Procured and prepared samples were homogenized, serially diluted and plated onto nutrient agar and soybean casein digest agar medium. A total number of 89 isolates were isolated and screened for caseinolytic and fibrinolytic activities. Out of those 89 isolates, nineteen were found to be potent protease producers. Amongst those isolates MS2, MS3, MS4 and MS9 had notable fibrinolytic activities as well. These isolates were subjected for biochemical and molecular characterization to determine their taxonomy and phylogeny. The strain with highest fibrinolytic potency was found to be Bacillus subtilis VITMS2 isolated from fermented milk of Vigna unguiculata. The clot lysing ability of the selected potent strain B. subtilis was found to be 79.98%. The total and specific activity of the same was found to be 788.82 U/mL and 41.73 U/mg, respectively. Other strains with comparatively low fibrinolytic activity were Pseudomonas aeruginosa VITMS3, Bacillus sp VITMS4 and Alcaligenes sp. VITMS9

    Novel Fibrinolytic Protease Producing Streptomyces radiopugnans VITSD8 from Marine Sponges

    Fibrinolytic enzymes have received more attention due to their medicinal potential for thrombolytic diseases. The aim of this study is to characterize the in vitro fibrinolytic nature of purified protease producing Streptomyces radiopugnans VITSD8 from marine brown tube sponges Agelas conifera. Three varieties of sponge were collected from the Rameshwaram Sea coast, Tamil Nadu, India. The fibrinolytic activity of Streptomyces sp. was screened and determined by casein plasminogen plate and fibrin plate methods respectively. The crude caseinolytic protease was purified using ammonium sulfate fractionation, affinity and ion-exchange chromatography. Based on the morphological, biochemical, and molecular characterization, the isolate VITSD8 was confirmed as Streptomyces radiopugnans. Maltose and peptone were found to be the best carbon and nitrogen sources for the production of fibrinolytic protease. The carbon and nitrogen source peptone showed (781 U/mL) enzyme activity. The optimum pH and temperature for fibrinolytic protease production was found to be 7.0 and 33 °C respectively. The purified enzyme showed a maximum specific activity of 3891 U. The blood clot lysis activity was compared with the standard, and it was concluded that a minimum of 0.18 U (10 µL) of purified protease was required to dissolve the blood clot. This is the first report which exploits the fibrinolytic protease activity of Streptomyces radiopugnans VITSD8 extracted from a marine sponge. Hence the investigation suggests a potential benefit of purified fibrinolytic protease which will serve as an excellent clot buster alternative

    Southern Greenland glaciation and Western Boundary Undercurrent evolution recorded on Eirik Drift during the late Pliocene intensification of Northern Hemisphere glaciation

The final version is available from Elsevier via the DOI in this record.We present new sedimentological and environmental magnetic records spanning ~3.2–2.2 Ma, during the intensification of Northern Hemisphere glaciation, from North Atlantic Integrated Ocean Drilling Program Site U1307 on Eirik Drift. Our new datasets and their high-fidelity age control demonstrate that while inland glaciers – and potentially also at times restricted marine-terminating ice-caps – have likely existed on southern Greenland since at least ~3.2 Ma, persistent and extensive marine-terminating glacial margins were only established in this region at 2.72 Ma, ~300 kyr later than in northeastern and eastern Greenland. Despite a dramatic increase in Greenland-sourced ice-rafted debris deposition on Eirik Drift at this time, contemporaneous changes in the bulk magnetic properties of Site U1307 sediments, and a reduction in sediment accumulation rates, suggest a decrease in the delivery of Greenland-sourced glaciofluvial silt, which we attribute to a shift in depositional regime from bottom-current-dominated to glacial-IRD- dominated between ~2.9–2.7 Ma in response to a change in the depth of the flow path of the Western Boundary Undercurrent relative to our study site

    Psychological and self-management support for people with vasculitis or connective tissue diseases: UK health professionals' perspectives

    © The Author(s) 2020. Objectives. CTD and systemic vasculitis impact on health-related quality of life. Treatment can be complex, involving multiple medical specialities. The aim of this study was to investigate psychological and self-management support for patients in secondary care. Methods. An online survey of health professionals in the UK, including 45 multiple-choice and freetext questions, was analysed descriptively. Free-text survey responses were analysed thematically to identify health professionals' perceptions of best practice and unmet needs. Results. The online survey included 120 health professionals (34% specialist nurses, 51% doctors and 12% allied health professionals), predominantly working in rheumatology (52.9%) and nephrology (21.5%) departments. Access to self-management programmes or clinics for people with CTD or vasculitis was available in 23% of rheumatology and 8% of nephrology departments. In response to 'How well is your team providing self-management support to people with CTD or vasculitis?', 38% of respondents reported 'not very well' or 'not well at all'. Direct access to psychological support was available in 76.9% of nephrology and 32.8% of rheumatology departments. More than 80% of respondents would like additional training. Key themes from the qualitative data (free-text survey responses) included the importance of: Dedicated psychological support and self-management programmes for people with CTD and vasculitis, a whole-team approach (specialist teams empowering people to manage their own care), staff training (e.g. brief psychological interventions) and signposting to resources, including patient charities. Conclusion. People with CTD and vasculitis have complex needs, and improvements in selfmanagement and psychological support are required in UK rheumatology and nephrology departments

    May Measurement Month 2022: Results from the global blood pressure screening campaign

    Introduction Elevated blood pressure (BP) is the major contributor to mortality and disease burden worldwide. May Measurement Month (MMM) is a global BP screening campaign, which aims to raise awareness of BP measurement and provide evidence to inform and influence related health policy. Methods This cross-sectional survey included individuals aged≥18 years recruited through opportunistic sampling at sites in 60 countries during MMM 2022. Each participant had three sitting BP measurements and a questionnaire was completed including demographics, comorbidities and lifestyle factors. Hypertension was defined as a systolic BP≥140 mm Hg and/or a diastolic BP≥90 mm Hg (average of the second and third readings) or taking antihypertensive medication. Multiple imputation was used to estimate BP readings where any participant's BP readings were missing. Linear mixed effects models were used to identify associations between participant characteristics and systolic or diastolic BP. Results Of the 715 518 participants surveyed (excluding 50 200 self-measured home BP screenees recruited via the ZOE Health Study app), 257 421 (36.0%) were identified as hypertensive, of whom 57.6% were aware and 49.3% were on antihypertensive medication. Of all participants with hypertension, 26.1% were controlled to <140/90 mm Hg and 12.0% to <130/80 mm Hg. Of those taking antihypertensive medication, 52.7% were taking only one drug class, 52.9% were controlled to <140/90 mm Hg and 24.4% to 130/80 mm Hg. In total, 190 314 (26.6% of total surveyed, 73.9% of hypertensives) participants screened were found to have untreated or inadequately treated hypertension. Only 27.6% of treated hypertensive participants were taking a statin. Substantial coexistence of diabetes, overweight and hypertension was apparent among participants. Conclusions MMM confirms a high global burden of hypertension with low rates of awareness, treatment and control. In the absence of systematic BP screening in many countries, the results from MMM underscore the continued need for BP screening to detect and thereby control raised BP. © Author(s) (or their employer(s)) 2024
