Renaissance of cancer therapeutic antibodies
In the past five years therapeutic monoclonal antibodies have established themselves as perhaps the most important and rapidly expanding class of therapeutic drugs. More than 25% of pharmacological agents that are currently under development are based on antibodies and the total income generated from them in 2002 exceeded 10–20 billion by 2010. Many experts feel that antibodies directed at cancer targets are likely to dominate the market for the foreseeable future. In this review, we will discuss some of the factors that, after more than 25 years of development, have led to this transformation in the antibody field.
Current technology makes the generation of fully-human antibodies a routine procedure. However, selection of appropriate disease targets still represents a challenge. Here we discuss factors that make an effective antibody target and provide an overview of the major issues that are likely to shape this exciting field over the next decade

Sustainable Chemical Processes and Products. New Design Methodology and Design Tools
The current chemical industry is not sustainable, which leads to the fact that innovation of chemical processes and products is too often hazardous for society in general and the environment in particular. It really is a challenge to implement sustainability considerations in the design activities of chemical engineers. Therefore, the main question of this thesis is: how can a trained chemical engineer develop a conceptual design of a chemical process or a chemical product in such a way that the final result clearly contributes to sustainable development? This question is answered after a profound discussion about the current chemical engineering practice and its relation to the sustainability debate. This dissertation claims that sustainable development of chemical engineering practices requires a general design methodology accompanied by a set of design tools. Such a combination of methodology and tools does not exist in the chemical engineering field. The author developed a new design methodology and seven new design tools that enable the incorporation of sustainability issues into the design practice of the chemical engineering field. The application and validity of the methodology and its tools are shown in seven, mainly industrial, case studies.Applied Science
Multi-proxy reconstruction of surface water pCO2 in the northern Arabian Sea since 29ka
We report here the results of a multi-proxy study to reconstruct surface water pCO2 concentrations in the northern Arabian Sea. Our results show that [delta]11B and Mg/Ca measurements of the planktonic foraminifer Globigerinoides sacculifer yield consistent pCO2 values with those reconstructed from the [delta]13C of alkenones when used in conjunction with foraminifera [delta]13C and Cd/Ca values. They reveal that this area of the oceans has been a constant source of CO2 to the atmosphere during the interval 5-29 ka, and that the intensity of this source was greatest between 11 and 17 ka, when atmospheric CO2 levels were rising rapidly. We interpret our data as reflecting variation in the strength of the Asian Summer Monsoon (ASM), thus indicating that the strength of the ASM varied in phase with summer insolation over the Tibetan plateau between 5 and 29 ka. In contrast to a previous study (Clemens and Prell, 2003), we observe no significant lag between the rise in insolation and the response of the ASM. Rather, our data support a recent study by Rohling et al. (2009) that northern hemisphere climatic forcing factors play a greater role in controlling the intensity of the ASM during times of intense monsoon activity, and that the southern hemisphere forcing is more important during times of weak monsoons
Mechanisms of G-CSF- or GM-CSF-stimulated tumor cell killing by Fc receptor-directed bispecific antibodies
Studies with gene-modified mice have recently reinforced the importance of Fc receptor-mediated effector mechanisms for the therapeutic efficacy of rituxan and herceptin — two clinically approved antibodies for the treatment of tumor patients. We investigated Fc receptor-dependent tumor cell killing by mononuclear and granulocytic effector cells — comparing human IgG1 antibodies against CD20 or HER-2/neu with their respective Fc?RI (CD64)-, Fc?RIII (CD16)-, or Fc?RI (CD89)-directed bispecific derivatives. With blood from healthy donors as effector source, human IgG1 and Fc?RIII (CD16)-directed bispecific antibodies proved most effective in recruiting mononuclear effector cells, whereas tumor cell killing by granulocytes was most potently triggered by Fc?RI-directed bispecific constructs. Granulocyte-mediated tumor cell lysis was significantly enhanced when blood from G-CSF- or GM-CSF-treated patients was investigated. Interestingly, however, both myeloid growth factors improved effector cell recruitment by different mechanisms, which were furthermore dependent on the tumor target antigen, and on the selected cytotoxic Fc receptor
