Neonatal inhibition of DNA methylation disrupts testosterone-dependent masculinization of neurochemical phenotype
Many neural sex differences are differences in the number of neurons of a particular phenotype. For example, male rodents have more calbindin-expressing neurons in the medial preoptic area (mPOA) and bed nucleus of the stria terminalis (BNST), and females have more neurons expressing estrogen receptor alpha (ERα) and kisspeptin in the ventromedial nucleus of the hypothalamus (VMH) and the anteroventral periventricular nucleus (AVPV), respectively. These sex differences depend on neonatal exposure to testosterone, but the underlying molecular mechanisms are unknown. DNA methylation is important for cell phenotype differentiation throughout the developing organism. We hypothesized that testosterone causes sex differences in neurochemical phenotype via changes in DNA methylation, and tested this by inhibiting DNA methylation neonatally in male and female mice, and in females given a masculinizing dose of testosterone. Neonatal testosterone treatment masculinized calbindin, ERα and kisspeptin cell number of females at weaning. Inhibiting DNA methylation with zebularine increased calbindin cell number only in control females, thus eliminating sex differences in calbindin in the mPOA and BNST. Zebularine also reduced the sex difference in ERα cell number in the VMH, in this case by increasing ERα neuron number in males and testosterone-treated females. In contrast, the neonatal inhibition of DNA methylation had no effect on kisspeptin cell number. We conclude that testosterone normally increases the number of calbindin cells and reduces ERα cells in males through orchestrated changes in DNA methylation, contributing to, or causing, the sex differences in both cell types.Fil: Cisternas, Carla Daniela. p19INK4d is involved in the cellular senescence mechanism contributing to heterochromatin formation
Background: During evolution, organisms with renewable tissues have developed mechanisms to prevent tumorigenesis, including cellular senescence and apoptosis. Cellular senescence is characterized by a permanent cell cycle arrest triggered by both endogenous stress and exogenous stress. The p19INK4d, a member of the family of cyclin-dependent kinase inhibitors (INK4), plays an important role on cell cycle regulation and in the cellular DNA damage response. We hypothesize that p19INK4d is a potential factor involved in the onset and/or maintenance of the senescent state. Methods: Senescence was confirmed by measuring the cell cycle arrest and the senescence-associated β-galactosidase activity. Changes in p19INK4d expression and localization during senescence were determined by Western blot and immunofluorescence assays. Chromatin condensation was measured by microccocal nuclease digestion and histone salt extraction. Results: The data presented here show for the first time that p19INK4d expression is up-regulated by different types of senescence. Changes in senescence-associated hallmarks were driven by modulation of p19 expression indicating a direct link between p19INK4d induction and the establishment of cellular senescence. Following a senescence stimulus, p19INK4d translocates to the nucleus and tightly associates with chromatin. Moreover, reduced levels of p19INK4d impair senescence-related global genomic heterochromatinization. Analysis of p19INK4d mRNA and protein levels in tissues from differently aged mice revealed an up-regulation of p19INK4d that correlates with age. Conclusion: We propose that p19INK4d participates in the cellular mechanisms that trigger senescence by contributing to chromatin compaction. General significance: This study provides novel insights into the dynamics process of cellular senescence, a central tumor suppressive mechanism.Fil: Sonzogni, Silvina Veronica. Morphological Analysis of the Flippers in the Franciscana Dolphin, Pontoporia blainvillei, Applying X-Ray Technique
Pectoral flippers of cetaceans function to provide stability and maneuverability during locomotion. Directional asymmetry (DA) is a common feature among odontocete cetaceans, as well as sexual dimorphism (SD). For the first time DA, allometry, physical maturity, and SD of the flipper skeleton—by X-ray technique—of Pontoporia blainvillei were analyzed. The number of carpals, metacarpals, phalanges, and morphometric characters from the humerus, radius, ulna, and digit two were studied in franciscana dolphins from Buenos Aires, Argentina. The number of visible epiphyses and their degree of fusion at the proximal and distal ends of the humerus, radius, and ulna were also analyzed. The flipper skeleton was symmetrical, showing a negative allometric trend, with similar growth patterns in both sexes with the exception of the width of the radius (P ≤ 0.01). SD was found on the number of phalanges of digit two (P ≤ 0.01), ulna and digit two lengths. Females showed a higher relative ulna length and shorter relative digit two length, and the opposite occurred in males (P ≤ 0.01). Epiphyseal fusion pattern proved to be a tool to determine dolphin's age; franciscana dolphins with a mature flipper were, at least, four years old. This study indicates that the flippers of franciscana dolphins are symmetrical; both sexes show a negative allometric trend; SD is observed in radius, ulna, and digit two; and flipper skeleton allows determine the age class of the dolphins. A Participatory Photo-Mapping (PPM) framework to observe and reflect on the transformation of public space: the case of the Paseo España Environmental Corridor in Bucaramanga, Colombia
Public participation in urban planning processes is key to ensuring projects can be successful, can address community needs, and can be sustainable into the future. The current sociodemographic and political circumstances in Colombia, namely the opportunities that the peace agreement between the government and guerrilla groups, and the advances of regulations that help ensure public participation in political processes across the country, have contributed to the increasing need of creating community engagement processes specifically in urban centers that support urban planning decision-making while supporting community development and relationship building. In 2021, Bucaramanga, a capital city of 500,000 inhabitants, developed a Walkable City Plan and an accompanying Revitalization of Public Spaces Plan. These aim to move forward a vision of creating lively public spaces that enable connectivity, sustainable mobility, and ultimately improve citizens’ quality of life. As part of these Plans, Bucaramanga aimed to complete 400 projects by December 2022 for the city’s 400th anniversary. The projects were chosen by the architecture firm TABUU, with their technical and social teams working together on prioritizing the most impactful possible interventions. While there are certain requirements for social engagement these interventions must comply with, there is room to strengthen these strategies by creating more timely, open and transparent processes, by ensuring project assessment and oversight during and after the infrastructural intervention, and by leveraging existing digital tools to democratize information and data. Thus, this thesis reviews the academic literature, official documents, and relevant precedents that can help guide better practices in community engagement processes in Bucaramanga, and explores the opportunity of utilizing a participatory photo-mapping framework to enable spaces to collaborate, exchange knowledge, and develop relevant skills in community planning to continue increasing participation moving forward.M.C.P
