    Performance of gravel aggregates in superpave mixes with 100/95 angularity

    The current aggregate requirement for aggregate angularity that the NYSDOT requires for their Superpave Hot Mix Asphalt (HMA) is 100/98. With this requirement, many of the gravel quarries in New York State cannot meet that specification. As a result, quarries that are further away from the job site have to transport the aggregates which in turn increases the cost of construction. This paper investigates a multitude of test methods to determine whether the NYSDOT can further lower their Superpave aggregate angularity requirement.     Epigenetic regulation and transcription factor programming enhances neurogenesis in neural stem cells

    In this thesis, we questioned how neuronal and glial phenotypes become specialized. Epigenetic chromatin modifiers and transcription factors were investigated on their roles in programming and maintaining neural lineage restriction. A relatively homogeneous population of cells was generated by deriving immortalizing neural clones from embryonic rat forebrains. Three phenotypes; neuronal, glial and multipotential (GE6, GE2, CTX8), provided contrasting lineages to probe the factors responsible for shaping cell fate. One particular clone, GE6, differentiated into a functional inhibitory like interneuron. Gene expression analysis showed several genes such as Ascl1, Dlx1, Dlx5, may be responsible for the interneuronal phenotype. Epigenetic regulation through histone modifications is believed to be an essential component within the developing nervous system, ultimately affecting cell fate. Testing chromatin signatures on specific neural genes with permissive and repressive histone “marks” shows that chromatin state in undifferentiated precursors correlates with current and predicts downstream gene expression. These results suggest that cell fate may already be predetermined. Furthermore, ChIP sequencing reveals global differences between the representative clones. Extrinsic growth factors, such as BMP2 promotes the neuronal and glial phenotypes in the multipotential cell CTX8. BMP2 asserts its phenotypic response in part by regulating global acetylation enrichment in specific neural gene networks, providing a mechanism to promote and maintain cell fate. Directly altering chromatin marks using a histone deacetylase inhibitor, valproic acid (VPA), globally acetylates the chromatin of CTX8 cells and enhances neurogenesis. VPA treatment was found to also maintain or increase acetylation in specific neuronal genes, such as Ascl1. In addition, several microRNAs thought to play a role in neurogenesis were also epigenetically regulated after VPA treatment. Finally, through the combination of gene expression and epigenetic analyses, direct programming through exogenous expression of Ascl1, Dlx1 and Dlx5 enhanced neurogenesis in CTX8. Gene expression and epigenetic signature mapping provides us with a deeper understanding of how lineage restriction occurs. Learning the programming rules will assist in directing homogeneous populations of neuronal cells to further probe the mechanisms of neurodegenerative diseases.Ph.     Heritability and Linkage Analysis of Appendicitis Utilizing Age at Onset

    Appendicitis usually afflicts the young, but there is a large tail in the distribution of onset age. The genetics of this disease are still not well understood. A heritability analysis and genome wide linkage analysis of a large twin dataset was undertaken. Treating age of onset of appendicitis as a censored survival trait revealed a heritability of 0.21, and found evidence of linkage to Chromosome 1p37.3.     Tracing Technological Development Trajectories: A Genetic Knowledge Persistence-Based Main Path Approach

    The aim of this paper is to propose a new method to identify main paths in a technological domain using patent citations. Previous approaches for using main path analysis have greatly improved our understanding of actual technological trajectories but nonetheless have some limitations. They have high potential to miss some dominant patents from the identified main paths; nonetheless, the high network complexity of their main paths makes qualitative tracing of trajectories problematic. The proposed method searches backward and forward paths from the high-persistence patents which are identified based on a standard genetic knowledge persistence algorithm. We tested the new method by applying it to the desalination and the solar photovoltaic domains and compared the results to output from the same domains using a prior method. The empirical results show that the proposed method can dramatically reduce network complexity without missing any dominantly important patents. The main paths identified by our approach for two test cases are almost 10x less complex than the main paths identified by the existing approach. The proposed approach identifies all dominantly important patents on the main paths, but the main paths identified by the existing approach miss about 20% of dominantly important patents

    Before main banks : a selective historical overview of Japan's prewar financialsystem

    The postwar experience of the Japanese banking system has received considerable attention recently partly because conditions in defeated Japan in 1945 (including high inflation and the need to switch from a military to a civilian economy) are similar to those in transition economies today. Policymakers in transition economies can learn a good deal from the experiences of Japan's postwar financial system but should remember that Japan also experienced extraordinary industrial growth and financial institution building in the late nineteenth and early twentieth centuries. Lessons to be learned from that experience include the following: Business conglomerates that did not continue to depend on government patronage were more successful than others in making the transition to a modern industrial economy. Banks that made a conscious effort to reduce their dependence on central bank credit were more successful than those that did not. The establishment of procedures for punishing defaulting borrowers helped the development of the payments system. Limits on the amount of lending to related parties appear to have contributed to financial stability (and could have contributed more if the newer"zaibatsu"had been as prudent as the older ones). Bank bailouts without accompanying reform (such as those the Bank of Japan undertook in 1920 and 1922) probably increased the likelihood of a more serious crisis, such as that of 1927. Capital standards - the minimum capital requirements established in the 1927 law - were a viable means of encouraging bank consolidation and more prudent lending.     Higher U (1)-gerbe connections in geometric prequantization

    We promote geometric prequantization to higher geometry (higher stacks), where a prequantization is given by a higher principal connection (a higher gerbe with connection). We show fairly generally how there is canonically a tower of higher gauge groupoids and Courant groupoids assigned to a higher prequantization, and establish the corresponding Atiyah sequence as an integrated Kostant-Souriau infinity-group extension of higher Hamiltonian symplectomorphisms by higher quantomorphisms. We also exhibit the infinity-group cocycle which classifies this extension and discuss how its restrictions along Hamiltonian infinity-actions yield higher Heisenberg cocycles. In the special case of higher differential geometry over smooth manifolds, we find the L-infinity-algebra extension of Hamiltonian vector fields - which is the higher Poisson bracket of local observables - and show that it is equivalent to the construction proposed by the second author in n-plectic geometry. Finally, we indicate a list of examples of applications of higher prequantization in the extended geometric quantization of local quantum field theories and specifically in string geometry
