    Modulation of laminin synthesis in human neuroblastoma cells during retinoic acid induced differentiation.

    A comparable pattern of morphological neuronal differentiation was induced in the human neuroblastoma cell line SMS-KCNR by treatment with either retinoic acid (RA) or exogenous laminin (LM). LM expression and synthesis by SMS-KCNR was increased upon RA treatment which involved the cell bound, rather than the secreted protein. These data suggest an involvement of LM in the neuroblastoma differentiation process manifested both as an ability of LM to induce a morphological neuronal differentiation and as a selective control on LM metabolism during RA induced neuronal differentiation

    the effects of training on muscle structure are dependent on adaptive changes induced by different intensities of physical exercise. evidence has shown that aerobic training is able to induce adaptive changes to muscle structure based on intensity. the aim of the present study was to investigate the effects of different methods of continuous aerobic training in mice using functional, morphological and biomolecular approaches. the continuous aerobic training methods used in the present study were uniform continuous training (UC), varying continuous training (VC) and progressive continuous training (PC). mice were made to run 3 times a week for 12 weeks on a motorized rotarod, following one of the three different training methods at different speeds. the results of the present study demonstrated that the various training methods had different effects on sarcomere length. ultrastructural analysis demonstrated that UC training resulted in a shortening of sarcomere length, PC training resulted in an elongation of sarcomere length and VC training showed similar sarcomere length when compared with the control sedentary group. additionally, succinate dehydrogenase complex flavoprotein subunit a levels in muscle tissue following VC training were higher compared with UC and PC training. overall, the present study showed that varying exercise methods resulted in different types of muscle plasticity, and that the VC protocol resulted in increased coordination and strength endurance in the functional tests, in agreement with the ultrastructural and biochemical profile. these observations support the view that VC training may be more efficient in increasing performance and may thus form the basis of training regimens when an improvement of motor efficiency is required

    Topical use of sucralfate in epithelial wound healing: clinical evidences and molecular mechanisms of action

    Sucralfate is a basic aluminium salt of sucrose octasulphate which was orally employed for prevention and treatment of several gastrointestinal diseases including gastroesophageal reflux, gastric and duodenal ulcer. Recent studies have employed sucralfate as a topical drug for the healing of several types of epithelial wounds such as ulcers, inflammatory dermatitis, mucositis and burn wounds. Epithelial wound healing is a well orchestrated process involving hemostasis, inflammatory reaction, cell proliferation and tissue remodelling which leads to granulation tissue development and filling of the wound space. This report will review clinical evidences on the use of topical sucralfate for the management of epithelial lesions and deal with the current knowledge on the molecular mechanisms of action of this compound towards the epithelial wound healing process and will also discuss relevant patents

    Inhibition of nuclear accumulation of phosphorylated ERK by tropomyosin-1-mediated cytoskeletal reorganization

    AIM: Most neoplastic cells express diminished levels of tropomyosin (TM) family of actin-binding proteins, which leads to the formation of poorly organized cytoskeleton. The aberrant cytoskeleton is hypothesized to contribute to the neoplastic phenotype through deregulation of intracellular signaling. The aim of the study is to evaluate whether reorganization of cytoskeleton modulates signaling pathways. Methods: We have utilized ras-transformed NIH3T3 (DT) cells and those transduced with TM1 (DT/TM1) as a model system. DT cells are highly malignant whereas the DT/TM1 cells contain reorganized cytoskeleton and exhibit revertant phenotype. Activation status of ras oncogene in DT and DT/TM1 cells was measured by GTP loading. Activation status and subcellular localization of extracellular signal regulated kinase (ERK) was measured in total, cytoplasmic and nuclear compartments by immunoblotting and confocal microscopy. Results: Transduction of TM1 does not alter the activation status of oncogenic ras. Both parental DT and DT/TM1 cells exhibit similar levels of activated ERK in total cellular lysates, whereas DT/TM1 cells contain significantly less phosphorylated ERK pERK) in the nuclear fraction. Disruption of cytosekeletal integrity results in increased nuclear content of pERK, suggesting that tropomyosin-1-induced microfilaments are critical for curtailing the nuclear accumulation of activated ERK, and may contribute to the anti-oncogenic effects of TM1. Conclusion: Our data suggest that spatiotemporal regulation of ERK by cytoskeleton is an important mechanism. Furthermore, aberrant microfilaments, present in neoplastic cells, fail to restrict nuclear localization of ERK, and hence contribute to deregulated ERK signaling. © 2008 MedUnion Press

    Hydrodynamic properties of whole arabic gum

    The most economically important of the hydrodynamic properties of a material are viscosity and density, which allow determining the intrinsic viscosity of raw materials used in the food industry. They serve as an indirect measure of molecular weight (M), hydrodynamic radius (RH), number of Simha, (ν(P)), Perrin parameter (P); hydration value (δ), Scheraga-Mandelkern parameter (β), and Flory parameters (0 and P0). Normally, these parameters are reported at a temperature of 25ºC, which limits their use at different temperatures. This work studies the temperature-dependence of whole arabic gum (WAG) in aqueous solution, finding that in aqueous solution, this biopolymer presents a random coil shape with ν(p) ≈ 2.55. The behavior of WAG in this system indicates that it behaves as a colloidal particle that tends to compact as temperature increases (RH decrease). The M of WAG calculated here are 760000 g/mol. The Mark-Houwink-Sakurada equation constants, a and k, for WAG in water solvent-temperature systems have been reported already, where the value of a ranges from 0.5496 to 0.5085 within a temperature range of 20 to 50°C.Fil: Masuelli, Martin Alberto.     Acacia caven gum studies of hydrodynamic parameters

    The acacia caven gum (ACG) is an exudate from Acacia caven tree, this gum is dissolved in water and purified by precipitation with ethanol. In this paper the intrinsic viscosity, [η], is calculated by dissimilar equations, as Huggins, Kramer, Arrhenius-Rother-Hoffmann, Staudinger & Heuer, Square (S), Square root (SR), S-SR average plot, average values of S-SR, and being Kraemer is taken as standard. The different methods are evaluated and compared. From [η] measurements are determined the molecular weight (MV) and hydrodynamic radius (RH), with values of 1060 Kg/mol and 24 nm, respectively. This polysaccharide acquires a random coil conformation with Mark-Houwink parameters of a=0.5507 and k=0.0225 cm3/g.
