    Molar concentration from sequential 2-D water-window X-ray ptychography and X-ray fluorescence in hydrated cells

    Recent developments in biological X-ray microscopy have allowed structural information and elemental distribution to be simultaneously obtained by combining X-ray ptychography and X-ray fluorescence microscopy. Experimentally, these methods can be performed simultaneously; however, the optimal conditions for each measurement may not be compatible. Here, we combine two distinct measurements of ultrastructure and elemental distribution, with each measurement performed under optimised conditions. By combining optimised ptychography and fluorescence information we are able to determine molar concentrations from two-dimensional images, allowing an investigation into the interactions between the environment sensing filopodia in fibroblasts and extracellular calcium. Furthermore, the biological ptychography results we present illustrate a point of maturity where the technique can be applied to solve significant problems in structural biology

    TeV gamma-ray emission from accreting binary pulsars

    The high-mass X-ray binary SMC X-1 has been observed during the interval 1986-1989 at a threshold γ-ray energy of 0.4 TeV using the University of Durham Mark III air Cerenkov telescope at Narrabri. The Cerenkov arrival time series recorded during these observations have been tested for periodicity at the contemporary pulse period of the SMC X-1 X-ray pulsar. This period analysis was performed using a new technique - introduced in the present work - which is designed for TeV γ-ray source candidates in binary systems and which simultaneously seeks evidence for pulsed TeV γ-ray emission and information on the location of the emission site within the binary. Results are also presented from the period analysis of data accumulated at Narrabri during observations of the low-mass X-ray binaries X0021.8-7221 (in the globular cluster 47 Tucanae) and GX 1+4. X-ray binaries as a class of TeV γ-ray source are considered and their properties compared. Previous models which invoke the interaction of beams of high-energy particles with material within the environment of a binary system are discussed. One of these models is adapted in the present work in order to propose a simple explanation of the narrow TeV 7-ray orbital light curve observed for the high-mass binary Centaurus X-3. The new model accounts for the γ-ray emission from Cen X-3 in terms of the collisions of ultrarelativistic protons with an accretion wake trailing the X-ray pulsar. This model, which incorporates the steering of charged particles in the magnetosphere of the massive primary star, is also applied to the Vela X-1 binary in order to investigate claims that TeV γ-ray outbursts observed during X-ray eclipse arise at particle collisions with the limb of the supergiant companion

    A search for TeV gamma ray emission from X-ray binary stars

    This work is concerned with the detection of pulsed TeV gamma ray emission from a number of X-ray binary systems by the use of the atmospheric Cerenkov technique. Chapters 1 and 2 give an overview of the development of gamma ray astronomy, with emphasis placed on progress made in the detection of TeV gamma rays by their Cerenkov radiation in the atmosphere. Chapters 3 and 4 describe the University of Durham atmospheric Cerenkov telescopes, which were used to make the observations reported in this work, and the standard data processing and analysis procedures adopted. The main part of the thesis deals with the application of these techniques to observations of five X-ray binaries. After a review of the properties of such objects in Chapter 5, Chapters 6 and 7 deal specifically with the results for two of the systems considered to be among the most likely candidates to give a detectable TeV photon flux; Centaurus X-3 and Vela X-1. A study of all data recorded on Cen X-3 over the course of six years suggests the presence of a weak gamma ray flux pulsed at the X-ray period. Previous reports of stronger emission near the ascending node of the orbit are confirmed here. For Vela X-1, the analysis of a dataset recorded during a single dark moon interval reveals evidence for two short outbursts of pulsed TeV gamma ray emission. Chapter 8 reports the series of observations made of SMC X-1, 4U1626-67 and X0G21-72, and upper limits are placed on the TeV gamma ray emission from each. Finally, the results reported here are compared with the predictions of a number of theoretical models, some of which are found to give good agreement with the limits and detections derived in this work. A discussion of the status of this field and future observational prospects is also given

    Active X-ray optics for the next generation of X-ray space telescopes

    Described within is the design, manufacture, metrology and X-ray testing of an active X-ray prototype intended for the next generation of X-ray telescopes. One of the challenges faced by the X-ray telescope community is how to combine high resolution and high sensitivity into one system, as weight limitations place constraints on the optics that can be launched. Therefore the mandate of the active X-ray prototype is to provide high sensitivity through the ability of the optics to be nested and to deliver high angular resolution through the active control of the optic’s form. Piezoelectric unimorph actuators provide the active component: it is intended that they will correct for figure errors within the optic and therefore increase the angular resolution capability. The prototype’s design is based upon an ellipsoidal segment which provides point-to-point focussing of an X-ray source. The prototype itself is composed of an electroformed nickel optic where the non-reflective surface is populated with 30 piezoelectric actuators and it is the production of the prototype that is the core of the presented research. Metrology of the actuators’ influence functions is presented and highlight the prototype’s ability to deform its optic surface by microns. In addition, the measured influence functions are compared against finite element models and a distinct similarity between the functions is observed. The prototype was tested at an X-ray beamline facility in November 2008 and the results showed the prototype’s ability to correct the optic to achieve an improved angular resolution: from 0.786 arc-minutes to 0.686 arc-minutes in terms of full width half maximum. Finally, difficulties in the manufacture of the prototype and X-ray testing shall be presented alongside future work in conclusion to this thesis

    A 2 h periodic variation in the low-mass X-ray binary Ser X-1

    Spectroscopy of the low-mass X-ray binary Ser X-1 using the Gran Telescopio Canarias have revealed a ?2 h periodic variability that is present in the three strongest emission lines. We tentatively interpret this variability as due to orbital motion, making it the first indication of the orbital period of Ser X-1. Together with the fact that the emission lines are remarkably narrow, but still resolved, we show that a main-sequence K dwarf together with a canonical 1.4 M? neutron star gives a good description of the system. In this scenario, the most likely place for the emission lines to arise is the accretion disc, instead of a localized region in the binary (such as the irradiated surface or the stream-impact point), and their narrowness is due instead to the low inclination (?10°) of Ser X-1

    Gamma ray Cerenkov telescope image analysis

    The subject of this thesis is ground based gamma ray astronomy using the imaging atmospheric Cerenkov technique. The first two chapters are introductory, and describe the field of gamma ray astronomy, the generation of extensive air showers in the atmosphere and the Cerenkov radiation they induce. Chapter three describes the atmospheric Cerenkov telescope, including the development of the imaging technique for background discrimination. The characteristics of the three University of Durham atmospheric Cerenkov telescopes and the processing and calibration of their data products are outlined. Chapter four is concerned with periodic sources of gamma ray emission and includes a review of candidate sources and time series analysis techniques. An analysis of the Mark 3 telescope SMC X-1 database is presented. An upper limit of 1.2 x 10(^-11) cm(^-2) s(^-1) above a cosmic ray threshold of 1 TeV is determined for the guard ring analysis of Mark 3 data. For an analysis of medium resolution Mark 3 imaging data, the upper limit is 2 x 10(^-10) cm(^-2) s(^-1) above a cosmic ray threshold of 500 GeV. Chapter five introduces a new method for the parameterisation of Cerenkov images of extensive air showers recorded by atmospheric Cerenkov telescopes. This method, involving the optimization of a bivariate Gaussian fit to the image, is shown to be significantly better than the standard moment based parameterisation using simulated images. In Chapter six, both of these methods are employed in an attempt to enhance the signal to noise ratio for observations of the pulsar PSR 1706-44 made with the Mark 6 telescope and some evidence for steady emission is seen. The implied fluxes are (2.6 ± 0.3 ± 0.1)x 10(^-11) cm(^-2) s(^-1) above 420 GeV for the bivariate Gaussian analysis and (1.7 ± 0.4 ± 0.2)x10(^-11) cm(^-2) s(^-1) above 500 GeV for the moment analysis
