Human bone marrow mesenchymal stem cell-derived extracellular vesicles attenuate neuroinflammation evoked by focal brain injury in rats
Background Ischemic stroke is the major cause of long-term severe disability and death in aged population. Cell death in the infarcted region of the brain induces immune reaction leading to further progression of tissue damage. Immunomodulatory function of mesenchymal stem cells (MSCs) has been shown in multiple preclinical studies; however, it has not been successfully translated to a routine clinical practice due to logistical, economical, regulatory, and intellectual property obstacles. It has been recently demonstrated that therapeutic effect of intravenously administered MSCs can be recapitulated by extracellular vesicles (EVs) derived from them. However, in contrast to MSCs, EVs were not capable to decrease stroke-induced neuroinflammation. Therefore, the aim of the study was to investigate if intra-arterial delivery of MSC-derived EVs will have stronger impact on focal brain injury-induced neuroinflammation, which mimics ischemic stroke, and how it compares to MSCs. Methods The studies were performed in adult male Wistar rats with focal brain injury induced by injection of 1 mu l of 50 nmol ouabain into the right hemisphere. Two days after brain insult, 5 x 10(5) human bone marrow MSCs (hBM-MSCs) labeled with Molday ION or 1.3 x 10(9) EVs stained with PKH26 were intra-arterially injected into the right hemisphere under real-time MRI guidance. At days 1, 3, and 7 post-transplantation, the rats were decapitated, the brains were removed, and the presence of donor cells or EVs was analyzed. The cellular immune response in host brain was evaluated immunohistochemically, and humoral factors were measured by multiplex immunoassay. Results hBM-MSCs and EVs transplanted intra-arterially were observed in the rat ipsilateral hemisphere, near the ischemic region. Immunohistochemical analysis of brain tissue showed that injection of hBM-MSCs or EVs leads to the decrease of cell activation by ischemic injury, i.e., astrocytes, microglia, and infiltrating leucocytes, including T cytotoxic cells. Furthermore, we observed significant decrease of pro-inflammatory cytokines and chemokines after hBM-MSC or EV infusion comparing with non-treated rats with focal brain injury. Conclusions Intra-arterially injected EVs attenuated neuroinflammation evoked by focal brain injury, which mimics ischemic stroke, and this effect was comparable to intra-arterial hBM-MSC transplantation. Thus, intra-arterial injection of EVs might be an attractive therapeutic approach, which obviates MSC-related obstacles
Zachowania młodych konsumentów na rynku m-zakupów
This article is of an empirical nature. Its main objective is to identify the behaviour of young consumers (aged 15–34) on the m-shopping market, taking into consideration three age groups: 15–18, 19–24 and 25–34. This segment is very sensitive to modern technological solutions and interested in novelties, which is often an expression of their personality and being in line with new consumer trends. The article is based mainly on the results of the report mShopper 2.0. Polacy na zakupach mobilnych from 2016 and the results of own research conducted in 2013 as a part of the research project Consumer Competence as a Stimulator of Innovative Behavior and Sustainable Consumption (2011/03/B/HS4/04417).</jats:p
A Comprehensive Survey on Antennas On-Chip Based on Metamaterial, Metasurface, and Substrate Integrated Waveguide Principles for Millimeter-Waves and Terahertz Integrated Circuits and Systems
Antennas on-chip are a particular type of radiating elements valued for their small footprint. They are most commonly integrated in circuit boards to electromagnetically interface free space, which is necessary for wireless communications. Antennas on-chip radiate and receive electromagnetic (EM) energy as any conventional antennas, but what distinguishes them is their miniaturized size. This means they can be integrated inside electronic devices. Although on-chip antennas have a limited range, they are suitable for cell phones, tablet computers, headsets, global positioning system (GPS) devices, and WiFi and WLAN routers. Typically, on-chip antennas are handicapped by narrow bandwidth (less than 10%) and low radiation efficiency. This survey provides an overview of recent techniques and technologies investigated in the literature, to implement high performance on-chip antennas for millimeter-waves (mmWave) and terahertz (THz) integrated-circuit (IC) applications. The technologies discussed here include metamaterial (MTM), metasurface (MTS), and substrate integrated waveguides (SIW). The antenna designs described here are implemented on various substrate layers such as Silicon, Graphene, Polyimide, and GaAs to facilitate integration on ICs. Some of the antennas described here employ innovative excitation mechanisms, for example comprising open-circuited microstrip-line that is electromagnetically coupled to radiating elements through narrow dielectric slots. This excitation mechanism is shown to suppress surface wave propagation and reduce substrate loss. Other techniques described like SIW are shown to significantly attenuate surface waves and minimise loss. Radiation elements based on the MTM and MTS inspired technologies are shown to extend the effective aperture of the antenna without compromising the antenna's form factor. Moreover, the on-chip antennas designed using the above technologies exhibit significantly improved impedance match, bandwidth, gain and radiation efficiency compared to previously used technologies. These features make such antennas a prime candidate for mmWave and THz on-chip integration. This review provides a thorough reference source for specialist antenna designers

Glucocorticoids upregulate decreased IL-7 receptor expression in asthmatic patients and simian immunodeficiency virus-infected non-human primates
Signaling through interleukin-7 receptor (IL-7R) is essential for regulation of T-cell homeostasis and survival. Previously, we and others have found diminished IL-7R levels in simian immunodeficiency virus (SIV) - infected non-human primates and human immunodeficiency virus (HIV) - infected patients. To date, it remains unknown whether changes in IL-7R expression could also be linked to non-infectious inflammatory diseases such as asthma or anti-inflammatory drug use. Here, we investigated through flow cytometry the levels of IL-7R expression on CD4+ and CD4- T-cells in asthmatic patients in relation to disease severity, immune status and glucocorticoid (GC) use. In addition, we sought to evaluate the effects of in vivo and in vitro GC treatment on IL-7R expression in both asthmatic patients and SIV-infected non-human primates. We demonstrated that expression of IL-7R on peripheral blood CD4+ T-cells was significantly decreased in clinically stable GC-naive mild and moderate asthmatic patients. Accordingly, the development of asthmatic reaction following bronchial allergen challenge performed in sensitized subjects was associated with a significant drop in levels of IL-7R on circulating CD4+ T-cells. In contrast, CD4+ T-cells from both, mild and moderate, but not severe asthmatics, treated with inhaled GC displayed levels of IL-7R similar to that seen in healthy controls. We did not find significant differences with serum or sputum interleukin-7 levels among healthy controls and GC-naïve and GC-treated asthmatic patients. Furthermore, both in vitro GC treatment and short-term oral GC administration to asthmatic patients resulted in a significant enhancement of IL-7R. Similarly, we demonstrated that GC-stimulated T-cells from SIV-infected non-human primates up-regulated IL-7R expression. Accordingly, experimental short-term systemic in vivo administration of GC to SIV-infected macaques led to enhancement of IL-7R expression on circulating T-cells. Our data indicate that GC bear potential to recover diminished IL-7R expression, as well in asthma as in lentiviral infection
Health on the net: the doctor answers
This study is intended to provide new insights into web-based discourse on medical exchanges in doctor-patient communication.
The global net system have certainly played an important role in the increase and the following modification of communication dynamics. However, in particular situation such as doctor-patient interviews, computer mediated communication is still a limit.
This paper is based on two lines of research in language studies, namely the studies of language corpus and studies of communication in health encounters. The corpus is formed by the medical entries that appeared in biomedical/health service sites.
The samples collected were analysed in order to find discourse patterns for structure and organization of moves together with their functional meaning. For each move, grammatical and lexical items were identified in order to assess the dimension of evidentiality, modality, and affect in online counseling.
The framework of this study has been shaped by the typologies in Chafe (1986), Willett (1988), Van Auwera and Plungian (1998), Plungian (2001), Fitneva (2001) and Marìn Arrese (2002, 2004) on the roles and expression of evidentiality and modality in English. Emotive load is studied through the lexical analyses of emotions expressed in first person following the Appraisal Framework by Martin and White (2005)
