Comparative kinetic analysis of glycerol 3-phosphate cytidylyltransferase from Enterococcus faecalis and Listeria monocytogenes
Background: Glycerol 3-phosphate cytidylyltransferase (GCT) is an enzyme central to the synthesis of teichoic acids, components of the cell wall in gram positive bacteria. Catalysis by GCT from Enterococcus faecalis and Listeria monocytogenes has been investigated and catalytic properties compared. Material/Methods: The genes encoding GCT were cloned from genomic DNA and recombinant proteins expressed in E. coli and purified. Enzyme assays were used to determine kinetic constants k(cat) and K-m. Chemical crosslinking provided a means to assess quaternary structure of each GCT. Results: Recombinant Enterococcus faecalis GCT had an apparent k(cat) value of 1.51 s(-1) and apparent K-m values of 2.42 mM and 4.03 mM with respect to substrates cytidine 5\u27-triphosphate (CTP) and glycerol phosphate. Listeria monocytogenes GCT had an apparent k(cat) value of 4.15 s(-1) and apparent K-m values of 1.52 mM and 6.56 mM with respect to CTP and glycerol phosphate. This resulted in k(cat)/K-m values of 0.62 s(-1)mM(-1) and 0.37 s(-1)mM(-1) for E. faecalis GCT and 2.73 s(-1)mM(-1) and 0.63 s(-1)mM(-1) for L. monocytogenes GCT with respect to CTP and glycerol phosphate, respectively. Conclusions: The genome of both Enterococcus faecalis and Listeria monocytogenes contain a gene that encodes a functional GCT. The genes are 67% identical at the nucleotide level and the encoded proteins exhibit a 63% amino acid identity. The purified, recombinant enzymes each appear to be dimeric and display similar kinetic characteristics. Studying the catalytic characteristics of GCT isoforms from pathogenic bacteria provides information important for the future development of potential antibacterial agents

Using A Creative Story Game to Enhance Eighth Grade Students' Active Participation

This study assessed the effects of the implementation of a creative story game on the improvement of eight grade students’ active participation in speaking. Based on the preliminary study, it found a problem dealing with the students’ involvement in teaching learning activity. The class consisted of 32 students, yet only few students,about 40% students,who engaged in the activity. Only one or two students in the group who presented the work who were actively involved during the presentation session. Others were not interested in the presentation and they did not pay attention to the presentation. Referring to the problem, this study proposed a creative story game which focused on the improvement of the eight grade students’ active participation in speaking. A creative story game was combined with a group work because a group work is one recommended techniques that can stimulate students’ active participation. The research problem of this study was formulated as follows;“How can a creative story game improve eight grade students’ active participation in speaking?” The research design of this study was classroom action research which consisted of four major stages; planning, implementing, observing, and reflecting. Students were divided into several groups and were asked to compose the narrative text in which the orientation had been set. The students were asked to discuss with their groups in composing the story. Next, each groups presented their works. Finally, the students voted the best story among the groups. The competitive element in this technique made this technique interesting for the students because it also stimulated the students to actively participate in the teaching learning activity. This study was conducted in two cycles. Cycle one was done in three meetings, whereas cycle two was done in two meetings. The study was conducted in VIII-E class at SMPN 2 Pandaan in second semester year 2013/2014. The instruments used in this study were observation sheet, field notes, questionnaire, and interview guide. The observation sheet was designed by modifying the six categories of student-active proposed by Flint of G. Moskowitz (1988 cited in Wulandari, 2010) covering: (1) Student answer the teacher’s or other students’ question orally, (2) Student asks the teacher if he/she does not understand about something, (3) Student gives his/her opinion, states his/her suggestion and expresses his/her feeling orally about something, (4) Student comes in front of the class to perform something, (5) Student is involved in-group activity, and (6) Student supportively responds to the teacher’s instructions. The field notes covered the data dealing with the implementation in the teaching learning process than could not be covered in observation sheet. The questionaiire was designed to kknow the students’ response on the implementation and the interview guide was designed as the guidance for the researcher when conducting the interview session. There were two types of interview guide, that are; (1) Interview guide in preliminary study, and (2) Interview guide in the implementation. The findings of this study showed that the implementation was successful in cycle two. It was found that 64.3% of the students participated actively in class
Evaluasi Kinerja Simpang Bersinyal Jl. Kapasari - Jl. Kalianyar - Jl. Kusuma Bangsa - JL. Ngaglik Surabaya
Simpang bersinyal pada Jl. Kapasari - Jl. Kalianyar - Jl. Kusuma Bangsa - Jl. Ngaglik merupakan salah satu persimpangan yang memiliki tingkat kepadatan yang cukup tinggi, terutama pada saat jam puncak tertentu. Beberapa faktor kemacetan dikarenakan adanya sekolah dan pasar tumpah yang menyebabkan antrian panjang terhadap kendaraan yang akan melewati persimpangan tersebut. Selain itu, banyaknya pemakai jalan yang tidak disiplin dalam berlalu lintas, keberadaan warung-warung serta kendaraan yang parkir sembarangan disekitar lokasi simpang juga menambah masalah yang terjadi. Permasalahan, bagaimana kinerja simpang bersinyal pada Tahun 2017 dan 5 tahun setelahnya yaitu 2022.
Analisa kinerja persimpangan menggunakan MKJI 1997 dengan program bantuan KAJI. Analisa diawali dari pengumpulan data-data jumlah penduduk, tata guna lahan dan pertumbuhan bangkitan jumlah kendaraan yang diperoleh dari BAPPEKO, BPS dan DISPENDUK CAPIL Surabaya sebagai data sekunder, sedangkan data primer didapat dari hasil survey di lapangan. Selanjutnya dilakukan analisa kondisi persimpangan dan segmen menggunakan acuan MKJI 1997.
Berdasarkan hasil analisa dan evaluasi kondisi eksisting tahun 2017 persimpangan Jl. Kapasari - Jl. Kalianyar - Jl. Kusuma Bangsa - Jl. Ngaglik didapatkan tingkat pelayanan (Level of Service) pada hari kerja jam puncak pagi berada pada tingkat E dengan tundaan rata-rata simpang sebesar 48,57 det/smp, pada puncak jam siang berada pada tingkat E dengan tundaan rata-rata simpang 42,44 det/smp dan pada jam puncak sore berada pada tingkat F dengan tundaan rata-rata simpang 115,47 det/smp. Dengan kondisi persimpangan yang sudah mencapai tingkat pelayanan LOS F terutama pada jam puncak sore, maka kinerja simpang saat ini pada tahun 2017 sudah tidak dapat dipertahankan. Selanjutnya, melakukan alternatif perbaikan simpang yaitu dengan mengubah waktu sinyal dan manajemen lalu lintas dari 3 fase menjadi 4. Dengan menggunakan alternatif terpilih tersebut, maka didapatkan tingkat pelayanan LOS E dengan (DI) 48,64 det/smp serta QL maksimum 170 m < 435,57 m (pada eksisting Tahun 2017). Hasil ini terlihat lebih baik dari kondisi eksisting.

The Signalized Intersection on Jl. Kapasari - Jl. Kalianyar - Jl. Kusuma Bangsa - Jl. Ngaglik is one of the intersections that has a high enough density, especially during certain peak hours. Several factors of congestion due to the existence of schools and spill markets that indirectly disrupt the road body causing long queues of vehicles going through the intersection. In addition, the number of road users who are not disciplined in traffic, the existence of stalls and vehicles parked carelessly around the intersection also adds to the problems that occur. Problem, how the performance of this intersection on 2017 and also 2022.
Analysis of intersection performance using MKJI 1997 with KAJI assistance program. The analysis begins with the collection of population data, land use and the growth of the number of vehicles obtained from BAPPEKO, BPS and DISPENDUK CAPIL Surabaya as sekuder data, while the primary data obtained from the survey results in the field. Futhermore, the analysis of junction and segment conditions using MKJI reference 1997.
Results, level of service (LOS) average delay for this intersection as performance indicator as mentioned, LOS E with 48.57 sec/smp for morning peak hour, LOS E with 42.44 sec/smp for noon peak hour and LOS F with 115.47 sec/smp for afternoon peak hour. With crossing conditions that have reached the service level of LOS F especially on afternoon peak hour, the current intersection performance in 2017 is not maintained. Then, improve alternatives signalized intersection by changing the signal time and traffic management from 3 phase become 4 phase. By using the selected alternative, performance increase LOS E with 48,64 sec/smp and maximum QL 170 m < 435,57 m (on existing year 2017). These results look better than the existing conditions
Logical Intelligence Enhancement Test: a measure for the evaluation of logical skills in primary school
Il presente lavoro si inserisce in uno studio pilota condotto nell’anno scolastico 2018-2019per valutare l’efficacia del Logical Intelligence Enhancement Programme (LIEP, Calvani, Peru,& Zanaboni, 2019), un programma composto da tavole di logica verbale e numerica volte asviluppare il ragionamento logico e le capacità metacognitive di studenti di quinta primaria.All’interno di questo studio è stato sviluppato il Logical Intelligence Enhancement test perstimare gli effetti del training sulle abilità logico-verbali. Il contributo presenta la costru-zione del test, le sue caratteristiche psicometriche e la validazione. A tal fine, lo strumentoè stato somministrato a un campione di 173 studenti di classe quinta primaria nell’area diPalermo e Firenze.Il LIEP Test si compone di 22 item e include otto dimensioni di esercizilogici come il completamento di tabelle o serie di numeri, la risoluzione di problemi nu-merici o geometrici e tavole di inferenza logica.This work is part of a pilot study conducted during 2018-2019 to evaluate the effectivenessof the Logical Intelligence Enhancement Programme (LIEP, Calvani, Peru & Zanaboni, 2019),a program which consists of logical-verbal and numeric exercises to enhance logical rea-soning and metacognitive skills of fifth grade students. During this study the Logical Intel-ligence Enhancement test was developed to measure the effects of the training on thelogical-verbal skills. This paper describes the development of the LIEP test, its psychometricfeatures and validation process. For the validation the measure was administered to a sam-ple of 173 fifth grade students in Palermo and Florence. The final version of the test consistsof 18 items and includes eight dimensions of logical exercises, such as tables to be com-pleted, numeric series, problem solving and logical inferences
The significant role of chloroplast thioredoxins in the regulation of photosynthesis
This is a review article presenting, comparing and analysing the recent studies on plant chloroplast thioredoxin regulation on photosynthesis. There was a seed paper provided for this essay (Okegawa and Motohashi, 2015). We needed to do further research in the corresponding area and find subsequent journals after this paper, and compare their results and present new findings, as well as showing the importance of these findings and suggest further research directions. Photosynthesis is rather important in plant which converts light into chemical energy to ensure plants’ growth and productivity. Thioredoxins (Trxs) are ubiquitous proteins involved in regulation of redox reaction in chloroplast in photosynthesis. There are multiple isoforms of thioredoxins, and their roles are explained in the article. Trx m is the most abundant protein in chloroplast stroma, which predominantly regulates redox-dependant Calvin cycle enzymes, and further studies demonstrated the role of Trx m in protecting plant photosynthesis from excessive light. Trx f is indispensable for activation of the important Calvin cycle enzymes FBPase and Rubisco activase, and Trx f is essential for plants’ adaptation to short-day conditions and of photosynthetic efficiency. It would be challenging to investigate Trx systems with in vivo studies and produce convincing results. More research is required as the inconsistencies in previous research and the lack of study done in vivo. Regarding the importance of Trx systems in photosynthetic efficiency in plants, it could be used in increasing crop yield for the increasing population worldwide
