    Editorial: Chronic autoimmune arthritis, infections and vaccines

    The link between autoimmunity and infection continues to represent an intriguing immunologic conundrum for scientist and a frequent clinical challenge for patients and physicians. Patients with chronic autoimmune arthritis indeed have an increased risk of infections, mainly due to the dysregulation of their immune system and the use of immunosuppressive therapy. Infections in these patients are more frequent, have a more severe clinical course, eventually with prolonged viral persistence, compared to the general population and represent a frequent cause of death. Besides, infections can trigger autoimmune diseases via different immunologic mechanisms such as molecular mimicry, epitope spreading, by-stander activation and can also induce disease relapses. SARS-CoV-2 infection represents a dramatic example of this complex connection. It is known, indeed, that different autoimmune manifestations can complicate SARS-CoV-2 infection such as uncontrolled host-immune response leading to life-threatening condition known as cytokine release syndrome, or autoimmune hemolytic anemia, immune thrombocytopenic purpura, Guillain-Barre syndrome, and the detection of different autoantibodies. This Research Topic includes seventeen contributions, fifteen original articles and two review articles, providing several new insights into the efficacy and safety of SARS-CoV-2 vaccine in autoimmune patients, immunologic biomarkers for diagnosis and therapeutic outcome of autoimmune arthritis

    Recent Results From the EU POF-PLUS Project: Multi-Gigabit Transmission Over 1 mm Core Diameter Plastic Optical Fibers

    Recent activity to achieve multi-gigabit transmission over 1 mm core diameter graded-index and step-index plastic optical fibers for distances up to 50 meters is reported in this paper. By employing a simple intensity-modulated direct-detection system with pulse amplitude or digital multi-tone modulation techniques, low-cost transceivers and easy to install large-core POFs, it is demonstrated that multi-gigabit transmission up to 10 Gbit/s over 1-mm core diameter POF infrastructure is feasible. The results presented in this paper were obtained in the EU FP7 POF-PLUS project, which focused on applications in different scenarios, such as in next-generation in-building residential networks and in datacom applications

    Monocytes of Patients with Systemic Sclerosis (Scleroderma) Spontaneously Release In Vitro Increased Amounts of Superoxide Anion

    It has been suggested that toxic oxygen free radicals can be involved in the pathogenesis of systemic sclerosis (scleroderma) (SSc). Because the cells that contribute to the generation of free radicals are not known, our aim was (i) to evaluate the ability of unmanipulated and phorbol 12-myristate 13-acetate-stimulated monocytes and polymorphonucleate neutrophils of SSc patients to generate superoxide anion (O2·–); and (ii) to investigate whether the O2·– produced by these cells involved the activation of nicotinamide-adenine dinucleotide diphosphate oxidase biochemical pathway. Employing the superoxide dismutase-inhibitable reduction of cytochrome c to evaluate the generation of O2·–, unmanipulated monocytes of SSc patients generated more O2·– than primary Raynaud’s phenomenon patients and normal control monocytes (p= 0.0001), and the release was higher in patients with diffuse cutaneous involvement and 5 y or less disease duration (p = 0.02). The involvement of nicotinamide-adenine dinucleotide diphosphate oxidase in the enhanced O2·– production was demonstrated by the finding that the cytosolic components of the enzyme, p47phox and p67phox, were both translocated to the plasma membrane of enriched but otherwise unmanipulated monocytes of SSc patients. The involvement of mitochondrial oxidases was excluded by the lack of inhibition of O2·– production when monocytes were incubated in the presence of rotenone, a mitochondrial oxidase inhibitor. Upon stimulation with phorbol 12-myristate 13-acetate, monocytes of SSc patients produced more O2·– than controls. In SSc patients untreated polymorphonucleate neutrophils generated significantly less O2·– than monocytes (p = 0.0001) and only slightly more than polymorphonucleate neutrophils of primary Raynaud’s phenomenon patients and normal controls (p = 0.03). In conclusion, we demonstrate that in patients with scleroderma, unmanipulated and phorbol 12-myristate 13-acetate-stimulated monocytes release in vitro increased amounts of superoxide anion through the activation of nicotinamide-adenine dinucleotide diphosphate oxidase and, thus, contribute to the oxidative stress found in this disease

    Collagen loss and impaired wound healing is associated with c-Myb deficiency

It is also an established target gene of the transcription factor, c-Myb. When c-myb-/- embryos were examined it was observed that their skin was markedly thinner than normal. Importantly, immunohistochemical investigation showed complete absence of collagen type I. Although these homozygous knock-out embryos fail to develop beyond day 15, fibroblasts established from these embryos (mouse embryonic fibroblasts [MEFs]) show defective proliferative responses. Furthermore, in vitro scratch wound assays demonstrated that these c-myb-/- MEFs also exhibit slower closure than their wild-type counterparts. Embryonic lethality has meant that examination of the role of c-Myb in adult mouse skin has not been reported to date. However, in view of the abundance of collagen type I in normal skin, its role in skin integrity and the in vitro data showing proliferative and migration defects in c-myb-/- MEFs, we investigated the consequences of heterozygous c-myb loss in adult mice on the complex process of skin repair in response to injury. Our studies clearly demonstrate that heterozygous c-myb deficiency has a functional effect on wound repair, collagen type I levels and, in response to wounding, transforming growth factor-β1 (an important collagen stimulating factor) induction expression is aberrantly high.     Dynamic Estimation of Vital Signs with mm-wave FMCW Radar

    In this paper, we propose a method for continuous monitoring of vital signs-in particular, respiration frequency-with a commercial mm-wave radar. The nearly constant frequency (NCF) model is adopted to represent chest displacement due to respiration and simulate radar response. Based on this model, an extended Kalman filter (EKF) based estimator is developed to track the breathing frequency of a person. The impact of dynamic model parameters is investigated in numerical simulation. The possibility to track breathing frequency with the proposed method is demonstrated by experimental data processing.     Digital beamforming receivers (RXs) support MIMO operation and offer great flexibility and accuracy in multi-beam formation and calibration. However, compared with analog phased-array and hybrid systems, due to the absence of any rejection for spatial in-band blockers, the RX/ADC dynamic range and linearity should be high enough to prevent array saturation. Therefore, the use of self-steering spatial notch filters (SNFs) is necessary to aid the digital beamformers and reduce RX/ADC power consumption while strong blockers exist. To address that, the sub-6GHz RXs in [1], [2] synthesize a baseband spatial notch impedance and translate it to RF by passive mixers. However, this technique cannot be directly applied at mm-wave frequencies as the impedance translational performance of the passive mixers degrades significantly. Hence, the mm-wave beamformer in [3] realizes a cascadable SNF at an intermediate frequency (IF). However, the front-end mm-wave components like mixers and phase shifters have to tolerate strong blockers, thus degrading RX linearity. Besides, it uses multiple IF buffers and VGAs for signal scaling and combining, which could be power-hungry if a similar method is adopted to realize a mm-wave SNF. To improve on those limitations, we propose a scalable SNF structure, which (1) suppresses the strongest in-band blocker at mm-wave frequencies, (2) supports N-input-N-output MIMOs, and (3) requires no active blocks except the phase shifters.     Experiencing the armed struggle : the Soweto generation and after

    Includes bibliographical references (p. 354-369).This study explores the experiences of the rank-and-file soldiers of Umkhonto we Sizwe and the Azanian People's Liberation Anny. Extensive interviews by the author and other researchers reveal the voices of the soldiers themselves. The African National Congress and Pan African Congress archives at the University of the Western Cape and the University of Fort Hare supplement and verify these oral testimonies, as do some published sources. Most previously published materials about the armed struggle against apartheid have already focused on diplomacy, strategy and tactics, operations, leadership, and human rights abuses to the neglect of the soldiers' actual experiences. This study complements these with significant new oral history materials from the Soweto generation of soldiers and their successors. When dealing with MK, many authors have documented issues of the camp structure in Angola, and operations inside South Africa, so much of this detail is only addressed briefly, leaving space to explore the soldiers' experiences. In the case of APLA, very little has been written on its history, and more detail is provided on these subjects. This study therefore deals with the soldiers' politicisation and motivation for joining the armed struggle, their experiences in leaving South Africa and training in exile, the crises in exile which limited their effectiveness for a time, their return to fight in South Africa, and their difficulties in the "new" South Africa. These materials reveal that vast problems remain facing these veterans of the struggle against apartheid, and that they have the potential, if properly supported and employed, to contribute substantially to the development of present day South Africa. Conversely, if their neglect continues, they also have the potential to bring vast harm to the country. Further use of the investigative tools of oral history, especially if extended to the former soldiers' vernacular languages, is necessary to augment the history of South Africa, and these soldiers' contributions
