    The giant protein titin forms a unique filament network in cardiomyocytes, which engages in both mechanical and signaling functions of the heart. TTN, which encodes titin, is also a major human disease gene. In this review, we cover the roles of cardiac titin in normal and failing hearts, with a special emphasis on the contribution of titin to diastolic stiffness. We provide an update on disease-associated titin mutations in cardiac and skeletal muscles and summarize what is known about the impact of protein–protein interactions on titin properties and functions. We discuss the importance of titin-isoform shifts and titin phosphorylation, as well as titin modifications related to oxidative stress, in adjusting the diastolic stiffness of the healthy and the failing heart. Along the way we distinguish among titin alterations in systolic and in diastolic heart failure and ponder the evidence for titin stiffness as a potential target for pharmacological intervention in heart disease

    Professionele muzikanten gaan naar een opnamestudio als ze een album willen opnemen van hoge geluidskwaliteit. Voor de beginnende muzikant zijn deze studio’s echter niet te betalen, aangezien een opname minstens \u801500 kost. Het aanschaffen van eigen opnameapparatuur is nog vele malen duurder. In die hoge prijs gaat het El Condor Soundsystem verandering brengen. Het El Condor Soundsystem is een draagbaar apparaat, dat een ruimteopname van gespeelde muziek vastlegt, bewerkt en opslaat als bestand dat leesbaar is op elke PC. Het El Condor Soundsystem kan dankzij zijn algoritmes verschillende bronnen onderscheiden zonder hier een fysieke verbinding mee te hebben. De verschillende geluidssporen worden gebruikt om de muziek automatisch te verfraaien en daarna worden deze weer gemixt tot één eindproduct met CD-kwaliteit. Voor het bewerken en wegschrijven is veel rekenkracht nodig.     Passive and active tension in single cardiac myofibrils

    Single myofibrils were isolated from chemically skinned rabbit heart and mounted in an apparatus described previously (Fearn et al., 1993; Linke et al., 1993). We measured the passive length-tension relation and active isometric force, both normalized to cross sectional area. Myofibrillar cross sectional area was calculated based on measurements of myofibril diameter from both phase-contrast images and electron micrographs. Passive tension values up to sarcomere lengths of approximately 2.2 microns were similar to those reported in larger cardiac muscle specimens. Thus, the element responsible for most, if not all, passive force of cardiac muscle at physiological sarcomere lengths appears to reside within the myofibrils. Above 2.2 microns, passive tension continued to rise, but not as steeply as reported in multicellular preparations. Apparently, structures other than the myofibrils become increasingly important in determining the magnitude of passive tension at these stretched lengths. Knowing the myofibrillar component of passive tension allowed us to infer the stress-strain relation of titin, the polypeptide thought to support passive force in the sarcomere. The elastic modulus of titin is 3.5 x 10(6) dyn cm-2, a value similar to that reported for elastin. Maximum active isometric tension in the single myofibril at sarcomere lengths of 2.1–2.3 microns was 145 +/- 35 mN/mm2 (mean +/- SD; n = 15). This value is comparable with that measured in fixed-end contractions of larger cardiac specimens, when the amount of nonmyofibrillar space in those preparations is considered. However, it is about 4 times lower than the maximum active tension previously measured in single skeletal myofibrils under similar conditions (Bartoo et al., 1993)

    Research Note: On the error behaviour of force and moment sources in simplicial spectral finite elements

    The representation of a force or moment point source in a spectral finite-element code for modelling elastic wave propagation becomes fundamentally different in degenerate cases where the source is located on the boundary of an element. This difference is related to the fact that the finite-element basis functions are continuous across element boundaries, but their derivatives are not. A method is presented that effectively deals with this problem. Tests on one-dimensional elements show that the numerical errors for a force source follow the expected convergence rate in terms of the element size, apart from isolated cases where superconvergence occurs. For a moment source, the method also converges but one order of accuracy is lost, probably because of the reduced regularity of the problem. Numerical tests in three dimensions on continuous mass-lumped tetrahedral elements show a similar error behaviour as in the one-dimensional case, although in three dimensions the loss of accuracy for the moment source is not a severe as a full order.Accepted Author ManuscriptApplied Geophysics and Petrophysic

    Variations of vessel diameter and δ13C in false rings of Arbutus unedo L. reflect different environmental conditions

    Woody species in Mediterranean ecosystems form intra-annual density fluctuations (IADFs) in tree rings in response to changes in environmental conditions, especially water availability.Dendrochronology, quantitative wood anatomy and high-resolution isotopic analysis (using a laser ablation technique) were used to characterize IADFs in Arbutus unedo shrubs grown on two sites with different water availability on the island of Elba (Italy).Our findings show that IADF characterization can provide information about the relationship between environmental factors and tree growth at the seasonal level. At the more xeric site, IADFs mainly located in the early and middle parts of the annual ring, showed a decrease in vessel size and an increase in delta 13C as a result of drought deficit. Opposite trends were found at the more mesic site, with IADFs located at the end of the ring and associated with a lower delta 13C. Moreover, at the first site, IADFs are induced by drought deficit, while at the second site IADFs are linked with the regrowth in the last part of the growing season triggered by favourable wet conditions.This combined approach is a promising way for dating problematic wood samples and interpreting the phenomena that trigger the formation of IADFs in the Mediterranean environment
