    Differential expression and phenotypic activities of osterix in equine mesenchymal stem cell populations

    Bone is one of the few tissues in vertebrates that possess the innate capacity for authentic regeneration, yet approximately 10% of fractures fail to heal adequately. There is considerable overlap in the cellular and molecular events occurring during embryological bone development and fracture repair. Mesenchymal stem cell (MSC) precursors undergo osteogenesis to give rise to osteoblasts, which are responsible for bone matrix synthesis and mineralization. Osteogenesis is regulated by two master transcription factors, runt-related transcription factor 2 (RUNX2) and Osterix (OSX), with interplay from signaling pathways such as the Bone Morphogenetic Protein (BMP) network. MSCs isolated from bone marrow (BM) and adipose tissue (ADI) are most commonly utilized in human and veterinary regenerative medicine applications. BM-MSCs have shown superior osteogenic capacity to ADI-MSCs in many studies, yet sample collection and culture techniques often vary between species. The overall objective of this dissertation was to determine factors that influence the in vitro osteogenic capacity differences of equine adult ADI- and BM-MSCs in order to standardize techniques and provide insights into avenues for improving osteogenesis in less-osteogenic MSC populations. The first study investigated the hypotheses that in vitro cell viability, proliferation, and osteogenic differentiation of MSCs collected from horses under general anesthesia and after euthanasia would be impaired compared to MSCs collected from horses during sedation. The second set of experiments characterized the RUNX2 and OSX response axes of ADI- and BM-MSCs during osteogenesis to test the hypotheses that 1) the relative osteogenic capacities are consistent with resting and inducible RUNX2 and OSX transcript levels, and 2) increasing OSX expression in ADI-MSCs improves their osteogenic capacity. The third series of experiments evaluated the comparative BMP signaling activities in ADI- and BM-MSCs to determine whether 1) osteogenic BMP ligand expression differs, 2) BMP inhibition in BM-MSCs impairs osteogenesis, and 3) BMP supplementation to ADI-MSCs improves osteogenesis. The results indicate that donor status during MSC collection does not affect in vitro cell viability, proliferation, and osteogenic capacity. BM-MSCs have higher OSX expression and intrinsic BMP signaling in both resting and osteogenic conditions than ADI-MSCs. Endogenous BMP activity is necessary for osteogenesis in BM-MSCs, and exogenous BMP supplementation, but not OSX over-expression, was sufficient to improve the osteogenic capacity of ADI-MSCs.     Giving voice to ideas : the role description plays in the diffusion of radical innovations

    Thesis (Ph.D.)--Massachusetts Institute of Technology, Dept. of Urban Studies and Planning, 1999.Includes bibliographical references (leaves 134-144).One of the more stable findings in the "diffusion and knowledge utilization" literature is that simple innovations. those compatible with the existing practices in a field. are spread more easily than those which challenge standard practice. Yet it is the more radical innovations that hold special promise for advancing the practice of a field. Using an action research methodology. the author studied the diffusion of radical innovations in two very different programmatic settings. first in an undergraduate affirmative action program on a university campus and later in a philanthropic-driven effort to fund charitable work with recoverable investments rather than grants--a practice that is called "program-related Investing." The two programs together served as test cases--one as a precipitating paradox and the other as a conscious experiment--in overcoming barriers to the diffusion of an important category of innovations: innovations that require individuals to practice in new ways and acquire new skills, that cause some disruption to the broader organization and that involve the "soft" technologies of knowledge rather than the "hard" material technologies. The literature treated diffuser's descriptions of their innovations as self-evident. whereas the author found that diffusers of these radical. practice innovations unintentionally gave incomplete and in some cases misleading descriptions of their work. An argument is made that effective description must do more than represent the original innovation with some accuracy. It must enable diffusers to teach those aspects of their practice which am difficult for them to make explicit by including opportunities for practicing side-by -side. whether these be through simulated practice worlds or actual ones. It must also enable appropriate transformation of the innovation. This can be best accomplished by structuring a dialogue between diffusers and (potential) users to lift up multiple descriptions of the practice. It is the process of comparing such descriptions that allows diffusers and users to build up an understanding both of the essence of the innovation and of ways in which transformations may preserve or damage this essence.by Kalyn Culler Cohen.Ph.D

    Google Scholar is an increasingly popular search engine with the largest bibliographic database for citation analysis that is accessible to anyone unlike other bibliographic databases which may incur monthly fees. With the increase of publications of scholarly papers, there is an increase in demand for quality measurement of papers for assessing the author’s publication works. The evaluations of author’s publications rely on citation analysis as to achieve an unbiased evaluation of an author’s work. H-index is one of the mostly used citation metric, thus, two unique additional calculations of h-index with unknown authors’ citations per paper and citations without self-citation of the author were included and compared against the original h-index calculation in this report. Hence, this report presents the study of the citation analysis of citation records based on Google Scholar. A python parser tool was implemented to parse information from Google Scholar based on three author name searches namely: “Xiaokui Xiao”, “Yunhao Liu” and “Mong LiLee”. Three different h-index versions were calculated and the results were analyzed. The results have shown that a relatively high self-citation by the authors have significant contribution to the overall citation counts that may affect the h-index value, leading to a slight increase in the value of h-index. However, a small value of self-citation does not affect the h-index value. When comparing the two unique h-index versions, version 2 and 3, version 2 has more impact on the overall value of the h-index. Overall, author “Yunhao Liu” has achieved the highest citation counts and the highest h-index value among the other authors, indicating that its publications have high significant contribution, quality and impact to the other authors. In addition, Google Scholar has limitation such as displaying issues, blockage problems and non-scholarly sources that may distort citation metrics. Thus, there are pros and cons when using Google Scholar as citation analysis source. Hence, the citation statistic results obtained from Google Scholar for the different authors reflect the significance and the impact of the authors ’publications.
