    Rational design of HIV vaccines and microbicides: report of the EUROPRISE network annual conference 2010

    Abstract Novel, exciting intervention strategies to prevent infection with HIV have been tested in the past year, and the field is rapidly evolving. EUROPRISE is a network of excellence sponsored by the European Commission and concerned with a wide range of activities including integrated developmental research on HIV vaccines and microbicides from discovery to early clinical trials. A central and timely theme of the network is the development of the unique concept of co-usage of vaccines and microbicides. This review, prepared by the PhD students of the network captures much of the research ongoing between the partners. The network is in its 5 th year and involves over 50 institutions from 13 European countries together with 3 industrial partners; GSK, Novartis and Sanofi-Pasteur. EUROPRISE is involved in 31 separate world-wide trials of Vaccines and Microbicides including 6 in African countries (Tanzania, Mozambique, South Africa, Kenya, Malawi, Rwanda), and is directly supporting clinical trials including MABGEL, a gp140-hsp70 conjugate trial and HIVIS, vaccine trials in Europe and Africa

    A Pattern-Based System to Support Collaborative Production of Learning Objects

    The use of Web 2.0 technologies has established itself even in corporate environments, profoundly changing both the organizational both operational practices. The so-called " knowledge workers", that are workers whose main object of their business is information, were the main protagonists of this change, adopting very frequently Web 2.0 communication tools for their activities. In previous works, we have identified a methodology that aims to identify and model the collaborative business activities by extracting the latter from a catalogue of models of collaborative activities. The application of the methodology and of the models catalogue to the re-engineering process has been successful in various contexts such as public administration, the ICT Company and Banking Company. In this work, we introduce the experience of using methodology and some collaborative pattern to model the Learning Objects production activities. It has the goal of designing an information system that gives support to collaborative activities related to these processes. The following work is developed as part of a collaborative project between the University of Salento and the CLIO SpA Lecce Company, which has developed a multichannel and interactive system of knowledge transmission called MediaBook. The goal of this work is the review of business processes related to the creation of a MediaBook, in order to improve the support and collaboration between the actors involved making the production phase more efficient. This activity presents different interaction moments between two main actors in the process, which are Author and Publisher. These interactions take place mainly through collaborative tools (chat, mail, etc.) and are not tracked and supported by the production system. Once having modelled the existing process of MediaBook production, we started a phase of review and optimization of the process, paying particular attention to stress collaborative activities carried out by the actors of the process. Re-engineering process was carried out by following a well-defined and methodological approach. In detail, after modelling the processes in an AS-IS version, we identified the major areas of interest and we applied to the latter some collaborative pattern. Subsequently we redesigned in the TO-BE version the MediaBook production process using the patterns. The new process highlights several collaborative aspects that were not raised in the AS-IS version of the process. It allows, in this way, to develop a truly collaborative model of activities to support the production of Learning Object. The used patterns allow to manage and coordinate the collaborative activities (eg: Deadline Agreement, Retrieve Contribution and Flexible Providers, Coordinates Contributions and Flexible providers), but also to "catch" the knowledge produced using the means of communication freely chosen by the actors (Aggregate Activity Loop). This result shows once again the efficiency and effectiveness that we can obtain modelling a process using the methodology and collaborative pattern

    ¿Hay espacio para aquello que ralentiza la comunicación, la tensión, la pregunta, el descontento? Algo de esto se pregunta Byung-Chul Han en una reciente entrevista (2020) para Zeit Wissen, revista bimensual alemana de divulgación científica. El filósofo asume la postura de que lo agrietado, las roturas, las lesiones, incluso las costuras quedan canceladas porque, en palabras del filósofo, hoy todo parece redondeado, pulido o suavizado. Se detiene a pensar que, en el mundo del Big Data, nos encontramos entre el ruido y el silencio, silencio-otro, puesto que no refiere a ese silencio elocuente que, al igual que la quietud, también produce lenguaje. En este sentido, Han enfatiza que la actual existencia resulta ser un tanto ruidosa, aunque sin palabras. Habrá información, sostiene, pero no conocimiento, puesto que el saber tiene una estructura temporal que abarca el pasado y el futuro, no sólo el presente. Este problema nos conduciría a la mudez y al desamparo. La crudeza del filósofo será apenas un disparador para reflexionar sobre el deseo y la posterior materialización de este libro, tanto o de igual modo que la escritura y la reflexión que ofrecen las y los autores al modo de actos contra-hegemónicos

    Bioética es un término de origen anglosajón, utilizado por primera vez en 1971 por van Rensselaer Potter; por ello en el 40 aniversario del surgimiento de esta disciplina, se revisó su traspaso de los Estados Unidos de Norteamérica a la América Latina, diferenciando entre moral y ética. Se destaca la labor del profesor José Alberto Mainetti, pionero en esta tarea, así como la importancia de diferentes instituciones, programas y federaciones, entre otras, por su encomiable labor al respecto. Se reseñan las raíces de la bioética en el país y se analizan la formación de sus distintas cátedras, las publicaciones sobre el tema y el apoyo brindado por organizaciones como la Sociedad Cultural "José Martí". El autor, basado en su experiencia, expone cómo ha continuado desarrollándose esta disciplina en Cuba.Bioethics is an Anglo Saxon origin word, used for the first time in 1971 by van Rensselaer Potter; that is why in the 40th anniversary of the emergence of this discipline, its transfer from the United States to Latin America was reviewed, making differences between moral and ethics. Professor José Alberto Mainetti`s work, pioneer in this task is highlighted, as well as the importance of different institutions, programs and federations, among others, for their commendable task on this respect. The roots of bioethics in the country are pointed out, and the formation of their different lectures, the publications on the topic and the support given by organizations such as "José Martí" Cultural Society are analyzed.     Nos limitaremos, en este escrito, a relatar nuestra experiencia personal y la técnica quirúrgica que hemos adoptado. La primera parte de esta cuestión comprende 38 tiroidectomías subtotales ultraradicales, en un solo tiempo, que hemos practicado en enfermas del Servicio de Clínica médica del profesor Rodolfo Rossi o en mi servicio de Cirugía del Hospital Italiano de La Plata desde enero de 1935 a marzo de 1940. El número de tiroidectomías que hemos practicado es mayor, pero aquí solo hemos de referirnos a los casos de bocio tóxico, por e^o hemos tenido cuidado de separar las enfermas con bocio simple, coloide o quístico, un caso de cáncer tiroideo y dos casos de bocio intra torácico. No entra tampoco en esta estadística las enfermas atendidas de su hipertiroidismo, que aunque pocas, no han llegado a la mesa de operaciones por circunstancias diversas.The author has effected 38, subtotal, ultra-radical thyroidectomies in one time for exophthalmic goitre. All sorts of cases of hyperthyroidism have been operated. Simple, serious and complicated cases. (Exophthalmic goitre and pregnancy, exophthalmic goitre and diabetes, exophthalmic goitre and nephrosclerosis, exophthlamic goitre and fibrillation, exophthlamic goitre and asystolia). The operative mortality has been 0 %. The fundamental in the Preoperative treatment has been rest and Lugol's solution. In order to decide the operative moment, it has been taken into account the patient's state of tranquillity, the pulse curve, the basal metabolism and the weight of the body. In the majority of cases this opportune moment has been at the 10 th. or 15 th, day from the time of administration of Lugol's solution. The only anaesthetic employed has been Novocaine 0.50 % without adrenalin, with previous injection of morphine.Facultad de Ciencias Médica

    Administration of aerosolized SARS-CoV-2 to K18-hACE2 mice uncouples respiratory infection from fatal neuroinvasion

    The development of a tractable small animal model faithfully reproducing human COVID-19 pathogenesis would arguably meet a pressing need in biomedical research. Thus far, most investigators have used transgenic mice expressing the human ACE2 in epithelial cells (K18-hACE2 transgenic mice) that are intranasally instilled with a liquid SARS-CoV-2 suspension under deep anesthesia. Unfortunately, this experimental approach results in disproportionate high CNS infection leading to fatal encephalitis, which is rarely observed in humans and severely limits this model’s usefulness. Here, we describe the use of an inhalation tower system that allows exposure of unanesthetized mice to aerosolized virus under controlled conditions. Aerosol exposure of K18-hACE2 transgenic mice to SARS-CoV-2 resulted in robust viral replication in the respiratory tract, anosmia, and airway obstruction, but did not lead to fatal viral neuroinvasion. When compared to intranasal inoculation, aerosol infection resulted in a more pronounced lung pathology including increased immune infiltration, fibrin deposition and a transcriptional signature comparable to that observed in SARS-CoV-2-infected patients. This model may prove useful for studies of viral transmission, disease pathogenesis (including long-term consequences of SARS-CoV-2 infection) and therapeutic interventions

    Opinião e nível de informação sobre a Lei Nacional de Identidade de Gênero e a autonomia progressiva dos adolescentes na escola: uma visão da Bioética

    Purpose/Context: In the context of the current social and legal transformations regarding gender, the objective of the research is to explore and analyze the opinion and level of information that high school students have about the rights stipulated in the National Gender Identity Law No. 26743 and about progressive autonomy, from a bioethical point of view. Methodology/Approach: An exploratory-descriptive, cross-sectional design was used with a mixed approach, with quantitative and qualitative data. As part of this, a purposive sample of 100 students between 17 and 18 years of age, from the cities of Necochea and Mar del Plata (Buenos Aires, Argentina) was formed. The participants answered a survey specifically designed for this research, with open and closed questions, online. Results/Findings: The results showed a low level of information on the Law and the principle of progressive autonomy, with the internet and social networks being the main source of information. Regarding the opinion on the rights stipulated in the Law and the principle of progressive autonomy, interest in this topic and a positive evaluation of its knowledge and teaching were observed. Discussion/Conclusions/Contributions: These results raised questions about the role of the secondary school in the training on these issues and may contribute to improve educational strategies and policies on this relevant social issue. Likewise, the bioethical perspective offers elements for this, by problematizing naturalized cultural constructions on gender, and to think about this issue also in infancy in order to guarantee the best interests of children.Propósito/Contexto: en el contexto de las actuales transformaciones sociales y legales con respecto al género, el objetivo de la investigación es explorar y analizar la opinión y el nivel de información que tienen estudiantes de escuelas secundarias, sobre los derechos estipulados en la Ley Nacional de Identidad de Género n.° 26743 y sobre la autonomía progresiva, desde una mirada bioética. Metodología/Enfoque: se utilizó un diseño exploratorio-descriptivo, transversal con un enfoque mixto, con datos cuantitativos y cualitativos. Como parte de ello se conformó una muestra intencional de 100 estudiantes de entre 17 y 18 años de edad, de las ciudades de Necochea y de Mar del Plata (Buenos Aires, Argentina). Los participantes contestaron una encuesta diseñada específicamente para esta investigación, de preguntas abiertas y cerradas, de manera virtual. Resultados/Hallazgos: los resultados evidenciaron un bajo nivel de información sobre la Ley y el principio de autonomía progresiva, siendo internet y redes sociales su principal fuente de información. Con respecto a la opinión sobre los derechos estipulados en la Ley y el principio de autonomía progresiva, se observó interés por esta temática y una valoración positiva de su conocimiento y enseñanza. Discusión/Conclusiones/Contribuciones: dichos resultados abrieron interrogantes sobre el rol de la escuela secundaria en la formación respecto a estas cuestiones, y pueden contribuir a mejorar estrategias y políticas educativas sobre este relevante tema social. Asimismo, la perspectiva bioética ofrece elementos para ello, al problematizar construcciones culturales naturalizadas sobre el género, y para pensar este tema también en las infancias en pro de garantizar el interés superior de los niños.Objetivo/Contexto: no contexto das atuais transformações sociais e legais em relação ao gênero, o objetivo da pesquisa é explorar e analisar a opinião e o nível de informação que os alunos do ensino médio têm sobre os direitos estipulados na Lei Nacional de Identidade de Gênero nº 26743 e sobre a autonomia progressiva, a partir de uma perspectiva bioética. Metodologia/Abordagem: foi utilizado um projeto exploratório-descritivo e transversal com uma abordagem mista, com dados quantitativos e qualitativos. Para isso, foi formada uma amostra intencional de 100 estudantes entre 17 e 18 anos de idade das cidades de Necochea e Mar del Plata (Buenos Aires, Argentina). Os participantes responderam on-line a um questionário criado especificamente para esta pesquisa, com perguntas abertas e fechadas. Resultados/Descobertas: Os resultados mostraram um baixo nível de informação sobre a Lei e o princípio da autonomia progressiva, sendo a internet e as redes sociais sua principal fonte de informação. Com relação à opinião sobre os direitos estipulados na Lei e o princípio da autonomia progressiva, observou-se interesse por esse tópico e uma avaliação positiva de seu conhecimento e ensino. Discussão/Conclusões/Contribuições: esses resultados abriram questionamentos sobre o papel das escolas de ensino médio na formação sobre esses temas e podem contribuir para o aprimoramento das estratégias e políticas educacionais sobre essa relevante questão social. Da mesma forma, a perspectiva bioética oferece elementos para isso, problematizando as construções culturais naturalizadas sobre o gênero, e para pensar sobre essa questão também na infância, a fim de garantir os melhores interesses das crianças
