    Hematopoietic Stem Cell Transplantation in Thalassemia

    Although recent advances in gene therapy are expected to increase the chance of disease cure in thalassemia major, at present hematopoietic stem cell transplantation (HSCT) remains the only consolidated curative approach for this disorder. The widest experience has been obtained in the HLA-matched family donor (MFD) setting, with probabilities of overall and thalassemia-free survival exceeding 90% and 85%, respectively. As for most patients a suitable MFD is not available, alternative donors (HLA-matched unrelated donor, unrelated cord blood, HLA-haploidentical relative) have been increasingly explored, translating into the expansion of the number of patients treatable with HSCT

    PERCHLORATES AS POWERFUL CATALYSTS IN MANY IMPORTANT ORGANIC TRANSFORMATIONS

    For long times, metallic perchlorates have been considered dangerous compounds[1] in that they function as explosives and as incontrollable oxidizers. Therefore, the fear of the great hazard connected with their manufacture and uses had prevented an extensive use both in research laboratories and in industrial processes.[2] However, recently it has been cleared that this bad reputation is due to the mistaken association of metallic perchlorates with the oxidizing potential of perchloric acid and the pyrotechnic performances of NH4ClO4. In this conference, we report that Magnesium and Zinc perchlorates can be used as powerful Lewis acids in several organic transformations which are commonly employed both in laboratory and industrial processes. As depicted in the Scheme, perchlorates are able to promote a series of reactions such as: a) Acylation of alcohols[3,4] b) Fisher esterification[5] c) Synthesis of b–enamino esters[6] d) Protection of the amino group as N-Boc derivative[7] Moreover, they can act as specific catalysts for the development of a new organic transformations. In fact, the treatment of an alcohol or a phenol with Boc2O in the presence of Mg(ClO4)2 leads to the unexpected formation of the corresponding t-butyl ethers.[8] This represents the first general method to synthesize aromatic t-butyl ethers. In an analogous manner, the treatment of phenols with diethyl dicarbonate results in a very efficient method to produce difficulty available aryl ethyl carbonates. Several of the above mentioned transformations can be carried out in the absence of the solvent, and often the catalyst can be easily recovered and reused without an appreciable loss of activity.     PHARMACOKINETICS OF GEMCITABINE HYDROCHLORIDE AND IRINOTECAN HYDROCHLORIDE ALONE AND IN COMBINATION IN RAT PLASMA BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY-DIODE ARRAY DETECTOR

    This communication reports an easy and quick HPLC-PDA method for the simultaneous analysis of Irinotecan Hydrochloride and Gemcitabine Hydrochloride in rat plasma samples both after single drug administration and drugs association. Gemcitabine Hydrochloride is commonly administered to treat non-small cell lung cancer (NSCLC), pancreatic adenocarcinoma, and in combination with paclitaxel for the treatment of breast cancer in the metastatic phase. Additionally Irinotecan Hydrochloride was used to treat colorectal cancer (CRC), gynecological cancers, carcinomas, non-small cell and small cell lung cancer. The drugs were detected simultaneously by using a Zorbax Extend C-18 column (250 mm × 4.6 mm; 5 m particle size) in gradient elution mode. The chromatographic analysis was performed in 15 minutes. The analytical method was calibrated and validated from 0.1 to 18 g/mL for both drugs. Rat plasma was used as biological samples during the analysis; while the 3-methylxanthine was used as internal standard. The performance of analytical method was further tested in rat plasma samples collected after single dose administration of drug or their association. Results demonstrated that HPLC-PDA method allows to detect the drugs in the range of concentrations herein reported and the analytical method is accurate and selective. The limit of quantification of the method was 0.1 g/mL. These values are similar or little higher to data published in literature, which are performed using sophisticated and expensive detectors such as mass spectrometer, and wich consider merely only one drug and not their association. The weighted-matrix matched standard curves showed a good linearity until 18 g/mL. The parallelism tests were also performed to evaluate if over-range samples can be analyzed after dilution, without affecting the performance of validated method. The intra- and inter-day precision (RSD%) values were ≤7.14% and ≤11.5%, respectively, for the full range of analysis. The intra- and inter-day trueness (Bias%) values ranged from -11.5% to 1.70% for the two drugs. At the best of our knowledge, this is the first HPLC-PDA method which allows to detect simultaneously Irinotecan Hydrochloride and Gemcitabine Hydrochloride in rat plasma, both after single and drugs association administration in order to evaluate how can interact and modify the pharmacokinetic parameters.
