Basic concepts of Magnetic Resonance Imaging
Fundamental aspects of the Magnetic Resonance Imaging (MRI) technique which is one of the preferred modalities for non-invasive clinical applications and "in vivo" medical and biological research, are presented. The physical principles of MRI and protocols recently introduced to investigate the microscopic details of pathological cerebral damages are introduced. The main results obtained from the analysis of T2 relaxation curves are briefly summarized

In recent years, several papers in the literature considered the Importance of Iron and other transition metal based compounds not onlythe preparation of magnetic nanostructured contrast media for MRI, SQUID and biosenslng detection, but alsothe manufacturing of labelled compounds able to track cells and/or tissues in vivo and ex vivo.this way objects for targeting specific tissues can be obtained, pushing the diagnosis capability to the molecular level.the last fifteen years, this diagnostics ability has been joint to the possibility of using the magnetic nanoparticles for delivering drugs and for releasing heat locally, thus allowing to obtain theranostlc agents for the realization of the find-fight and follow approachthe treatment of tumour and of other diseases

Sono stati studiati nuovi composti da utilizzare come agenti di contrasto
(CA) per immagini di Risonanza Magnetica. Lo scopo del lavoro era di trovare sistemi
con rilassivita’ paragonabile o migliore di quella dei CA esistenti in commercio ma con
dimensioni e forma uniforme, e tali da essere utilizzati per “targeting” molecolare. La
prima classe di composti è costituita da un nucleo magnetico rivestito da Polietilenimmina
(PEI) o da Polietilenimmina carbossilata (PEI-COOH). La seconda classe di
composti è costituita da nanocristalli magnetici colloidali (NCs) costituiti da un nucleo
di magnetite rivestito da materiale organico e aventi forma e dimensioni controllabili.
Mediante misure di profili di dispersione NMR si dimostra che entrambi i sistemi possono
essere utilizzati come mezzi di contrasto con rilassività paragonabile o superiore a
quella degli agenti di contrasto attualmente in uso

Inhibition of lysozyme fibrillogenesis by hydroxytyrosol and dopamine : an Atomic Force Microscopy study
Protein aggregation underlies many human diseases characterized by the deposition of normally soluble proteins in both fibrillar and amorphous aggregates. Here, Atomic Force Microscopy (AFM) has been applied to investigate the ability to inhibit hen egg white lysozyme (HEWL) fibrillogenesis by hydroxytyrosol (HT), one of the main phenolic components of olive oil. In this framework, HEWL is a useful and well-studied model protein whose amyloid-like fibril formation can be induced under experimental conditions where HT is more stable. HEWL fibrils, obtained at pH 1.6 and at 65 °C, exhibited a height of about 3 nm and a fibril length on average of about 3 μm. The presence of HT reduced the HEWL fibril number and length with respect to the control sample. Interestingly, also dopamine, a compound with a chemical structure similar to HT, decreased both the fibril number and the fibril length. AFM experimental data were supported by Thioflavin T assay and Fourier transform infrared spectroscopy. Our results show that HT is an effective inhibitor of HEWL aggregation, thus suggesting possible future applications of this natural compound for potential prevention or treatment of amyloid diseases, or as a lead molecular structure for the design of improved modulators
