    Cyclic nucleotide-gated (CNG) ion channels are Ca2+ permeable nonselective cation channels that are directly gated by binding of cAMP or cGMP, thus providing a linkage between two important signal molecules, cyclic nucleotides and calcium. They are known to play an important role in sensory transduction and in second-messenger modulation of synaptic neurotransmitter release. Previous studies showed that besides in neuronal cells, CNG were found also in non-neuronal tissues including heart, kidney, blood vessels and spleen, they are reported to be involved in a variety of cell function.Ion channels play an indispensable role in endothelial cells, which is a unique signal-transducting surface in the vascular system that is responsible in altering vascular tone. The present study investigated the expression and functional roles of the cyclic nucleotide gated channels (CNG) in regulating the intracellular calcium level of vascular endothelial cells using molecular and calcium measurement techniques.The present study provided evidence that the CNG channels, especially that of CNGA2, were expressed in vascular tissues. I used a variety of different methods, including RT-PCR, northern blot, in-situ hybridization, immunohistochemistry and western blot to study the localization of CNGA2 channels. RT-PCR amplify a CNGA2 fragment of 582bp from RNAs isolated from bovine vascular endothelial cell line, rat vascular smooth muscle cell line and rat aorta. Northern blot analysis detected a 2.3-kilobase (kb) CNGA2 transcript in rat aorta mRNA. The cellular distribution of CNGA2 was further studied by in-situ hybridization, which demonstrated expression of CNGA2 mRNA in human vascular endothelial and vascular smooth muscle cells. Immunohistochemistry data also agreed with those generated from in-situ hybridization. Western blot data also demonstrated proteins of CNG2 was expressed in both human vascular endothelial cells and vascular smooth cells layer. Subcellular localization of CNGA2 inside the vascular endothelial cells was also investigated with the use of a GFP linked CNGA2 channel gene. Taken together, the results showed that CNGA2 proteins were expressed on the plasma membrane of the vascular endothelial cells.     Scheduling shared data acquisition for real-time decision making

    This work investigates scheduling policies for the acquisition of possibly overlapping sets of data items required to make multiple decisions by different deadlines. The work is motivated by military IoT applications in which a large number of sensors must collect intelligence data needed to make multiple decisions. For example, data from several cameras in a contested city might be needed to decide where targets of interest are. This work is based on the assumption that network bandwidth is limited, creating a significant resource bottleneck (perhaps between the sensors and the command center where decisions are made). This might be the case, for example, due to active interference by a determined adversary. A relieved sub-problem is first discussed with a corresponding optimal algorithm. Then, an improved heuristic algorithm based on the insights from the optimal algorithm of the sub-problem is presented.     M.F.A.In the sense of an artistic interpretation, the role of the human body can act as a form, dependent object, container, or movable sculpture. As physical roles and physical symbols transmit the inner thoughts and emotions of a body, a person’s state of mind can be outwardly expressed, and communication takes place. This paper will be an analysis of the author’s artistic attempts to record a person’s inner emotional state when that is expressed through a certain visual medium—the human body.In further detail, this text documents the journey of the author over two years’ time as she uses artistic mediums to: 1) encourage the expression of an art viewer’s inner emotional state; 2) capture that expression as a tangible record; 3) analyse the art viewer’s unique responses; 4) incorporate those responses into the author’s other artistic works as part of an ongoing process of inspired exploration, particularly the author’s final exhibition piece as part of the Master of Fine Arts programme at the Chinese University of Hong Kong.Further broken down into the following five research questions, the thesis takes on a particular philosophical direction:How can a person’s body operate as a communicative tool?How can a person’s inner world be explored via a body, in the context of time and space?Can body traces become visualisations for our experiences in life?How can body traces be applied in sculpture and installation practices?How does bodily engagement of the audience transform to become part of an artwork?Some of the major challenges of documenting a person’s mood or mental state at a particular time, and the effect that mood has on the person's thinking and behavior, include but are not limited to: 1) there are many barriers to expression of a state of mind, such as reluctance towards disclosure or the person’s own acuity of awareness of his or her feelings; 2) a state of mind is a temporary psychological state, meaning that it is fairly fragile and subject to rapid change; 3) it is challenging to find a suitable method for documenting in concrete terms what is essentially an abstract thing (such as emotions and thought).This thesis also explains the author’s period of works, from 2012 to 2019. The theme of the art works was a gradual shift from being a reflection on the nature of the body—which took place in the form of body-related objects: 1) the casting of specific body parts, 2) installations with body-related objects, using materials where the medium’s shape would emphasise the artist’s message—to culminating in a markedly different final exhibition for the Master of Fine Arts course. In that exhibition, the focus was on a self-reflected written expression of the viewer’s inner state of mind, an abstraction that was then translated into a tangible format and incorporated into part of the art installation itself.The collection and analysis of research assets mainly include photographic records of the viewer responses collected during that same exhibition (the exhibition having taken place from May to June 2019 at the Chinese University of Hong Kong).Beyond collecting and analysing the responses of art audiences, the scope of the author’s thesis also looks at the works of other artists, in the hope that the author might build upon the outcome of previous artists’ works to consider alternative perspectives and methodologies.Lastly, this paper draws conjectures from the author’s observations and analyses, and compares what significance those musings may have anthropologically and philosophically.我們身體跟內心的思緒最大的分別在於身體是與身俱來的,而思緒則是可以不斷地改變,有如陶土輪上的粘土可以一直加添塑造各種形狀。在這短暫的一生裡,我們的身體會否留下什麼痕跡?除了認知的用途外,還有什麼方面的探知在等待著我們的身體?從藝術的角度去探知身體的角色,我們先要尋問身體可以作為什麼?如果先把認識的身體拋開,假定身體的可變;我們發現身體可以以形態出現、可以稱作物件依存、可以是一個載體、也可以是一件流動的雕塑。如何發揮並善用這些身體角色去呈現內在的思想和情感正是創作方向的重心。本文想探討的卻不只限於此;觀乎身體經過年月的洗禮,身上總會留下一些印記,這些痕跡可以看作是身體上的符號,因著人類在生命裡遇過相似的經驗,我們會產生共識去理解其含意和歷史。本文將論證身體在這些方面的轉變,並將時間和情感附注其中以作為研究的核心。本論文紀錄了作者在中文大學兩年的專題藝術研究旅程; 1) 推動觀眾在觀賞展出作品時抒發其內在情感;2) 並將該情感視為有形紀錄; 3) 繼而分析觀眾對作品所產生的各種獨有思緒反應; 4) 將這些回應灌注到作者的藝術裝置裡,作為一種持續的啟發和探究,提出觀眾在作品裡的演譯和影響。文章討論將從身體的認知到如何引用身體去創作為結尾,為了清楚明辯,將分成五點論述;- 人的身體如何作為一種溝通的工具?- 如何運用身體通過時間和空間轉變下去探知人的內在世界?- 身體痕跡能否體現人的生活經歷?- 如何運用身體痕跡呈現在雕塑和裝置藝術創作上?- 如何透過觀眾身體的互動和參與將之成為作品一部份?整個創作和研究過程中最具挑戰的是紀錄人在特定時間裡的情緒和精神狀態,而該狀態的變化是如何透過思想和行為顯露人前。其中包括但不限於:1) 在表達情感時會遇上許多心理上的障礙,例如不願意披露給別人知曉或是意識到自己的感受而作出保護;2) 精神狀態能隨時間、情感或事件而變,意味著它既相當脆弱,亦容易發生難以預料的變化;3)尋找合適的方法去紀錄本質上是抽象的議題-思緒和行為。文中同時概論了作者從2012年至2019年間的創作歷程和轉化。由最初以身體形態為主導的表現手法 (例如,通過身體倒模而取材或是以關乎身體等物件去創作的作品);到近年以探討內在情感與身體表現的關係和衝擊為重。作者通過討論其他藝術家在處理有關身體的題材上之分野,去進行反思和突破,從而在尋找抒發內在情感的手法上得以推進;在中文大學的畢業作品裡,能看出作者新的演譯手法,把觀眾在作品裡的角色轉化為參與者,讓作品變得更完整性、層次更豐富和獨有。最後,考慮到身體在空間領域和展覽廳中可成為作品的一部份,當觀眾走過裝置作品時,他們有機會與之產生互動。每個個體對作品的反應將成為他們對身體和空間相互動態下的反思,以此回應論文的主題:身體來呈現精神狀態。Tai, On Yau Monica.Thesis M.F.A.
