    Applications and Utility of Three-Dimensional In Vitro Cell Culture for Therapeutics

    The field of 3D cell culture and its applications is rooted in the understanding of cell biology, tissue engineering, tissue morphology, disease mechanisms, and drug action. For many years, traditional 2D cell culture systems have been widely used but have proven to be limited in their ability to accurately replicate the complex microenvironment of tissues. This often results in issues with cell proliferation, aggregation, and differentiation. 3D cell culture systems have emerged as a solution to this problem and have demonstrated a more accurate simulation of in vivo physiology. This has had a major impact on drug discovery and includes the use of spheroids, organoids, scaffolds, hydrogels, and organs. This review has addressed fundamental questions and exploited utility in 3D in vitro mode of cell culture in view of therapeutics

    Testing of traditional ways of predicting lactation yield for selecting dairy cattle

    514-517There are many conventional methods for predicting the first lactation milk yield in 305 days (FLMY) based on fortnightly part milk yield data in dairy cattle. The different methods used are based on data available over different years as implicated under farm and field conditions. These fortnightly test day milk yields could be utilized in the estimation of 305-day milk yield by ratio method and multiple regression methods. The Sahiwal cattle can be selected for higher FLMY utilizing fifth fortnightly day milk yield (FTDY-5), FTDY-6, FTDY-7, FTDY-8, FTDY-11, and FTDY-18 test day yields. Selection can be carried out for higher first lactation milk yield based on fortnightly test day milk yields in the case of Sahiwal and its crossbred cattle

    Surgical management of pterygium in a rural hospital using bare sclera versus conjunctival autograft technique

    Background: Pterygium is defined as a triangular fibrovascular subepithelial ingrowth of degenerative bulbar conjunctival tissue over limbus onto the cornea. Commonly seen among farmers, fishermen, stone cutters and welders. Both blue and ultraviolet light have been implicated in its causation. Histologically the true Pterygium shows elastotic degeneration of the subconjunctival connective tissue. Pterygium warrant treatment when they encroach the visual axis, induces significant astigmatism or become cosmetically bothersome. My present study was undertaken to evaluate the efficacy, safety and recurrence rates of conjunctival autograft and bare sclera techniques in treatment of progressive true pterygium in a rural hospital.Methods: The prospective study was done in the Department of Ophthalmology, Adichunchanagiri Institute of Medical Sciences, B.G Nagara, Mandya District, Karnataka, India. The study was conducted for a period of 18 months. About 70 patients who presented with progressive pterygium were included in the study.Results: In our study pterygium recurred in 10 cases (14.28%), among which pterygium recurred in 8 cases (22.85 %) using bare sclera technique and in 2 patients (5.71%) using conjunctival autograft technique, best corrected visual acuity remained stable in 37 cases (52.85 %), improved in 33 cases (47.14 %) and worsened in none (0%) of the cases.Conclusions: Conjunctival autografting is a safe and effective procedure in the management of pterygium. The recurrence rate following limbal conjunctival autografting is significantly lower than that following primary conjunctival closure by bare sclera technique.</jats:p

    Revisiting the Existing Grading Systems of Glenohumeral Joint Arthropathy and Proposing a Modified Classification

    Osteoarthritis (OA) of the glenohumeral joint may affect up to 20% of the elderly population. The glenohumeral joint OA is classified in several ways based on different criteria. The notable ones include the Samilson and Prieto classification, Modified Samilson and Prieto classification by Allain, Modified Samilson and Prieto classification by Gerber, Guyette classification, and Kellgren and Lawrence classification. Through this letter, the authors wish to bring it to esteemed editors that a more comprehensive radiological classification is needed to study and categorize the patient and to predict the severity of glenohumeral arthritis. We suggest that X-ray views in anatomical position as well as 45° external rotation and 45° internal rotation will be best suited to plain radiographic classification, which is needed to classify glenohumeral arthritis

    Identification, expression analysis and molecular modeling of <i>Iron deficiency specific clone 3</i> (<i>Ids3</i>) like gene in hexaploid wheat

    AbstractGraminaceous plants secrete iron (Fe) chelators called mugineic acid family phytosiderophores (MAs) from their roots for solubilisation and mobilization of unavailable ferric (Fe3+) ions from the soil. The hydroxylated forms of these phytosiderophores have been found more efficient in chelation and subsequent uptake of minerals from soil which are available in very small quantities. The genes responsible for hydroxylation of phytosiderophores have been recognized as iron deficiency-specific clone 2 (Ids2) and iron deficiency-specific clone 3 (Ids3) in barley but their presence is not reported earlier in hexaploid wheat. Hence, the present investigation was done with the aim:(i) to search for the putative Hordeum vulgare Ids3 (HvIds3) ortholog in hexaploid wheat, (ii) physical mapping of HvIds3 ortholog on wheat chromosome using cytogenetic stocks developed in the background of wheat cultivar Chinese Spring and (iii) to analyze the effect of iron starvation on the expression pattern of this ortholog at transcription level. In the present investigation, a putative ortholog of HvIds3 gene was identified in hexaploid wheat using different bioinformatics tools. Further, protein structure of TaIDS3 was modelled using homology modeling and also evaluated modelled structure behavior on nanoseconds using molecular dynamics based approach. Additionally, the ProFunc results also predict the functional similarity between the proteins of HvIds3 and its wheat ortholog (TaIds3). The physical mapping study with the use of cytogenetic stocks confines TaIds3 in the telomeric region of chromosome 7AS which supports the results obtained by bioinformatics analysis. The relative expression analysis of TaIds3 indicated that the detectable expression of TaIds3 induces after 5th day of Fe-starvation and increases gradually up to 15th day and thereafter decreases till 35th day of Fe-starvation. This reflects that Fe deficiency directly regulates the induction of TaIds3 in the roots of hexaploid wheat.</jats:p

    Adaptive ribosomal DNA polymorphism in wild barley at a mosaic microsite, Newe Ya'ar in Israel

    In eukaryotic genomes, ribosomal DNA is organized in tandem repeat units at the nucleolar organizing regions (NORs) on each satellited chromosome. Each repeat unit consists of a coding region and an intergenic spacer (IGS), the latter generally varying in length at different loci of the same genotype and also at the same locus in different genotypes. The spacer length variants (slvs) are sometimes correlated with climatic and ecological variables. In a study of 42 accessions of wild barley (Hordeum spontaneum) collected from four different microniches (sun-deep soil, sun-shallow soil, shade-deep soil and shade-shallow soil) at the Newe Ya'ar microsite (3182 m2), in Israel, we observed eight slvs constituting 10 different rDNA phenotypes, correlated with edaphic conditions prevalent at the four microniches. We conclude that rDNA diversity in wild barley is distributed nonrandomly and adaptively in the four microniches of Newe Ya'ar. A novel feature in this study is also the homogenization of IGS length at the two loci located at two nonhomologous chromosomes.     Over-Indebtedness in Nepalese Microfinance Institution – A Study of Clients of Nirdhan Utthan Bank

    This paper analyses the over-indebtedness of micro-borrowers of Nirdhan Utthan Bank in Birgunj, Nepal. Descriptive analytical research design is attempted to explore the scenario of factor of over-indebtedness and impact of multiple borrowing. The research is primary data based on 150 sample respondents from Birgunj and convenient and structured questionnaire had been used to collect response of the respondents and data was collected through face to face interview with the clients. In this study majority of the clients are involved in multiple borrowing 82 %, it finds that loss of income and multiple borrowing is the major factor that leads to over-indebted. The paper also shows the association between factors of over-indebtedness and multiple borrowing and how frequently micro-borrower repeats them and for this cross tabulation analysis and chi-square test was used to analyze the data. The results show that over-indebtedness is significantly associated with multiple borrowing. The risk of over-indebtedness further increases with the occurrence of multiple borrowing. Results show that small size of loans issued by MFIs and needs of the clients was one of the major reasons for multiple loans contracts. Other reasons for multiple borrowing include family obligation, loan recycling, and relaxed or simple loan procedures, and influence from friends who are taking multiple loans, delayed loan disbursement from the MFIs in which the client first applied.</jats:p
