Bielefeld (Germany): Bielefeld University; 2007.Advances in sequencing technologies provide the opportunity to rapidly produce vast genomic data sets at a low price. In particular, the recently developed, ultra-fast 454 pyrosequencing has dramatically reduced the cost and time requirements per sequenced base pair. Furthermore, novel methods have been developed that enable sequencing of the 99 percent of microbes that are difficult to access with conventional, culture-dependent approaches. These culture-independent methods launch the exciting field of metagenomics - the study of the collective genomes (metagenomes) of free-living microbial communities.
In light of the immense data sets produced, sequence analysis is still a contemporary, ongoing challenge in computational biology. Additionally, new demands arise from the short length of sequence reads produced by ultra-fast sequencing techniques. In the field of whole-genome research, accurate methods are required for identifying and functionally characterizing the gene content of organisms, thus reducing the required manual effort while at the same time producing high-quality annotations. On the other hand, metagenomes are nowadays routinely sequenced, and an increasing number of metagenomic projects is expected in the near future (Pennisi, 2007), but their computational analysis is still in its infancy.
In the presented thesis, state-of-the-art machine learning techniques as well as algorithmic and statistical methods are employed for the high-throughput analysis and characterization of large genomic data sets. First, the gene finding software GISMO was developed, which combines the search for protein domains using profile hidden Markov models (pHMMs) with a sequence composition-based classification using a Support Vector Machine. This combined strategy is able to unveil almost the complete gene content of prokaryotic genomes in a fully automated manner. GISMO has already been extensively employed in the international effort to "Annotate a Thousand Genomes" as well as in various genome annotation and re-annotation projects.
Furthermore, a novel gene finding algorithm for metagenomic data sets was developed. It is robust for most problems encountered when predicting genes in metagenomes, including short sequence length and low sequence quality. Thereby, the algorithm allows to hunt for novel, unknown genes carried by organisms that cannot be sequenced using conventional, culture dependent techniques.
Finally, methods were devised for characterizing short-read metagenomes obtained by pyrosequencing. Following the pHMM-based identification of gene fragments, the latter are categorized into functional groups. Additionally, the source organisms (taxonomic origins) of gene fragments are predicted. The resulting genetic and taxonomic profiles can in turn be used to unveil important trends in the gene content, metabolism, and species composition of the underlying microbial communities

Dohromady bylo zkoumáno šest dílčích období voleb, a celkový vzorek epizod Show Jana Krause je tím pádem tvořen šesti menšími vzorky. Cílem práce bylo zjistit, jakou podobu politická satira v pořadu ve sledované době měla, jak často se v něm objevovala a kteří političtí aktéři byli nejčastěji jejím terčem. Výzkumnou metodou je kvantitativní obsahová analýza s jejíž pomocí hledá práce odpovědi na následující čtyři výzkumné otázky. VO1: Jak velký prostor v Show Jana Krause byl politické satiře věnován?; VO2: Jakých prvků politické satiry Show Jana Krause využívá?; VO3: Jací politici, respektive jaké politické subjekty, byly v pořadu nejčastěji satiricky zobrazovány?; VO4: Jakým tématům se politická satira v Show Jana Krause věnovala nejvíce? Práce se skládá ze čtyř kapitol. První kapitola teoreticky vymezuje klíčové pojmy a nastiňuje historii politické satiry. Ve druhé kapitole je podrobněji rozebrán žánr televizní politické satiry a jeho vývoj zejména se zaměřením na české prostředí. Předmětem třetí kapitoly je představení samotné Show Jana Krause jakožto české verze late-night show. Práce se zabývá okolnostmi vzniku tohoto pořadu, jeho podobou i...The diploma thesis deals with political satire in the Show Jana Krause at the time of selected elections in the Czech Republic in the years 2010-2019. Altogether six sub-periods in which the chosen elections were held were examined, which means that the general sample of episodes of the Show Jana Krause is formed from six smaller samples. The aim of the work was to find out what form political satire had in the show in the monitored period, how often it appeared in it and which political actors were most often its target. The research method was a quantitative content analysis with the help of which the work seeks answers to the following four research questions. RQ1: How much space was devoted to political satire in the Show Jana Krause? RQ2: What elements of political satire does the Show Jana Krause use? RQ3: What politicians, or which political subjects were most often satirically portrayed on the show? RQ4: What topics did political satire on the Show Jana Krause focus on the most? The thesis consists of four chapters. The first chapter theoretically defines key concepts and outlines the history of political satire. V práci se využívá teoretických poznatků z oblasti strategického podnikového řízení a praktických zkušeností autora, které nasbíral během studia, zaměstnání v softwarové firmě a vlastního podnikání. Cílem práce je sestavení podnikatelského plánu minimálně do roku 2016. V teoretické části se autor zaměřuje na porovnání různých definic podnikání, strategického podnikového řízení a založení startupu. In the theoretical part, author focuses on the comparison of various definitions of entrepreneurship, strategic corporate management and startup establishment. The beginning of the practical part describes company and its products. Next chapter forms definition of the market category, market and strategic analysis.
