    Etablierung genetischer Methoden für Clavibacter michiganensis subsp. michiganensis und Charakterisierung von Mutanten mit veränderter Morphologie, Physiologie und Virulenz

Bielefeld (Germany): Bielefeld University; 2003.Die vorliegende Arbeit untergliedert sich in zwei Teile. Ein Schwerpunkt lag in der Entwicklung und Etablierung von molekulargenetischen Techniken für Clavibacter michiganensis subsp. michiganensis (Cmm), dem Erreger der "Bakteriellen Welkekrankheit" der Kulturtomate (Lycopersicon esculentum Mill.). Der zweite Schwerpunkt lag in der Identifizierung und Charakterisierung von Mutanten von Cmm, insbesondere mit veränderter Virulenz, die mit den neu etablierten Techniken erzeugt worden waren. Als eine essentielle Voraussetzung für viele molekulargenetische Techniken, die eine hohe Transformationseffizienz mit heterologer DNA in Cmm benötigen, wurde die Elektrotransformation nach Cmm u.a. mit Hilfe einer Glycinbehandlung und dem Einsatz von nicht methylierter DNA optimiert. Mit einer Erhöhung der Transformationsrate um etwa den Faktor 1000 auf ~ 5 · 10 6 Transformanten/µg DNA konnte die Voraussetzung für die Transposon-Mutagenese, zur Erzeugung von zufälligen Mutationen im Chromosom von Cmm, und die Kassettenmutagenese mittels "gene-disruption"/"gene-replacement" geschaffen werden. Mit dem Transposon Tn1409C - abstammend von IS1409 aus Arthrobacter sp. (GARTEMANN und EICHENLAUB, 2001) - konnte ein Transposon-Mutagenese-System in Cmm und anderen Clavibacter michiganensis-Subspezies etabliert werden, bei dem bis zu 3.000 Transposanten/µg DNA erhalten werden können. Ausgehend von einer pigmentfreien Mutante wurde ein Operon identifiziert, welches die gesamte Carotinoid-Biosynthese von Cmm bis zu dem potentiellen C50-Carotin kodiert. In einer Mutante mit verändertem EPS konnte ein Gen identifiziert werden, das für ein sehr großes, wahrscheinlich zellwandassoziiertes Protein kodiert und Ähnlichkeiten zu Rhs-Proteinen aufweist, dessen Funktion aber noch unklar ist. Unter den ca. 1500 in planta getesteten Transposon-Mutanten von Cmm konnten mit Hilfe eines verfeinerten Welkeindices drei Mutanten identifiziert und charakterisiert werden, die eine deutlich verringerte Virulenz besaßen. In zwei von ihnen war das Transposon in die gleiche DNA-Region inseriert und hatte dort im Abstand von ca. 3,5 kb zwei ORFs unterbrochen, deren Genprodukte Ähnlichkeiten zu Proteinen unbekannter Funktion aus den pflanzenpathogenen Bakterien Xylella fastidiosa und Xanthomonas axonopodis pv. citri aufweisen. Bei diesen könnte es sich um sekretierte Serin-Proteasen handeln. Hierin zeigen sie Ähnlichkeiten zu den sechs ebenfalls in dieser Großregion liegenden Homologen zu dem Pathogenitätsprotein Pat-1 des endogenen Plasmids pCM2. Diese auf einem ca. 130 kb großen VspI-Fragment liegende DNA-Region ist zumindest zum Teil in der dritten Mutante deletiert, die die Wirtspflanze Tomate nicht mehr erfolgreich kolonisieren kann. Alles deutet darauf hin, dass mit dieser DNA-Region ein für die Bakterien-Pflanzen-Interaktion wichtiger Bereich gefunden wurde, in dem erstmals chromosomale Gene identifiziert werden konnten, die zusätzlich zu den bekannten plasmidkodierten Genen pat-1 und celA für die Virulenz von Cmm notwendig sind

    National Security Cultures: Patterns of Global Governance

    This edited collection examines changes in national security culture in the wake of international events that have threatened regional or global order, and analyses the effects of these divergent responses on international security. Tracing the links between national security cultures and preferred forms of security governance the work provides a systematic account of perceived security threats and the preferred methods of response with individual chapters on Canada, China, France, Germany, Italy, Japan, Mexico, Russia, UK and USA. Each chapter is written to a common template exploring the role of national security cultures in shaping national responses to the four domains of security governance: prevention, assurance, protection and compellence. The volume provides an analytically coherent framework evaluating whether cooperation in security governance is likely to increase among major states, and if so, the extent to which this will follow either regional or global arrangements. By combining a theoretical framework with strong comparative case studies this volume contributes to the ongoing reconceptualization of security and definition of threat and provides a basis for reaching tentative conclusions about the prospects for global and regional security governance in the early 21st century.     Real-World Predictions from Ab Initio Molecular Dynamics Simulations

    In this review we present the techniques of ab initio molecular dynamics simulation improved to its current stage where the analysis of existing processes and the prediction of further chemical features and real-world processes are feasible. For this reason we describe the relevant developments in ab initio molecular dynamics leading to this stage. Among them, parallel implementations, different basis set functions, density functionals, and van der Waals corrections are reported. The chemical features accessible through AIMD are discussed. These are IR, NMR, as well as EXAFS spectra, sampling methods like metadynamics and others, Wannier functions, dipole moments of molecules in condensed phase, and many other properties. Electrochemical reactions investigated by ab initio molecular dynamics methods in solution, on surfaces as well as complex interfaces, are also presented

    Absence of the lactase-persistence-associated allele in early Neolithic Europeans

    Lactase persistence (LP), the dominant Mendelian trait conferring the ability to digest the milk sugar lactose in adults, has risen to high frequency in central and northern Europeans in the last 20,000 years. This trait is likely to have conferred a selective advantage in individuals who consume appreciable amounts of unfermented milk. Some have argued for the ‘‘culture-historical hypothesis,’’whereby LP alleles were rare until the advent of dairying early in the Neolithic but then rose rapidly in frequency under natural selection. Others favor the ‘‘reverse cause hypothesis,’’ whereby dairying was adopted in populations with preadaptive high LP allele frequencies. Analysis based on the conservation of lactase gene haplotypes indicates a recent origin and high selection coefficients for LP, although it has not been possible to say whether early Neolithic European populations were lactase persistent at appreciable frequencies. We developed a stepwise strategy for obtaining reliable nuclear ancient DNA from ancient skeletons, based on (i) the selection of skeletons from archaeological sites that showed excellent biomolecular preservation, (ii) obtaining highly reproducible human mitochondrial DNA sequences, and (iii) reliable short tandem repeat (STR) genotypes from the same specimens. By applying this experimental strategy, we have obtained high-confidence LP-associated genotypes from eight Neolithic and one Mesolithic human remains, using a range of strict criteria for ancient DNA work. We did not observe the allele most commonly associated with LP in Europeans, thus providing evidence for the culture-historical hypothesis, and indicating that LP was rare in early European farmers.J.
