Copyright @ 2011 Li et al.BACKGROUND: Understanding of how transcription factors are involved in lymphocyte development still remains a challenge. It has been shown that Egr-2 deficiency results in impaired NKT cell development and defective positive selection of T cells. Here we investigated the development of T, B and NKT cells in Egr-2 transgenic mice and the roles in the regulation of distinct stages of B and T cell development. METHODS AND FINDINGS: The expression of Egr1, 2 and 3 were analysed at different stages of T and B cell development by RT-PCT and results showed that the expression was strictly regulated at different stages. Forced expression of Egr-2 in CD2+ lymphocytes resulted in a severe reduction of CD4+CD8+ (DP) cells in thymus and pro-B cells in bone marrow, which was associated with reduced expression of Notch1 in ISP thymocytes and Pax5 in pro-B cells, suggesting that retraction of Egr-2 at the ISP and pro-B cell stages is important for the activation of lineage differentiation programs. In contrast to reduction of DP and pro-B cells, Egr-2 enhanced the maturation of DP cells into single positive (SP) T and NKT cells in thymus, and immature B cells into mature B cells in bone marrow. CONCLUSIONS: Our results demonstrate that Egr-2 expressed in restricted stages of lymphocyte development plays a dynamic, but similar role for the development of T, NKT and B cells.This article is provided by the Brunel Open Access publishing fund

    Solving factored MDPs using non-homogeneous partitions

    AbstractWe present an algorithm for aggregating states in solving large MDPs (represented as factored MDPs) using search by successive refinement in the space of non-homogeneous partitions. Homogeneity is defined in terms of stochastic bisimulation and reward equivalence within blocks of a partition. Since homogeneous partitions that define equivalent reduced-state-space MDPs can have a large number of blocks, we relax the requirement of homogeneity. The algorithm constructs approximate aggregate MDPs from non-homogeneous partitions, solves the aggregate MDPs exactly, and then uses the resulting value functions as part of a heuristic in refining the current best non-homogeneous partition. We outline the theory motivating the use of this heuristic and present empirical results. In addition to investigating more exhaustive local search methods we explore the use of techniques derived from research on discretizing continuous state spaces. Finally, we compare the results from our algorithms which search in the space of non-homogeneous partitions with exact and approximate algorithms which represent homogeneous and approximately homogeneous partitions as decision trees or algebraic decision diagrams

    A methodology for KEE systems target cascading

    The main aim of this paper is to present a methodology developed within the European Project VIVACE to guide the design and implementation of a Knowledge Enabled Engineering (KEE) system in a Virtual Enterprise. The proposed methodology tries to overcome some of the limitations which characterise traditional methods for Target Cascading, promoting a more collaborative and iterative approach to derive system specifications (in terms of advanced knowledge functionalities) from initial high-level targets. Social and behavioural aspects of Knowledge Management play a crucial role when many different users, knowledge experts, and process owners are involved in the Knowledge Management System (KMS) development. A well designed methodology is needed, therefore, to enhance communication and information sharing among design teams, to promote requirements merging and to take care both of the technological and behavioural aspects of the implementation. Initial business targets have been step-by-step decomposed into a set of sub-problems (Service Requirements, Knowledge Issues, and Knowledge Challenges) in the form of simple sentences in natural language. Then Quality Function Deployment (QFD) matrixes have been used to identify the set of functionalities to be implemented in the system, addressing the most important knowledge-related problems outlined in the cascading activity.Faste Laborator

    Extending the TAM for a t-cornmerce

    Broadband communication at home allows television to become an e-commerce medium that merges video, voice, and transactional data. t-commerce is electronically mediated commerce using interactive television. Theoretical and empirical analysis was undertaken and reported here to explain the factors that influence potential users&apos; adoption of t-commerce by extending the technology acceptance model. The study also compared the factors influencing t-commerce adoption between experienced and inexperienced users. Seven factors were identified: perceived ease of use, perceived usefulness, perceived enjoyment, trust, attitude, normative belief of family and friends, and subjective norm. Although the factors were not the same between the two groups, the differences were not large. Perceived enjoyment is the most important factor affecting attitude and behavioral intention toward t-commerce. From a theoretical point of view, the study extended the TAM for t-cornmerce adoption by potential users in the home environment.     Simple approach to one-laser, broadband coherent anti-Stokes Raman scattering microscopy

    Coherent anti-Stokes Raman scattering (CARS) microscopy is emerging as a powerful method for imaging materials and biological systems, partly because of its noninvasiveness and selective chemical sensitivity. However, its full potential for species-selective imaging is limited by a restricted spectral bandwidth. Recent increases in bandwidth are promising but still are not sufficient for the level of robust component discrimination that would be needed in a chemically complex milieu found, for example, in intracellular and extracellular environments. We demonstrate a truly broadband CARS imaging instrument that we use to acquire hyperspectral images with vibrational spectra over a bandwidth of 2500 cm^-1 with a resolution of 13 cm^-1.Tak W.
