Journal of Animal Science, 93(9), 4368-4376. doi:10.2527/jas2015-9171The objective of this study was to evaluate the efficacy of protecting PUFA within ground flaxseed against ruminal biohydrogenation by encapsulating them in a matrix consisting of a 1:1 blend of ground flaxseed and dolomitic lime hydrate (L-Flaxseed). Crossbreed heifers (n = 462, 346 +/- 19 kg) were blocked by weight and randomly assigned to pens. Pens were assigned to 1 of 6 dietary treatments in a randomized complete block design. Treatment 1 consisted of a combination of 54.6% steam-flaked corn (SFC), 30.0% wet corn gluten feed, 8.0% roughage, and supplement (0% flaxseed). In treatments 2 and 3, a proportion of SFC was replaced with 3 and 6% flaxseed, respectively; in treatments 4, 5, and 6, SFC was replaced with 2, 4, or 6% L-Flaxseed, respectively. Cattle were fed for 140 or 168 d and then harvested in a commercial abattoir where carcass data were collected. Approximately 24 h after harvest, carcasses were evaluated for 12th-rib fat thickness, KPH, LM area, marbling score, and USDA yield and quality grades. Samples of LM were also obtained for determination of long-chain fatty acid profiles. Cattle that were fed diets with 4 and 6% L-Flaxseed consumed less feed than other treatments (P 0.05). Supplementation with flaxseed increased (P 99%; increases for Flaxseed and L-Flaxseed of 0.095 and 0.140 mg of ALA/g of tissue for each percentage of flaxseed added). This study indicates that a matrix consisting of dolomitic lime hydrate is an effective barrier to ruminal biohydrogenation of PUFA; however, adverse effects on DMI limit the amounts that can be fed

Journal of Animal Science, 94(3), 1170-1178. doi:10.2527/jas2015-8779Fifty-two Holstein steers (573 +/- 9.92 kg BW) were used to determine if oral administration of crystalline menthol would induce changes in endogenous secretions of IGF-1 and circulating concentrations of glucose, lactate, and plasma urea nitrogen (PUN). Steers were blocked by BW and assigned within block to treatment. Treatments consisted of 0, 0.003, 0.03, or 0.3% crystalline menthol (DM basis) added to the diet. Animals were housed in individual, partially covered pens equipped with feed bunks and automatic water fountains. On d 1 of the experiment, blood samples were obtained via jugular venipuncture at 0, 6, 12, 18, and 24 h after feeding. Treatment administration commenced on d 2, and blood samples were again drawn at 0, 6, 12, 18, and 24 h after feeding. This blood-sampling schedule was repeated on d 9, 16, 23, and 30. Plasma was analyzed for PUN, glucose, and lactate concentrations. Serum was used to analyze IGF-1 concentration. Body weights were measured on d 1, 9, 16, 23, and 30. To accompany the live animal phase, in vitro fermentations were performed using ruminal fluid cultures. Measurements included VFA concentrations and fermentative gas production for cultures containing crystalline menthol at 0, 0.003, 0.03, or 0.3% of substrate DM. Addition of menthol to the diet of steers resulted in a treatment x day interaction (P 0.21). In conclusion, menthol supplementation minimally affected blood parameters associated with growth or ruminal fermentative activity

Journal of Animal Science, 94(1), 412-423. doi:10.2527/jas2015-9862The objective of this study was to evaluate the effect of steak location and postmortem aging on cooked meat tenderness and myofibrillar protein degradation of steaks from M. semitendinosus (ST). Following harvest and a 6 d chill period, the left ST was removed from carcasses of crossbred feedlot steers (n = 60, average hot carcass weight 427 +/- 24 kg). Each ST was fabricated into ten 2.54-cm thick steaks originating from the proximal to distal end of the muscle. Steaks cut adjacent to each other were paired, vacuum packaged, and randomly assigned to 7, 14, 21, 42, or 70 d of aging at 2 +/- 1 degrees C. After aging, within each steak pair, steaks were randomly assigned to Warner-Bratzler shear force or myofibrillar proteolysis analysis (calpain activity and desmin and troponin-T degradation). Muscle fiber type and size were also determined at the 2 ends of the muscle. There was no location x d of aging interaction (P = 0.25) for ST steak WBSF. Steak location affected (quadratic, P 0.13). Type I, IIA, and IIX muscle fibers were larger at the proximal end of the muscle than the distal end (P < 0.01). Increasing d of aging improved WBSF (quadratic, P < 0.01) for the duration of the 70 d postmortem period. As d of aging increased, intact calpain-1 activity decreased (quadratic, P < 0.01) with activity detected through 42 d. Day of aging affected autolyzed calpain-1 (linear, P < 0.01) and calpain-2 activity (quadratic, P < 0.01). Through d 70 of aging, the intensity of intact 55 kDa desmin band decreased (linear, P < 0.01), while there was an increase (linear, P < 0.01) in the degraded 38 kDa band. Similarly, d of aging increased troponin-T proteolysis, indicated by a decrease (quadratic, P < 0.01) in intensity of the intact 40 kDa band and an increase (linear, P < 0.01) in the 30 kDa degraded band. Intramuscular WBSF differences are not due to proteolytic activity or myofibrillar degradation and seem related to muscle fiber size. The improvement of ST steak WBSF through 70 d of aging is partly due to continued degradation of desmin and troponin-T. Calpain proteolytic analysis indicates that autolyzed calpain-1 and calpain-2 may be involved in extended postmortem myofibrillar protein proteolysis

    Mass Transport of Condensed Species in Aerodynamic Fallout Glass from a Near-Surface Nuclear Test

    In a near-surface nuclear explosion, vaporized device materials are incorporated into molten soil and other carrier materials, forming glassy fallout upon quenching. Mechanisms by which device materials mix with carrier materials have been proposed, however, the specific mechanisms and physical conditions by which soil and other carrier materials interact in the fireball, as well as the subsequent incorporation of device materials with carrier materials, are not well constrained. A surface deposition layer was observed preserved at interfaces where two aerodynamic fallout glasses agglomerated and fused. Eleven such boundaries were studied using spatially resolved analyses to better understand the vaporization and condensation behavior of species in the fireball. Using nano-scale secondary ion mass spectrometry (NanoSIMS), we identified higher concentrations of uranium from the device in 7 of the interface layers, as well as isotopic enrichment (&gt;75% 235U) in 9 of the interface layers. Major element analysis of the interfaces revealed the deposition layer to be chemically enriched in Fe-, Ca- and Na-bearing species and depleted in Ti- and Al-bearing species. The concentration profiles of the enriched species at the interface are characteristic of diffusion. Three of the uranium concentration profiles were fit with a modified Gaussian function, representative of 1-D diffusion from a planar source, to determine time and temperature parameters of mass transport. By using a historical model of fireball temperature to simulate the cooling rate at the interface, the temperature of deposition was estimated to be ∼2200 K, with 1σ uncertainties in excess of 140 K. The presence of Na-species in the layers at this estimated temperature of deposition is indicative of an oxygen rich fireball. The notable depletion of Al-species, a refractory oxide that is highly abundant in the soil, together with the enrichment of Ca-, Fe-, and 235U-species, suggests an anthropogenic source of the enriched species, together with a continuous chemical fractionation process as these species condensed

    Use of exogenous growth promotants in finishing cattle

DrouillardExogenous growth promotants, such as the synthetic beta agonist zilpaterol hydrochloride (ZH), have been shown to increase carcass weight by repartitioning energy toward increased skeletal muscle at the expense of adipose tissue, which is associated with a decline in tenderness. More recently, essential oils such as menthol have been observed to have growth promoting properties in livestock. The objectives of this research were to determine effects of ZH on blood parameters and long chain fatty acids in plasma and adipose tissue, to determine if the decline in tenderness can be negated by temporary depletion of calcium during ZH supplementation, and to determine effects of crystalline menthol on blood parameters. Blood samples were collected in 7-d intervals during ZH administration. Zilpaterol hydrochloride decreased concentrations of plasma urea nitrogen and whole blood glucose (P 0.10). Total long chain fatty acids of plasma and adipose tissue were unaffected (P > 0.10); however, ZH supplementation increased HCW, dressing percentage, and LM area (P 0.10) were observed for Warner-Bratzler shear force values, live animal performance, or carcass measurements. Addition of 0, 0.003, 0.03, 0.3% menthol (diet DM) to diets of steers resulted in a menthol × time within day interaction (P 0.05). Furthermore, concentrations of VFA were not different (P > 0.05), but production of fermentative gas was decreased (P < 0.01) when menthol was added at 0, 0.003, 0.03, 0.3% of substrate DM in a 24 h in vitro fermentation trial. Results from these studies suggest ZH improved efficiency of nutrient utilization for increased skeletal muscle growth; however, the decline in tenderness was not negated by the temporary depletion of calcium in the diet. Overall, ZH affected components related to increased skeletal muscle growth, but menthol did not affect blood parameters associated with growth

    Mineral supplementation of feedlot cattle

DrouillardFour studies evaluated effects of mineral supplementation on feedlot performance, carcass characteristics and ruminal fermentation of finishing cattle. Study 1 supplemented 0 or 3.3 g/d yeast combined with Cr propionate to steers separated into light and heavy groups. No treatment x weight group interactions were observed for ADG, DMI, final BW, carcass traits, or plasma glucose of lactate concentrations (P ≥ 0.06). A treatment x weight group interaction was observed for G:F (P = 0.03). In study 2, steers were supplemented 60 or 300 mg Zn/kg DM with or without zilpaterol hydrochloride (ZH). No interactions or effects of Zn or ZH were observed for IGF-1, plasma glucose, or lactate concentrations (P > 0.05). Plasma urea nitrogen (PUN) concentration decreased with ZH (P < 0.01). No interactions or effects of Zn or ZH were detected for ADG, DMI, final BW, G:F, and carcass traits were minimally affected (P ≥ 0.05). Study 3 evaluated effects of supplementing 30 or 100 mg Zn/kg DM (30 or 100Zn) with and without ractopamine hydrochloride (RH; 200 mg/d). No interactions or effects of Zn were observed for feedlot performance or PUN (P ≥ 0.07). Final BW, ADG, and HCW increased when heifers were fed RH (P ≤ 0.02). Zinc x RH interactions were observed for LM area and yield grade (P ≤ 0.01), but other carcass traits were not affected (P ≥ 0.08). In study 4, heifers were supplemented 0, 30, 60, or 90 mg Zn/kg DM. Zinc supplementation did not affect final BW, ADG, or DMI (P ≥ 0.07), but G:F increased linearly (P = 0.02). Carcass traits were not affected by Zn supplementation (P ≥ 0.07). Effects of in vitro Zn titration (0, 30, 60, 60, 90, 120, or 150 mg/kg Zn) were evaluated using ground corn and soybean meal as substrate. In vitro fermentation was not affected by added Zn (P ≥ 0.05). These studies suggest Cr and Zn supplementation minimally affected carcass traits, but Zn supplementation up to 60 mg/kg improved feed efficiency with minimal impact on ruminal fermentation. Supplementing increased Zn concentrations may alter fat and muscle deposition when fed with RH

    Accelerated step-up regimes for feedlot heifers following oral dosing with Lactipro (Megasphaera elsdenii strain NCIMB 41125)

    Cattle entering feedlots typically are adapted to finishing diets over a period of 2 to 4 weeks by gradually replacing forages with concentrate feeds using a series of step-up diets. Without proper adaptation, naïve cattle are highly susceptible to ruminal acidosis, a disorder associated with excessive production and accumulation of organic acids within the rumen. One of the key metabolic intermediates associated with the manifestation of acidosis is lactic acid, which is derived from fermentation of readily available starches and sugars. Streptococcus bovis is a prolific, rapidly growing, and opportunistic organism that thrives in the presence of readily fermented starches and sugars, and is an important inhabitant of the rumen that is recognized for its ability to produce large quantities of lactate. In unadapted cattle, the relative absence of lactate-utilizing bacteria can lead to the accumulation of lactate, thus predisposing the animals to acidosis. In traditional step-up programs, the gradual replacement of roughages with concentrate feeds provides ample time for proliferation of lactate-utilizing species of bacteria, the most important of which is Megasphaera elsdenii. Lactipro (MS Biotech, Inc., Wamego, KS) is a novel class of probiotic consisting of a highly prolific strain of Megasphaera elsdenii. Because Megasphaera elsdenii is an obligate anaerobe, it must be administered orally to avoid exposure to oxygen. In previous experiments with Lactipro, we have observed that a single oral dose results in rapid colonization of Megasphaera elsdenii within the rumen, effectively preventing the accumulation of lactate following an abrupt diet change from forage to concentrate. The present study was designed to evaluate different step-up regimens, with the objective of decreasing the time and number of diets required to place cattle on high-concentrate finishing diets
