    Metabotropic glutamate receptor 4-immunopositive terminals of medium-sized spiny neurons selectively form synapses with cholinergic interneurons in the rat neostriatum

    Metabotropic glutamate receptor 4 (mGluR4) is localized mainly to presynaptic membranes in the brain. Rat neostriatum has been reported to contain two types of mGluR4-immunoreactive axon varicosities: small, weakly immunoreactive varicosities that were distributed randomly (type 1) and large, intensely immunoreactive ones that were often aligned linearly (type 2). In the present study, most type 1 terminals formed asymmetric synapses on dendritic spines, whereas type 2 terminals made symmetric synapses on dendritic shafts, showing immunoreactivity for GABAergic markers. After depletion of neostriatal neurons, type 2 but not type 1 varicosities were largely decreased in the damaged region. When medium-sized spiny neurons (MSNs) were labeled with Sindbis virus expressing membrane-targeted green fluorescent protein, mGluR4 immunoreactivity was observed on some varicosities of their axon collaterals in immunofluorescence and immunoelectron microscopies. Furthermore, type 2 varicosities were often positive for substance P but mostly negative for striatal interneuron markers and preproenkephalin. Thus, striatonigral/striato- entopeduncular MSNs are likely to be the largest source of type 2 mGluR4-immunopositive axon terminals in the neostriatum. Next, in the double-immunofluorescence study, almost all choline acetyltransferase (ChAT)-immunopositive and 41% of NK1 receptor-positive dendrites were heavily associated with type 2 mGluR4-immunoreactive varicosities. Neuronal nitric oxide synthase (nNOS)-positive dendrites, in contrast, seemed associated with only a few type 2 varicosities. Conversely, almost all type 2 varicosities were closely apposed to NK1 receptor-positive dendrites that were known to be derived from cholinergic and nNOS-producing interneurons. These findings indicate that the mGluR4-positive terminals of MSN axon collaterals selectively form synapses with neostriatal cholinergic interneurons. © 2006 Wiley-Liss, Inc

    Activity-dependent adenosine release may be linked to activation of Na+-K+ ATPase : an in vitro rat study

    In the brain, extracellular adenosine increases as a result of neuronal activity. The mechanisms by which this occurs are only incompletely understood. Here we investigate the hypothesis that the Na+ influxes associated with neuronal signalling activate the Na+-K+ ATPase which, by consuming ATP, generates intracellular adenosine that is then released via transporters. By measuring adenosine release directly with microelectrode biosensors, we have demonstrated that AMPA-receptor evoked adenosine release in basal forebrain and cortex depends on extracellular Na+. We have simultaneously imaged intracellular Na+ and measured adenosine release. The accumulation of intracellular Na+ during AMPA receptor activation preceded adenosine release by some 90 s. By removing extracellular Ca2+, and thus preventing indiscriminate neuronal activation, we used ouabain to test the role of the Na+-K+ ATPase in the release of adenosine. Under conditions which caused a Na+ influx, brief applications of ouabain increased the accumulation of intracellular Na+ but conversely rapidly reduced extracellular adenosine levels. In addition, ouabain greatly reduced the amount of adenosine released during application of AMPA. Our data therefore suggest that activity of the Na+-K+ ATPase is directly linked to the efflux of adenosine and could provide a universal mechanism that couples adenosine release to neuronal activity. The Na+-K+ ATPase-dependent adenosine efflux is likely to provide adenosine-mediated activity-dependent negative feedback that will be important in many diverse functional contexts including the regulation of sleep

    Version 2020/03/08Kyoto University provides courses on 'The Practice of Basic Informatics' as part of its Liberal Arts and Sciences Program. The course is taught at many schools and departments, and course contents vary to meet the requirements of these schools and departments. This textbook is made open to the students of all schools that teach these courses. As stated in Chapter 1, this book is written with the aim of building ICT skills for study at university, that is, ICT skills for academic activities. Some topics may not be taught in class. However, the book is written for self-study by students. We include many exercises in this textbook so that instructors can select some of them for their classes, to accompany their teaching plans. The courses are given at the computer laboratories of the university, and the contents of this textbook assume that Windows 10 and Microsoft Office 2016 are available in these laboratories. In Chapter 13, we include an introduction to computer programming; we chose Python as the programming language because on the one hand it is easy for beginners to learn, and on the other, it is widely used in academic research. To check the progress of students' self-study, we have attached assessment criteria (a 'rubric') of this course as an Appendix. Current ICT is a product of the endeavors of many people. The "Great Idea" columns are included to show appreciation for such work. Dr. Yumi Kitamura and Dr. Hirohisa Hioki wrote Chapters 4 and 13, respectively. The remaining chapters were written by Dr. Hajime Kita. In revision for 2018 edition and after, Dr. Hiroyuki Sakai has participated in the author group, and Dr. Donghui Lin has also joined for English edition 2019. The authors hope that this textbook helps you to improve your academic ICT skill set. The content included in this book is selected based on the reference course plan discussed in the course development team for informatics at the Institute for Liberal Arts and Sciences.     Abstract: The present study deals with structural, morphological, dielectric and electrical properties of Zinc doped SrTiO3 powder particles which have been prepared via conventional solid-state route method. These were ball milled for 8 hr and furthermore calcined at 1050 0 C for 8hrs and sintered at 1250 0 C for 2hrs.The dielectric constant (K), dielectric loss (tan�) and thermoelectric powers (S) were computed. The obtained results using HIOKI 3532-50 LCR HiTESTER showed that the sample exhibited high dielectric constant of 246.6 at RT (100 Hz) and low loss of 0.50195 at RT (10 kHz).The samples were characterized using XRD, SEM,FTIR, EDAX and TEP for structural, micro structural, functional group, elemental and thermoelectric powe

Ośrodki te charakteryzują się różnymi zdolnościami sorpcji wody, która jako składnik silnie polarny wpływa na polaryzację elektryczną. Wyniki tych badań potwierdzają przewidywane, na podstawie wcześniejszych badań, istotnie różne wartości częstotliwości odpowiadające obserwowanej maksymalnej wartości kąta j dla piasku drobnoziarnistego i bentonitu.A prototype research stand based on modern impedance meter Hioki 3522 type was made in the Laboratory of Oil Engineering, Faculty of Drilling, Oil and Gas, AGH-UST. It was used for realization of a research project no. N524 348335 within which phase angle j of modeled fine sand and bentonite samples was tested for frequencies available with the meter, i.e. 0.001 Hz to 100 kHz in air-dry conditions. These media have different water sorption qualities; being strongly polar, water influences electrical polarization. The results of these experiments confirm results expected on the basis of previous tests. Significant differences were revealed between frequencies corresponding to the observed maximum value of angle j for fine sand and bentonite
