Hybridarchitekturen basierend auf kohlenstoffreichen pi-Systemen und Porphyrinen
The focus of this thesis “Hybrid architectures based on carbon rich pi-systems and porphyrins” was set on hybrid systems containing porphyrin units combined with carbon-rich pi-systems and the way towards those.
The first part of this thesis described the investigation of precursor fragments of two of the herein used pi-systems, the hexaarylbenzenes (HABs) and their oxidized species, the hexa-peri-hexabenzocoronenes (HBC), which are required for the donor-acceptor hybrid architectures. In that regard, the multigram synthesis of HABs via the “functionalization of para-nitroaniline” (FpNA) route, developed in the Jux group, is shown. This route offers substitution geometries of HABs that are of relevance for this thesis. In particular, the meta arrangements such as A3BAB and A3BAC, which have not been accessible before. This method was utilized further and adopted for a similar approach, the “functionalization of ortho-nitroaniline” (FoNA), with the goal to prepare HABs with up to six different substituents. Therefore, comparable reaction conditions were applied: brominations, Sandmeyer and Suzuki reactions. Attempts towards regioisomerically pure functional HABs with dipole, or quadrupole arrangement were performed utilizing FoNA. The key step was the regioisomeric bromination. Even though directed brominations of one isomer over the other one could be achieved, a complete regioisomerically pure reaction was never reached. Yet, the results are encouraging and offer the possibility for further investigations.
FpNA has been applied for the synthesis of meta and para substitution patterns of HABs, which formed the basis for the herein studied zinc-porphyrin (ZnPor)-HBC/HAB-C60 hybrid architectures. These were investigated with respect to the arrangement of the donor and acceptor units in space to each other: ortho, meta and para. The therefore necessary new aldehyde HAB structures and other novel HAB derivatives have been synthesized successfully, were compared to literature known systems and tested for post-functionalization reactions using electrophilic aromatic substitution or Iridium-catalyzed CH-activation procedures. However, several post-functionalization attempts were unsuccessful. Thus, we carried out theoretical studies by DFT (density functional theory) calculations. Theoretical results were compared to literature-known successfully post-functionalized HABs, and a reasonable connection between successful and unsuccessful experiments could be made by comparing the highest occupied molecular orbitals (HOMOs), local ionization potential and electrostatic potential maps. These calculations give an idea on possible active sites for an electrophilic aromatic substitution and reveal not only the structural difference, but as a consequence thereof the electronical dissimilarity of HAB derivatives. Herein, the decisive aspect was not, as expected, the HOMO, but rather the local ionization potential. Taking this into account, this finding offers the possibility regarding future theoretical investigations prior to experimental work, in order to predict reactions or the behavior of HABs. Accompanied with that, preparation and investigation of their oxidized species, the HBCs, were conducted. Similar to the HAB systems, they have been tested for literature-known post functionalization via CH-activation using an Iridium catalyst, as well. Unfortunately, those attempts did not result in the desired functionalized HBC systems.
With regards to the desired hybrid architectures, a further functionalization of aforementioned HAB/HBC derivatives was not possible, so that an alternative pathway was developed. By targeted design of suitable precursors for a statistical Suzuki reaction, trimethylsilyl (TMS)-aldehyde-HABs could be successfully synthesized. Separation of the mixture was possible due to the strong difference in polarity of the functional groups (aldehyde, TMS). Then, by applying a Silyl-Halogen-exchange reaction, iodine-substituted meta and para HAB aldehydes were successfully synthesized, and could be structurally compared to each other and the ortho HAB. With this tool in hand, the structural isomeric row of ortho, meta and para HABs were prepared. The same is true for the corresponding HBC systems which were obtained after Scholl reaction. With Suzuki and Prato reactions, the desired ZnPor-HBC-fullerene hybrids were successfully synthesized. The hybrid systems, as well as the precursors were studied with nuclear magnetic resonance (NMR) spectroscopy. With variable temperature (vt) proton NMR, dynamics within the hybrids were studied and in order to find the best characterization conditions for each structure. The corresponding donor-acceptor HAB systems were prepared additionally and studied for similar aspects, as well as their precursors. Furthermore, literature-known ZnPor-benzene-fullerene systems were prepared as reference materials. All pi-system containing hybrid architectures were compared, regarding structural and electronic behavior depending on the geometrical alignment, and the type of pi-system. Unlike in symmetrical bis-porphyrin systems, in which differences in ground state communication was observed among different isomers, photophysical investigations of the donor acceptor conjugates revealed that the excited state communication, studied by emission spectroscopy, played the crucial role, and which altered with structural orientation (ortho, meta and para), as well as the type of bridging pi-systems. Further structural and electronical differences were compared using DFT calculations.
The second part of this thesis described the preparation of polycyclic aromatic hydrocarbon (PAH)-substituted tetrabenzoporphyrins (TBPs) as basis for novel graphyrin hybrid systems (nanocaps, nanocones, molecular nanodots, buckyballs). Herein, PAH-TBP hybrid species were synthesized in good yields based on an optimized, recently reported method, and modified in the aforementioned regards. Photophysical studies revealed communication between the PAH and the TBP moieties, which was particular evident in their fluorescence spectra. Further emission studies were performed in this regard with a row of pyrenyl-TBPs carrying different substituents in the d-positions of the TBP moiety, in which intramolecular exciplex formation was observed. Additional DFT calculations were carried out in order to study structural differences between the PAH-TBPs. Insights in interesting CH-pi interactions and packing motif were obtained by the crystal structure of triphenylphenyl-TBP.
Concerning the novel graphyrin precursors, benzo[g]chrysene-TBP, offers a large structural variety due to the possible formation of atropisomers. As studied theoretically, this can result in structurally different conformers in on surface studies, which could potentially lead to different nanocaps. Out of these atropisomers and conformers, nanocaps for nanotubes with varying chiralities could be formed. Meaning that this one molecule theoretically results in three different single-walled-carbon-nanotubes (SWCNTs), which contain a nitrogen-functionalized endcap that might be of potential interest to nano-driven applications. Based on triphenylphenyl-substituted TBP, for the first time, a novel nanocone architecture consisting of a pi-extended carbon-rich surrounding and an intrinsic nitrogen-functionalized top stemming from the porphyrin core, could be formed on Pt(111) surface via surface-assisted cyclodehydrogenation (SACDH). Furthermore, it was possible to embed this cone into a graphene lattice, which was grown from carbon bulk impurities by annealing of the Pt-crystal. Accompanied with that, a theoretical explanation for the geometrical interconversion of a squared nanocone seed into the hexagonal structure of graphene was given. The pyrenyl-TBPs were investigated towards wet-chemical cyclodehydrogenation for buckyball or molecular nanodot architectures. All herein conducted experiments resulted in the formation of nearly insoluble materials, that were studied via UV/Vis spectroscopy as standard technique. In all experiments, a loss of the fine structure features, as well as broadening and bathochromic shift of the signals were observed, which indicated a pi-extension of the hybrid molecules. Lastly within this part, a pyrene-TBP was studied on graphene by TEM, towards the preparation of a novel buckyball compound class. By metalation with Pb as heavy metal, giving a high contrast in TEM, atom molecule interactions and dynamics were studied and for the first time, evidence for the existence of such a graphyrin buckyball was collected, which was further supported by DFT calculations, in order to understand and reconstruct the formation of this porphyrin-buckyball. All investigations in the first and second part of this thesis confirmed that these novel hybrids are not just a linear combination of the utilized compound classes in terms of properties, but display rather novel and unique architectures with own characteristics.
The last part of this thesis was devoted to the synthesis and purification of porphyrin mixtures, prepared from two different aldehydes through a statistical approach. Hereby, automated techniques were explored, in order to find efficient methods towards low symmetrical porphyrins. A modified literature approach using a microwave assisted synthesis was used for porphyrin mixtures with different functional groups. This synthesis showed how to control the reaction, and driving it towards good yields of the desired isomers by variation of equivalents of the starting materials. Furthermore, automated purification by programmed medium pressure liquid chromatography (MPLC) protocols revealed the separation in an efficient way, including time, solvent amount and recycling, as well as reuse of silica gel columns for several separation experiments.
Summarizing, this thesis offered insights into novel carbon-rich pi-system based architectures, which are of fundamental importance in order to understand electronic communication behavior across molecular hybrid materials. Moreover, it contributes new aspects in the field of molecular based novel nanomaterials, based on carbon-rich pi-systems and porphyrins, and provides fundamental synthetic procedures and also concepts, relying on automated methodologies.Im Fokus dieser Dissertation “Hybridarchitekturen basierend auf kohlenstoffreichen pi-Systemen und Porphyrinen“ standen Hybridsysteme, welche sich aus Kombinationen von Porphyrinen und kohlenstoffreichen pi-Systemen zusammen setzten, sowie deren Vorstufen und der Weg zu eben diesen Hybriden.
Der erste Teil dieser Arbeit beschreibt Untersuchungen von Vorstufen oder Fragmenten von Hexaarylbenzolen (HABs) und deren oxidierten Spezies, den Hexa-peri-hexabenzocoronenen (HBCs), auf welchem die Donor-Akzeptor Hybrid Architekturen beruhen. Aus diesem Grund wurde die Multigramm Synthese von HABs, welche über die „Funktionalisierung von para-Nitroanilin“ (FpNA) erreicht und in der Arbeitsgruppe Jux entwickelt wurde, erwähnt. Diese Route bietet die Möglichkeit der Synthese verschiedener Substitutionsmustern an HABs, welche unter anderem für diese Dissertation relevant sind. Hierbei besonders interessant sind die meta Substitutionsmuster A3BAB und A3BAC, welche bisher nicht zugänglich waren. Zudem wurde diese Methode weiter verwendet und adaptiert für die auf ähnlichen Reaktionen (Bromierungen, Sandmeyer- und Suzuki-Reaktionen) beruhende „Funktionalisierung von ortho-Nitroanilin“ (FoNA), mit dem Ziel, ein HAB mit sechs verschiedenen Substituenten herzustellen. Im Speziellen wurde mit Hilfe der FoNA versucht, funktionelle HABs mit Dipol- oder Quadrupol-Anordnungen regioisomeren-rein herzustellen. Obwohl eine gezielte Bromierung, welche ein Isomer bevorzugt, erreicht werden konnte, konnte eine regioisomeren-reine Reaktion bisher nicht ermöglicht werden. Nichtsdestotrotz sind die bisher erzielten Ergebnisse vielversprechend und bieten die Möglichkeit für weiterführende Untersuchungen.
Die FpNA wurde für die Synthese von meta and para Substitutionsmustern von HABs angewandt, welche das Fundament für Herstellung und Untersuchung der Zinkporphyrin (ZnPor)-HBC/HAB-C60 Hybdride bilden. Jene wurden im Hinblick auf die räumliche Anordnung von Donor und Akzeptor Einheit zueinander untersucht: ortho, meta and para. Daher konnten entsprechende neue Aldehyd-HAB Strukturen und weitere neuartige HAB Derivate erfolgreich hergestellt werden, mit literaturbekannten Systeme verglichen und für Postfunktionalisierungen, wie elektrophile aromatische Substitution und Iridium katalysierte CH Aktivierung, getestet werden. Allerdings waren verschiedene Postfunktionalisierungsversuche, obwohl anders erwartet, nicht erfolgreich. Aus diesem Grund wurden theoretische Studien mittels Dichtefunktionaltheorie (DFT) Rechnungen durchgeführt. Theoretische Ergebnisse wurden mit literaturbekannten, erfolgreich postfunktionalisierten HABs verglichen; ein plausibler Zusammenhang zwischen erfolgreichen und nicht erfolgreichen Experimenten konnte durch den Vergleich der Verteilung der höchst-besetzten molekularen Orbitale (highest occupied molecular orbitals, HOMOs), lokalem Ionisierungspotential und elektrostatischem Potential, gefunden werden. Diese Kalkulationen dienen als Orientierung für mögliche aktive Angriffspunkte für eine elektrophile aromatische Substitution an HABs und decken nicht nur strukturelle, sondern als Konsequenz auch elektronische Ungleichheiten von HAB-Derivaten auf. Der entscheidende Faktor hierbei war nicht, wie erwartet die Verteilung des HOMOs, sondern vielmehr des lokalen Ionisierungspotentials. Mit diesen Ergebnissen werden neue Möglichkeiten für zukünftige theoretische Untersuchungen im Vorfeld von experimentellen Arbeiten geschaffen, um Reaktionen und das mögliche Verhalten von HABs vorherzusagen. Weiterführend wurden deren oxidierte Spezies (HBCs) hergestellt und untersucht. Ähnlich zu den HABs wurden auch diese hinsichtlich literaturbekannter Postfunktionalisierungsmethoden durch CH-Aktivierung mit Hilfe eines Iridium-Katalysators getestet, welche jedoch leider nicht in den gewünschten funktionalisierten HBC Systemen resultierten.
Durch gezieltes Design geeigneter Vorstufen für eine statistische Suzuki Reaktion, konnten Trimethylsilyl(TMS)-Aldehyd-HABs erfolgreich synthetisiert werden. Eine Trennung der verschiedenen Produkte war aufgrund der starken Polaritätsunterschiede der funktionellen Gruppen (Trimethylsilyl, Aldehyd) möglich. Anschließend konnten durch einen Silyl-Halogen-Austausch Iodid-substituierte meta und para HAB-Aldehyde gezielt erfolgreich hergestellt werden, welche strukturell miteinander sowie dem entsprechenden ortho HAB verglichen wurden. Mithilfe dieser zweier Reaktionen konnte die vollständige Reihe der Strukturisomere der ortho, meta and para HABs synthetisiert werden.
Das Gleiche gilt für die entsprechenden HBC Systeme, welche nach einer Scholl-Reaktion der HABs erhalten wurden. Mit Suzuki- und Prato-Reaktionen konnten die gewünschten ZnPor-HBC-Fulleren-Hybride abschließend erfolgreich dargestellt werden. Die Hybridsysteme, wie auch deren Vorstufen wurden mittels NMR-Spektroskopie untersucht. Durch temperaturabhängige Protonen NMR-Spektroskopie wurden strukturelle Dynamiken innerhalb der Hybride aufgezeigt und es konnten die optimalen Charakterisierungsbedingungen für jede einzelne Struktur gefunden werden. Die entsprechenden Donor-Akzeptor-HAB-Systeme wurden ebenfalls zusätzlich hergestellt und wurden ähnlich studiert, ebenso wie deren Vorstufen. Außerdem wurden literaturbekannte ZnPor-Benzol-Fulleren-Systeme zum Vergleich hergestellt und herangezogen. Alle pi-System enthaltenden Hybridarchitekturen wurden hinsichtlich ihrem strukturellen und elektronischen Verhalten abhängig von der geometrischen räumlichen Anordnung und der Art des verbrückenden pi-Systems studiert und verglichen. Im Gegensatz zu symmetrischen Bis-Porphyrin-Systemen, in denen Unterschiede in der Kommunikation im Grundzustand zwischen den verschiedenen Strukturisomeren beobachtet wurden, deckten photo-physikalische Untersuchungen, vor allem mittels Fluoreszenz Spektroskopie, der hier vorgestellten Donor-Akzeptor Konjugate, eine Kommunikation im angeregten Zustand, verändert durch die strukturelle Orientierung (ortho, meta, para) sowie durch die Art des verbrückenden pi-Systems, auf. Weitere strukturelle und elektronische Unterschiede wurden mit Hilfe von DFT Rechnungen verglichen.
Im zweiten Teil der Arbeit wird die Synthese von mit polyzyklisch-aromatischen-Kohlenwasserstoffen (PAKs)-substituierten Tetrabenzoporphyrinen (TBPs) als Basis für neuartige Graphyrin (=Graphen-Porphyrin-Hybrid)-Systeme (Nanocaps = Nanotube Endkappen, Nanocones, molekulare Nanodots, Buckyballs) vorgestellt. Hierbei wurden die PAK-TBP Konjugate in guten Ausbeuten basierend auf einer modifizierten, optimierten, kürzlich veröffentlichten Methode synthetisiert. Durch photophysikalische Untersuchungen konnte die Kommunikation zwischen der PAK- und der TBP-Einheit aufgezeigt werden, welche sich besonders offensichtlich in den Fluoreszenzspektren ausdrückte. Weitere Emissionsstudien wurden für eine Reihe von Pyrenyl-TBPs durchgeführt, welche unterschiedliche Substituenten in den d-Position der TBP-Einheit trugen, wobei ein intramolekularer angeregter Komplex (Exciplex) beobachtet wurde. Zusätzliche DFT-Rechnungen unterstützten die Analyse struktureller Unterschiede zwischen den unterschiedlichen PAK-TBPs. Einblicke in interessante CH-pi Interaktionen und Packungsverhalten konnten durch die Kristallstruktur von Triphenylphenyl-TBP gewonnen werden. Benzo[g]chrysen-TBP bietet aufgrund möglicher Atropisomere eine große strukturelle Variation als neuartige Graphyrin-Vorstufe. Die hier theoretisch durchgeführte Studie zeigt daher als Folge strukturell unterschiedliche Konformere in Oberflächen Studien auf, welche potenziell in verschiedenen Nanocaps resultieren. Aus diesen Atropisomeren und Konformeren können Nanocaps für Nanotubes mit variierbarer Chiralität entstehen. Diese bedeutet, dass dieses eine Molekül theoretisch drei verschiedene einwandige Kohlestoffnanoröhren (single-walled-carbon-nanotubes = SWCNTs), welche eine Stickstoff-funktionalisierte Endkappe besitzen, hervorbringen kann. Basierend auf Triphenylphenyl-substituiertem TBP konnte zum ersten Mal durch Oberflächen assistierte Cyclodehydrogenierung (surface-assisted cyclodehydrogenation = SACDH), eine neuartige Nanocone-Architektur bestehend aus einer pi-erweiterten kohlenstoffreichen Umgebung und einer aus dem Porphyrinkern stammenden intrinsischen Stickstoff-funktionalisierterten Spitze auf einer Pt(111) Oberfläche realisiert werden. Weiterhin war es möglich, diesen Cone in eine Graphenfläche einzubetten, welche durch aus dem Pt-Kristall austretende Kohlenstoffverunreinigungen durch thermisches Ausglühen gewachsen ist. Begleitet wurde dieses Ergebnis durch ein theoretisches Gedankenexperiment für die geometrische Umwandlung des quadratischen Nanocones in die hexagonale Struktur von Graphen. Pyrenyl substituierte TBPs wurden hinsichtlich nass-chemischer Cyclodehydrogenierung für Buckyball oder molekulare Nanodot-Architekturen untersucht. Hierbei resultierten alle durchgeführten Versuche in der Bildung eines nahezu unlöslichen Materials, welches nur durch UV/Vis-Spektroskopie als gängige Methode studiert werden konnte. Alle Experimente zeigten den Verlust der Feinstruktur der Spektren im Vergleich zu den PAK-TBP-Hybrid Vorstufen, sowie der Verbreiterung und der bathochromischen Verschiebung der Banden, was auf eine Erweiterung des pi-Systems des Hybrids schließen lässt. Zuletzt wurde innerhalb dieses Teils der Arbeit ein Pyren substituiertes TBP auf einer Graphenoberfläche mit Hilfe von Transmissionselektronenmikroskopie (TEM) zur Herstellung einer neuartigen Buckyball Verbindungsklasse untersucht. Durch gezielte Metallierung mit Blei als Schwermetall, welches einen hohen Kontrast unter TEM-Bedingungen hervorruft, konnten Metallatom-Molekül-Interaktionen und -Dynamiken studiert werden. Zum ersten Mal wurde hiermit ein Anzeichen für die Existenz eines solchen Graphyrin Buckyballs beobachtet, welches weiterhin durch DFT-Kalkulationen unterstützt wurde, um seine Bildung zu verstehen und nachzuvollziehen.
Alle Untersuchungen im ersten und zweiten Teil dieser Arbeit bestätigen, dass diese neuen Hybride nicht nur eine Linearkombination der verwendeten Verbindungsklassen in Bezug auf Eigenschaften, sondern vielmehr neuartige und einzigartige Architekturen mit eigenen Charakteristika, darstellen.
Der letzte Teil der Arbeit ist der Synthese und Aufreinigung von Porphyrin Mischungen, die aus zwei verschiedenen Aldehyden statistisch synthetisiert wurden, gewidmet. Automatisierte Techniken wurden hierbei untersucht, um effiziente Methoden zur Aufreinigung niedersymmetrischer Porphyrine zu finden. Eine modifizierte Literaturmethode, welche sich Mikrowellen-assistierter Synthese bedient, wurde für Porphyrinmischungen mit unterschiedlichen funktionellen Gruppen verwendet. Die Synthese zeigte, wie die Reaktion hin zu guten Ausbeuten des gewünschten Isomers durch Variation der Äquivalente der Startmaterialien gesteuert werden konnte. Außerdem konnte durch automatisierte Aufreinigung mit Hilfe von programmierten „medium-pressure liquid chromatography (MPLC)“ Protokollen die Trennung der Isomere effizient in Bezug auf Zeit, Lösungsmittelmenge und Recycling, sowie der mehrmaligen Wiederverwendung von Silica-Gel-Säulen für Trennungsversuche, realisiert werden.
Zusammenfassend bietet diese Arbeit Einblicke in neuartige Kohlenstoff-reiche pi-System- und Porphyrin-basierte-Architekturen, welche von fundamentaler Bedeutung für das Verständnis von elektronischer Kommuni
Mehrfache „Click” Reaktionen an Porphyrinen
The concept of the 1,3-dipolar “click” alkyne-azide cycloaddition was successfully applied to ortho-benzylic fuctionalized tetraphenylporphyrins presented by JUX in 2000. As the further modification of these porphyrins is indispensable the “click” cycloaddition was optimized for these basic porphyrins in order to obtain good yields and easy working procedures. Thus, over 40 sophisticated and complex molecular structures were prepared. All of these new compounds feature the triazole ring as binding motif and could be characterized by standard chemical analytic methods. The optimized reaction conditions were determined successfully to be CuSO4*H2O as Cu(I)-source and sodium ascorbate as reducing agent. As solvent, a mixture of dichloromethane, ethanol and water delivered the best results. Very important is the addition of the sterical demanding base N,N’-diisopropylethylamine, which increases the yield enormously. Thus, diverse acetylene compounds were attached to various azidoporphyrins with different symmetries. The ortho-benzylic fuctionalized tetraphenylporphyrins synthesized turned out to be favorable precursors for this reaction class as the azido functionalized derivatives can be generated in a few easy steps which all proceed under full conversion. In this manner, tetraphenylporphyrins with up to eight azido units in the ortho-benzylic positions can be obtained easily. As acetylene compounds ethynyl ferrocene, phenyl acetylene and a dendritic ferrocene alkyne conjugate were chosen and were reacted with the different azidoporphyrins. Thus, a plenitude of triazole connected porphyrin conjugates was prepared. This modified “click” method allowed the easy attachment of new functional groups to the basic porphyrins. The yields of the less often functionalized systems (mono- and bis-substituted porphyrins) were satisfactory with around 60%, but excellent results were achieved for the highly-substituted systems (tetra- and octa-substituted porphyrins). These reactions proceeded with complete conversion, which means that the yield for each single reaction center on the porphyrin is higher than 99%. An autocatalytical mechanism and the stabilization of the catalytically active Cu(I)-species by the triazole moieties were supposed to be the reason for this excellent success. The structures of these systems were determined by NMR spectroscopy as well as by mass spectrometry. The ferrocene conjugates were also examined via cyclic voltammetry as the ferrocene unit offers interesting electrochemical behavior. Since this “click” approach turned out to work very well with the JUX porphyrins bridged porphyrins modeled on BALDWIN’S capped porphyrins were created. As bridging unit ferrocene or a dendritic compound were used and anchored to the azido groups of two different azidoporphyrins. In this way, simple- and double-capped ferrocene porphyrins as well as simple- and double-capped dendritic porphyrins were formed. The electrochemical properties of these systems were investigated via cyclic voltammetry and showed an astonishing additional oxidation step, whose origin can be found in the strained configuration of the porphyrin. The capability of the 1,3-dipolar “click” reaction with our porphyrin systems was underlined by creating oligomers consisting of different porphyrins. An array of conjugates built up of different porphyrins, ferrocene units and dendritic NEWKOME branches was generated. One approach, for instance, was the synthesis of a pentamer consisting of one porphyrin in the core and four porphyrins attached to it. A tetrakisazidoporphyrin was used as core porphyrin and four alkyne porphyrins were connected via the 1,3 dipolar SHARPLESS cycloaddition. Due to the autocatalytical process caused by the ortho-position of the azido groups this reaction gave full conversion. Other examples for synthesized oligomers are porphyrins which are linked via a ferrocenyltriazolyl bridge or dendritic ferrocenyl-bridged porphyrins. At last, a long desired approach was realized with this Cu(I)-catalyzed alkyne-azide coupling on the azidoporphyrins: the side-selective functionalization of the tetrakis and octakisazidoporphyrin. This synthesis is challenging due to a lot of isomers that can be formed when the porphyrins are not converted on every of its four or eight reaction centers. By taking the right stoichiometrically deficient proportion of the alkyne compound, the triazole units were built only on one side of the porphyrin plane. Thus, the side-selective modification was carried out successfully via the “click” approach. The side-selective functionalization can clearly be determined via the NMR spectra which prove the difference between the upper and the lower side of the porphyrin plane. Thus, hydrophilic and hydrophobic groups can be attached to the highly-substituted porphyrin in a regioselective manner.Im Rahmen der vorliegenden Arbeit wurde die 1,3-dipolare SHARPLESS Alkin-Azid Cycloaddition sehr erfolgreich auf die von JUX im Jahr 2000 vorgestellten ortho-benzylisch substituierten Tetraphenylporphyrinen übertragen. Durch diese Reaktion gelang es über 40 komplexe und anspruchsvolle Moleküle in sehr guter Ausbeute zu synthetisieren. Als gemeinsames Verknüpfungsmotiv besitzen diese Strukturen den durch die Cycloaddition gebildeten Triazolring. Die dargestellten Verbindungen wurden durch die gängigen analytischen Methoden charakterisiert. Bei der 1,3-dipolaren Cu(I)-katalysierten Cycloaddition wurden die Bedingungen auf die Umsetzung mit den vorliegenden ortho-funktionalisierten Porphyrinen erfolgreich optmimiert und bestimmt. Als Cu(I)-Quelle wurde CuSO4*5 H2O verwendet, welches in situ durch Natriumascorbat reduziert wurde. Eine Mischung aus Dichlormethan, Ethanol und Wasser diente als Lösungsmittel und als zusätzliche Base lieferte N,N‘-Diisopropylethylamin die besten Resultate. Auf diese Weise konnten verschiedene Alkinkomponenten an die entsprechenden Porphyrine gekoppelt werden und so eine Fülle an komplizierten Porphyrinstrukturen mit verschiedenen Symmetrien erhalten werden. Die Azidopophyrine, welche die Ausgangsverbindungen für die entsprechenden „Click“ Reaktionen sind, wurden sehr einfach aus den von JUX et al. vorgestellten ortho-benzylisch funktionalisierten Porphyrinen erhalten. Als Alkinkomponente wurden Phenylacetylen, Ethinylferrocen und ein dendritisches Ferrocenderivat eingesetzt. Bei den durchgeführten „Click“ Cycloadditionen fiel auf, dass die Ausbeuten je Reaktionszentrum zunahm, desto mehr Substituenten das Porphyrin trägt. So verlief die Umsetzung von einfach und doppelt funktionalisierten Azidoporphyrinen mit etwa 50 - 60% zwar mit einer zufriedenstellenden Ausbeute, doch eine quantitative Umsetzung wurde für die Reaktion der hochsubstituierten Systeme (vier und achtfach funktionalisierte Porphyrine) beobachtet. Als Erklärung hierfür wurde ein autokatalytischer Prozess postuliert, der durch die steigende Anzahl an Triazoleinheiten hervorgerufen wird. Die Triazolringe können das katalytisch-aktive Cu(I) stabilisieren, was zu einer signifikanten Verbesserung der Ausbeute führt. Aufgrund der guten Ergebnisse, wurden überbrückte Porphyrinsysteme nach dem Beispiel von „BALDWINS capped porphyrin“ synthetisiert. Als Brückeneinheit wurde ein Ferrocendialkin und ein dendritisches Dialkin verwendet, welche jeweils an den Azidogruppen des Porphyrins verankert wurden. Dieser Ansatz gelang mit dem zweifach substituierten syn-Porphyrin und dem vierfach substituierten Porphyrin. Auf diese Art und Weise konnten einfach und doppelt verbrückte Porphyrine in guten Ausbeuten erhalten werden. Die elektrochemischen Eigenschaften dieser Konjugate wurden mit Hilfe von zyklischer Voltammetrie untersucht. Zu den typischen zwei Ein-Elektronen-Oxidationen des Porphyrins wurde bei diesen Systemen eine zusätzliche Oxidation detektiert, deren Ursache die verzerrrte Konfiguration der Porphyrine ist. Weiterhin wurde durch die „Click“ Chemie eine Reihe von oligomeren Konjugaten aus verschiedenen Porphyrinen synthetisiert. Einerseits gelang es Oligomere, die durch Ferroceneinheiten verknüpft sind darzustellen andererseits wurden auch verschiedene Porphyrine durch Triazolringe zu höheren Konjugaten verknüpft. Die Idee der einseitigen Funktionalisierung an den beiden verschiedenen Seiten des Porphyrins konnte durch die optimierte „Click“ Reaktion verwirklicht werden. Durch den richtig-berechneten stöchiometrischen Unterschuss wurde der Triazolring nur an einer Seite der Porphyrinebene gebildet. Dadurch können verschiedene Substituenten auf beiden Seiten eingeführt werden und die beiden Ebenen selektiv modifiziert werden. So kann durch die SHARPLESS Cycloaddition z. B. eine hydrophile und eine hydrophobe Halbsphäre um das Porphyrin erzeugt werden
Neuartige Funktionalisierte Porphycene
The first part of this thesis dealt with highly charged porphycenes. Simultaneously with the formation of an eightfold positively charged porphycene the syntheses of three model systems – tetrapropylporphycene, tetraphenylporphycene, and tetra-t-butylphenylporphycene – were carried out and optimized: The pyrroles were synthesized by reaction of the respective aldehydes with two equivalents of ethyl isocyanoacetate, and the iodopyrroles were generated using iodine monochloride. Regarding the iodopyrrole that would lead to the tetrapropylporphycene, a review of the Ullmann reaction conditions did not show a positive effect of the protection of the pyrrolic NH group. On the contrary, the Ullmann reaction proceeded quantitatively without protection. The subsequent saponification and decarboxylation of the bipyrrole tetracarboxylic esters were investigated with the help of the bipyrrole most similar to that leading to the eightfold positively charged porphycene. Here, the most crucial point was to conduct the experiment in such a high concentration so that the product could be obtained as solid. The following Vilsmeier-Haack formylations and McMurry reactions yielded the desired porphycenes. In cooperation with the Guldi research group (Erlangen) the three model compounds were subjected to a detailed analysis of their physico-chemical properties, including ground and excited state characterization by steady-state as well as by time-dependent measurements. In cooperation with the Marbach research group (Erlangen) the molecular arrangement of tetraphenylporphycene on a Cu(111) surface was investigated with the help of scanning tunneling microscopy.
With respect to the syntheses of the precursors of the eightfold positively charged porphycene, the most commonly used methods for the pyrrole formation did not work because of the steric demands of the starting material. The desired pyrrole could only be obtained via the reaction of the aldehyde with two equivalents of ethyl isocyanoacetate. Similarly, the following iodination only met with success, when the treatment with iodine and potassium iodide was replaced by the reaction with iodine monochloride. Optimization afforded the iodopyrrole in a quantitative manner. After the Ullmann coupling and the combined saponification and decarboxylation reaction, the free bipyrrole was subjected to a formylation under Vilsmeier-Haack conditions. Although a partial chlorination took place, the mixture of the dialdehydes was carried on to a McMurry reaction and yielded a mixture of porphycenes with a varying degree of chlorination. This mixture was treated with hydrogen bromide and gave the octabromoporphycene which led to the eightfold positively charged porphycene via an SN2 reaction with t-butylpyridine. In cooperation with the Guldi research group this molecule was characterized by steady-state as well as time-resolved methods and then incorporated into graphene oxide photoelectrodes via an LbL approach.
The second part of this thesis was devoted to the implementation of porphycenes into dye-sensitized solar cells (DSSCs). For that reason a library of porphycenes was generated. The synthesis started with the generation of a porphycene bearing a catechol anchoring group. This molecule was used by the Guldi research group to exfoliate graphene and then to generate DSSCs. The results were an incentive to focus on the optimization of porphycenes for the application in p-DSSCs. Five different porphycenes were generated. The linker between the anchoring group and the porphycene did not contain a heteroatom anymore but was a conjugated C4 unit, which would facilitate the electronic communication. The incorporation of these five porphycenes into DSSCs and their investigation were conducted in cooperation with the Guldi research group.
The third part of this thesis explored the modification of the porphycene by extension of the conjugation pathway. Several approaches were evaluated with the aim of finding the best route to a fused porphycene. The attachment of t-butylphenyl groups at the meso-positions gave a porphycene, which could provide two phenanthrene-like extensions after an oxidation. However, its insolubility after purification by column chromatography prevented subsequent reactions. Using 4,5,6,7-tetrahydro-2H-isoindole as a building block furnished another porphycene that would lead to a fused porphycene after an oxidation, but only in very low yields. The most promising procedure was the bicyclo[2.2.2]octadiene-based route relying on a retro-Diels-Alder reaction as the final step to the fused porphycene. The required bipyrrole was obtained via a recently published route to the respective pyrrole in combination with an oxidative coupling procedure, and it was subsequently converted to a diketone. This molecule was used to evaluate the conditions of the retro-Diels-Alder reaction. Most likely owing to its steric demand, a symmetrical McMurry reaction did not give any porphycene. By carrying out a McMurry reaction with the diketone and a sterically less demanding dialdehyde an asymmetrically substituted porphycene was obtained. The final retro-Diels-Alder reaction yielded the desired dibenzoporphycene. The synthetic route to a porphycene with this substitution pattern was considerably shortened.Der erste Teil der Arbeit befasste sich mit hochgeladenen Porphycenen. Zeitgleich mit der Darstellung eines achtfach positiv geladenen Porphycens wurde die Synthese dreier Modelsysteme – Tetrapropylporphycen, Tetraphenylporphycen und Tetra-tert-butylphenylporphycen – durchgeführt und optimiert: Die Pyrrole wurden durch die Reaktion der entsprechenden Aldehyde mit zwei Äquivalenten Isocyanessigsäureethylester synthetisiert, und die nachfolgenden Iodpyrrole wurden durch Reaktion mit Iodmonochlorid erhalten. Eine Überprüfung der Bedingungen der Ullmann-Reaktion am Beispiel des Iodpyrrols, das zum Tetrapropylporphycen führen würde, zeigte keinen positiven Einfluss der Schützung der pyrrolischen NH Gruppe. Vielmehr verlief die Ullmann-Reaktion ohne Schützung quantitativ. Die darauffolgende Verseifung und Decarboxylierung der Bipyrroltetracarbonsäureester wurde anhand desjenigen Bipyrrols untersucht, welches dem, das zu dem achtfach positiv geladenen Porphycen führen würde, am ähnlichsten war. Die Reaktion musste in einer so hohen Konzentration durchgeführt werden, dass das Produkt als Feststoff erhalten werden konnte. Die nachfolgenden Vilsmeier-Haack-Formylierungen und McMurry-Reaktionen erbrachten die gewünschten Porphycene. In Zusammenarbeit mit der Arbeitsgruppe von Prof. Dr. Guldi (Erlangen) wurden diese drei Moleküle einer eingehenden Analyse ihrer physikalisch-chemischen Eigenschaften unterzogen. Ihre Grund- und angeregten Zustände wurden durch stationäre und zeitaufgelöste Messungen charakterisiert. In Zusammenarbeit mit der Arbeitsgruppe von Priv. Doz. Dr. Marbach (Erlangen) wurde die Molekülanordnung von Tetraphenylporphycen auf einer Cu(111) Oberfläche mit Hilfe von Rastertunnelmikroskopie untersucht.
Bei der Synthese der Vorstufen des achtfach positiv geladenen Porphycens erwiesen sich die allgemein gebräuchlichen Methoden der Pyrroldarstellung auf Grund des räumlichen Anspruchs des Ausgangsmaterials als nicht erfolgreich. Das gewünschte Pyrrol konnte schließlich über die Reaktion des Aldehyds mit zwei Äquivalenten Isocyanessigsäureethylester gewonnen werden. Die folgende Iodierung zum Iodpyrrol wurde durch die Reaktion mit Iodmonochlorid ermöglicht und konnte nach Optimierung mit quantitativen Ausbeuten durchgeführt werden. Im Anschluss an die Ullmann-Kupplung und die Verseifung und Decarboxylierung wurde das freie Bipyrrol einer Vilsmeier-Haack-Formylierung unterworfen. Trotz einer teilweisen Chlorierung wurde die Mischung der Dialdehyde in eine McMurry-Reaktion eingebracht und ergab ein Gemisch aus unterschiedlich oft chlorierten Porphycenen. Für die anschließende Umsetzung zum Oktabromoporphycen wurde dieses Gemisch mit Bromwasserstoffsäure versetzt. Eine SN2 Reaktion mit tert-Butylpyridin ergab das achtfach positive geladene Porphycen. In Zusammenarbeit mit der Arbeitsgruppe von Prof. Dr. Guldi wurde dieses Porphycen mit Hilfe von stationären und zeitaufgelösten Methoden untersucht und anschließend mittels der sogenannten LbL-Technik in Graphenoxid-Photoelektroden eingebracht.
Der zweite Teil dieser Arbeit war auf Porphycene für den Einsatz in farbstoffsensibilisierten Solarzellen ausgerichtet. Zu diesem Zweck wurde eine Auswahl an Porphycenen dargestellt. Zunächst wurde ein Porphycen, das eine Catechol-Ankergruppe trug, synthetisiert. Dieses wurde anschließend im Arbeitskreis von Prof. Dr. Guldi zur Exfoliierung von Graphen und Herstellung farbstoffsensibilisierter Solarzellen verwendet. Die erzielten Ergebnisse waren Anlass, die Porphycene für die Anwendung in farbstoffsensibilisierten Solarzellen zu optimieren. Fünf verschiedene Porphycene wurden synthetisiert. Die Verbindungseinheit von Ankergruppe und Porphycen enthielt kein Heteroatom mehr, sondern bestand aus einer konjugierten C4 Einheit, welche die elektronische Kommunikation erleichtern sollte. In Zusammenarbeit mit der Arbeitsgruppe von Prof. Dr. Guldi wurden die Integration dieser Porphycene in farbstoffsensibilisierte Solarzellen und deren Untersuchung durchgeführt.
Der dritte Teil dieser Arbeit war der Modifizierung von Porphycenen durch die Vergrößerung des Konjugationsweges gewidmet. Verschiedene Synthesestrategien wurden mit dem Ziel untersucht, den besten Weg zu einem „fused porphycene“ zu finden. In der Annahme, dass eine nachfolgende Oxidation zwei phenanthren-ähnliche Erweiterungen liefern könnte, wurde ein Porphycen mit vier tert-Butylphenyl Gruppen an den meso-Positionen synthetisiert. Allerdings verhinderte seine Unlöslichkeit nach der säulenchromatographischen Reinigung weiterführende Reaktionen. Ausgehend von 4,5,6,7-Tetrahydro-2H-isoindol konnte ein Porphycen, das nach einer Oxidation ein „fused porphycene“ ergeben würde, dargestellt werden, allerdings nur in sehr geringer Ausbeute. Die vielversprechendste Methode war die auf einem bicycylo[2.2.2]octadien-substituierten Porphycen basierende Route mit einer retro-Diels-Alder-Reaktion als letztem Schritt zum „fused porphycene“. Das dafür benötigte Bipyrrol wurde nach einer vor kurzem veröffentlichten Synthesevorschrift für das entsprechende Pyrrol, gefolgt von einer oxidativen Kupplung, dargestellt und anschließend zu einem Diketon umgesetzt. Dieses Diketon ermöglichte die Evaluation der Bedingungen der retro-Diels-Alder-Reaktion. Höchstwahrscheinlich auf Grund des räumlichen Anspruchs führte eine symmetrische McMurry-Reaktion zu keinem Porphycen. Durch eine McMurry-Reaktion des Diketons mit einem sterisch weniger anspruchsvollen Dialdehyd wurde das asymmetrisch substituierte Porphycen erhalten. Die abschließende retro-Diels-Alder-Reaktion ergab das gewünschte Dibenzoporphycen. Der Weg zu Porphycenen mit diesem Substitutionsmuster wurde somit erheblich verkürzt
Wasserlösliche Metalloporphyrine als biomimetische Modelle für die Sauerstoffaktivierung
The aim of this work was the synthesis and characterization of novel metalloporphyrins that can serve as model systems for the cytochrome P450 superfamily. Thus, novel iron porphyrins with covalently attached cystein-S donor were synthesized. The model systems feature one or two triazole moieties as common binding motif, which were introduced by the 1-3 dipolar copper-I catalyzed alkyne-azide cycloaddition reaction. A bisphenol A-strapped porphyrin with topologically different faces was synthesized from a novel dipyrrylmethane and postmodified to the related zinc azidoporphyrin. Subsequent Click-reaction with a bidentate alkynyl peptide, lead to a zinc conjugate that was converted to the related iron porphyrin. In that case the peptide is attached to the porphyrin via two triazole moieties. This double junction effectively stabilizes the peptide ligand and prevents dissociation of the ligated thiolate. It was possible to reproduce the characteristic absorption band at about 450 nm in the UV/Vis for that iron porphyrin in DMSO. In native cytochrome P450 enzymes the thiolate is embedded in the so-called cysteine-loop. Inspired by nature’s cysteine-loop novel alkynyl peptides and azido peptides were synthesized and attached via one triazole ring to azidoporphyrins and alkynylporphyrins, respectively. Due to their inherent higher flexibility the peptide ligands are capable of mimicking the cysteine-loop. Thus, it was possible to generate the characteristic absorption band at about 450 nm in the UV/Vis for these iron porphyrins in DMSO. A further challenge was the synthesis of water-soluble cytochrome P450 model systems. Following the idea of the cysteine-loop a carboxylated iron porphyrin and a hexacationic iron porphyrin were synthesized. For both species the pH dependence of the effect of the thiolate ligand was studied in aqueous media. For the carboxylated iron porphyrin the spectroscopic results showed no influence of the pH value. Due to the formation of mü-oxo-dimers it was not possible to generate the characteristic absoption band at about 450 nm. The hexacationic iron porphyrin showed after reduction to the FeII-species and coordination of CO gas a shoulder at 445 nm at pH = 7. Due to the six positive charges of the pyridinium groups a formation of the thiolate ligated FeII-CO complex becomes possible. A further goal of this thesis was the synthesis of novel water-soluble ruthenium porphyrin complexes. A free-base porphyrin with four malonates was metallated with trisrutheniumdodecacarbonyl to obtain the corresponding ruthenium complex in very good yields from which the related pyridine and acetonitrile complexes were synthesized in quantitative manner. After saponification of the malonates under basic conditions it was possible to isolate a water-soluble ruthenium porphyrin with eight carboxylates and two axial ethanol ligands. By metallation of a fourfold methoxymethylated free-base porphyrin with trisrutheniumdodecacarbonyl a ruthenium dimer was synthesized and successfully converted to the related monomeric pyridine complex. Subsequent bromomethylation lead to a pyridine complex and to a compound without axial ligand, which was successfully complexed with acetonitrile. Thereupon it was possible to functionalize the four bromomethylene moieties of that compound with 4-t-butylpyridine, which lead to a fourfold quaternized ruthenium porphyrin with an axial 4-t-butylpyridine and coordinated CO ligand. A eightfold methoxymethylated free-base porphyrin was metallated with trisrutheniumdodecacarbonyl to a ruthenium dimer. After complexation with pyridine and bromomethylation the eightfold bromomethylated pyridine complex was isolated together with another eightfold bromomethylated compound. The latter species was successfully complexed with acetonitrile and reacted with n-butylimidazole and 4-t-butylpyridine. n-Butylimidazole was capable of displacing the CO ligand which leads to a ruthenium complex with two axial n-butylimidazole ligands. 4-t-butylpyridine was not capable of displacing the axial CO ligand which leads to a ruthenium complex with one axial 4-t-butylpyridine ligand and one axial CO ligand.Das Ziel dieser Arbeit war die Synthese und Charakterisierung neuartiger Metalloporphyrine, die als Modellsysteme für die Cytochrom P450 Überfamilie dienen können. Um dies zu erreichen, wurden verschiedene Eisenporphyrine dargestellt die einen kovalent angebundenen Cysteamin-Donor besitzen. Als gemeinsames Bindungsmotiv besitzen alle diese Substanzen einen oder zwei Triazolringe, die mittels 1-3 dipolarer Kupfer-I katalysierten Alkin-Azid Cycloaddition erzeugt wurden. Hierfür wurde zunächst ein Bisphenol A-überbrücktes Porphyrin aus einem neuartigen Dipyrrylmethan dargestellt. Nach Modifizierung zu einem Zink-Azidoporphyrin und anschließender Reaktion mit einem zweizähnigem alkinylierten Peptid konnte das mit Zink metallierte Konjugat erhalten werden welches zum entsprechenden Eisenporphyrin modifiziert wurde. Durch doppelte Anbindung des Peptidliganden an das Porphyrin mittels zweier Triazol-Einheiten wird das Peptid besser stabilisiert und somit eine Dissoziation des angebundenen Thiolatliganden verhindert. Für das Eisenporphyrin konnte die charakteristische Absorptionsbande bei ungefähr 450 nm nachgewiesen werden. In natürlichen Cytochrom P450 Enzymen liegt das Thiolat eingebunden im sogenannten Cystein-loop vor. Davon inspiriert, wurden neuartige Alkinylpeptide und Azidopeptide synthetisiert und nachfolgend über einen Triazolring an Zink-Azidoporphyrine und Zink-Alkinylporphyrine gebunden. Durch die inhärente Flexibilität der Peptidliganden sind diese in der Lage, den Cystein-loop zu imitieren, wodurch es gelang, die charakteristische Absorptionsbande bei ungefähr 450 nm für diese Eisenporphyrine zu erzeugen. Eine weitere Herausforderung war die Synthese wasserlöslicher Cytochrom P450 Modellsysteme. Aufbauend auf der Idee des Cystein-loops wurde ein dreifach anionisches Eisenporphyrin und ein sechsfach kationisches Eisenporphyrin dargestellt. Mittels dieser beiden Systeme war es möglich, den Effekt des Thiolatliganden pH-abhängig in Wasser zu untersuchen. Für das dreifach anionische Eisenporphyrin zeigten die UV/Vis spektroskopischen Befunde keine pH-Abhängikeit. Durch die Ausbildung eines mü-oxo-Dimers gelang es nicht, die charakteristische Absorptionsbande bei 450 nm im UV/Vis Spektrum zu erzeugen. Das sechsfach kationische Eisenporphyrin zeigte nach Reduktion zur FeII-Spezies und Koordination von CO eine Schulter bei 445 nm im UV/Vis Spektrum bei pH 7. Eine Deprotonierung des Thiols zum Thiolat wurde ermöglicht durch die sechs kationischen Pyridiniumgruppen, die sich in unmittelbarer Nähe zum Peptidliganden befinden. Ein weiteres Ziel dieser Arbeit war die Darstellung wasserlöslicher Rutheniumporphyrine. Durch Metallierung eines Porphyrinsystems mit vier Malonestergruppen mit Trisrutheniumdodekacarbonyl gelang die Darstellung eines Ruthenium-Komplexes in sehr guten Ausbeuten. Anschließende Komplexierung mit Pyridin und Acetonitril führte zu zwei neuartigen Komplexen. Nach basischer Verseifung der Malonestergruppen gelang es ein wasserlösliches Rutheniumporphyrin mit acht Carboxylatgruppen und zwei axialen Ethanol Liganden herzustellen. Nach Metallierung eines mit vier Methoxymethylengruppen substituierten Porphyrinsystems mit Trisrutheniumdodekacarbonyl konnte ein Ruthenium-Dimer in guten Ausbeuten isoliert werden. Es gelang dieses Ruthenium-Dimer durch Zugabe von Pyridin in den entsprechenden Pyridin-Komplex zu überführen. Die anschließende Spaltung der Methoxymethylengruppen lieferte den entsprechenden vierfach bromomethylierten Pyridin-Komplex und eine vierfach bromomethylierte Verbindung, welche erfolgreich mit Acetonitril komplexiert werden konnte. Daraufhin gelang es, die vier bromomethylen Gruppen dieser Spezies mit 4-t-Butylpyridin zu funktionalisieren und somit ein Rutheniumporphyrin mit vier Pyridiniumgruppen und axialem 4-t-Butylpyridin sowie einem axialen CO-Liganden darzustellen. Ein mit acht Methoxymethylengruppen substituiertes Porphyrinsystem wurde mit Trisrutheniumdodekacarbonyl metalliert, wodurch das entsprechende Ruthenium-Dimer erhalten wurde. Komplexierung mit Pyridin und Etherspaltung der Methoxymethylengruppen des Pyridin-Komplexes lieferte den achtfach bromomethylierten Pyridin-Komplex und eine weitere achtfach bromomethylierte Verbindung. Letztere ließ sich quantitativ mit Acetonitril komplexieren wodurch ein achtfach bromomethylierter Acetonitril-Komplex erhalten wurde. Dieser wurde im Anschluß mit n-Butylimidazol und 4-t-Butylpyridin zur Reaktion gebracht. Das elektronenreiche n-Butylimidazol ist in der Lage, den Axial gebundenen CO-Liganden zu verdrängen. Dadurch konnte ein mit acht Imidazoliumgruppen substituiertes Rutheniumporphyrin, welches zwei axiale n-Butylimidazol Liganden koordiniert, isoliert werden. Das 4-t-Butylpyridin hingegen zeigte keine Verdrängung des axialen CO, wodurch ein Rutheniumporphyrin mit acht Pyridiniumgruppen sowie einem axialen 4-t-Butylpyridin Liganden isoliert werden konnte
Naturbasierte und vollsynthetische, chirale Zykloketo-Porphyrinsysteme - Entwicklung neuartiger Photosensibilisatoren
Several semi-natural and synthetic cycloketo-porphyrins were synthesized and their properties and usability for different applications was subjected to detailed investigations. It was dealt with porphyrin systems with an exocyclic connection of a meso-position or -substituent to a beta-pyrrolic position of the macrocycle formally originating from an acylation reaction to give a ketone functionality. Such systems can be found in nature amongst the chlorophylls. Thus, derivatives of chlorophyll a were firstly in focus. Synthetic analogs, deriving from “porphyrin building blocks” with one to eight ortho-bromomethyl side-chains, introduced by N. Jux, were made accessible, characterized and investigated within the second part of this thesis. For the semi-natural systems, an optimized, highly efficient isolation method for pheophorbides from spirulina was worked out to easily and quickly deliver large amounts of that materials. With those materials at hand, it was possible to reproduce and verify recent findings in connection with their applicability in photosensitizing systems for photodynamic therapy (PDT). The materials have been used for detailed investigations on their photophysical and electrochemical properties to be able to compare them with other sensitizers. To access artificial analogs, a synthetic strategy has been developed, starting from an mono-functionalized o-(bromomethyl) „porphyrin building block“ providing a novel cycloketo-porphyrin system via a cyanomethyl derivative and the porphyrin ethanoic acid. This system exhibits a seven-membered exocycle as major structural feature. The newly established methodology was also applied to other building blocks with up to 8 functionalities leading to a library of free base cycloketo-porphyrins and their metal complexes which have been investigated by chemical and photophysical means. As the characterization data showed, the systems contain a distorted pi-system arising from the structural features being analog to those already known for dodeca-substituted porphyrins. But in contrast, the ones under investigation here turned out to be highly stable in configuration, as VT NMR demonstrated. They thus represent stable inherently chiral systems. The racemic mixtures obtained from synthesis have been resolvable by chiral HPLC. So, it was possible to characterize the individual enantiomers by CD spectroscopy. Ffor the first time, we were able to describe stable inherently chiral porphyrins whose chirality is due to a distorted pi-system. Via temperature-dependent NMR spectroscopy, it was also possible to show that the lowering of symmetry is leading to the presence of distinguishable tautomers within the mono-exocyclic cycloketo-porphyrin. Interestingly, those tautomers turned out to be differentiable by photophysical means. So, a novel spectroscopic methodology for the detection and characterization of tautomeric porphyrin structures was developed. The conducted photophysical experiments showed that the structural elements and the thus arising deviation from planarity within the pi-system is leading to an enhanced spin-orbit-coupling with a high intersystem crossing probability and hence, an efficient excitation into triplet states. It was also shown that the excitation energy can be transferred onto present triplet oxygen nearly quantitatively and it was demonstrated that the corresponding performance of the systems can be effectively tuned via variations in the architecture of the exocycle as well as by degree and regiochemistry of poly-annulation. Thus, novel singlet oxygen generators were made accessible captivating through their high stability and outstanding quantum yields. Such systems are of great interest for hygiene applications (e.g. for detoxification of water) and also for medicinal means (foremost for applications in terms of PDT) as, in comparison to pheophorbides, they exhibit a far higher stability, a better solubility and an increased efficiency. Also corresponding metal complexes (e.g. indium(III)) and coupleable derivatives with additional amino functionality were made accessible without significant changes in the photophysical performance. Those systems allowed investigations on novel concepts of application. For example, it has been shown that the Förster resonance energy transfer can be utilized within photosensitizing concepts as the results on the conjugate of pyropheophorbide a and the mono-exocyclic cycloketo-porphyrin illustrated. Thus, excitation wavelengths can be effectively tuned. Furthermore, the investigations on indium(III) complexes implied that novel combinational therapies could be opened up. The implementation of a radioactive metal center (e.g. In-111) could then be used for tumor localization while the ligand, being an effective photosensitizer, would allow for an in situ treatment by PDT.Es wurden hierbei naturstoffbasierte und vollsynthetisch zugängliche Zykloketo-Porphyrine dargestellt und ihre Eigenschaften und Anwendungsmöglichkeiten genauer untersucht. Dabei handelte es sich um Porphyrine, die einen Exozyklus aufweisen, der eine meso-Position oder einen meso-Substituenten mit einer beta-pyrrolischen Position verbindet. Die Art der Verbrückung basiert auf einer Acylierungsreaktion und beinhaltet somit ein Keton. Solche Systeme finden sich in der Natur unter den Chlorophyllen, sodass die Derivate des Chlorophylls im zunächst im Focus der Arbeit standen. Synthetische Analoga, die sich von „porphyrin building blocks“ mit ein bis acht ortho-Brommethylseitenketten ableiten, wurden dann zugänglich gemacht, charakterisiert und untersucht. Bei den naturstoffbasierten Systeme ist es gelungen, ein optimiertes, hocheffizientes Verfahren zur Gewinnung von Pheophorbid a aus Spirulina zu erarbeiten, das leicht und schnell Gramm-Mengen zugänglich macht. Anhand dieser Verbindungen konnten die bisher erzielten Ergebnisse über Anwendbarkeit in Photosensibilisatorsystemen für die PDT nachvollzogen und bestätigt werden. Sie wurden auch für photophysikalische und elektrochemische Untersuchungen herangezogen, um Vergleiche mit anderen Sensibilisatorsystemen vorzunehmen. Um vollsynthetische Analoga zu erhalten, wurde, ausgehend vom monofunktionalen o-brommethyl-substituierten „porphyrin building block“, eine Synthesestrategie entwickelt, die über das Cyanomethyl-Derivat und die Porphyrin-Essigsäure zu einem neuartigen Zykloketo-Porphyrinsystem führte. Dieses verfügt über einen siebengliedrigen Exozyklus als maßgebliches Strukturmerkmal. Die neu etablierte Verfahrensweise konnte auf weitere „building blocks“ (bis zu 8 funktionellen Gruppen) angewendet werden und ergab so eine Bibliothek von freibasigen Zykloketo-Porphyrinen und ihren Metallkomplexen. Auch diese wurden chemisch wie photophysikalisch untersucht. Wie die Charakterisierungsdaten zeigten, liegt in diesen Systemen, von Strukturmerkmalen bedingt, ein deformiertes Pi-System vor, wie es z. B. für dodeka-substituierte Porphyrine bekannt ist. Im Gegensatz zu diesen, sind die hier untersuchten Systeme außerordentlich konfigurationsstabil und stellen inhärent chirale Systeme dar. Die synthetisch erhaltenen Racemate konnten mittels chiraler HPLC aufgetrennt werden und es gelang die Charakterisierung der Antipoden mittels CD-Spektroskopie. Somit war erstmalig die Beschreibung stabiler, inhärent chiraler Porphyrine möglich, deren Chiralität durch Verzerrung des Pi-Systems bedingt wird. Mittels temperaturabhängiger NMR-Spektroskopie konnte gezeigt werden, dass die Symmetrieerniedrigung zu unterscheidbaren Tautomeren des mono-exozyklischen Zykloketo-Porphyrins führt. Diese konnten auch photophysikalisch nachgewiesen werden. Dies führte zur Entwicklung eines neuartigen spektroskopischen Verfahrens zur Unterscheidung und Beschreibung tautomerer Strukturen in Porphyrinen. Die Untersuchung der photophysikalischen Eigenschaften zeigte, dass die Strukturelemente und die einhergehende Verzerrung des Pi-Systems zu einer effizienten Spin-Bahn-Kopplung führen. Diese bedingt eine hohe Intersystem Crossing-Wahrscheinlichkeit und somit eine effektive Anregung in Triplett-Zustände, welche die Anregungsenergie nahezu quantitativ auf Triplett-Sauerstoff übertragen können. Es konnte auch gezeigt werden, dass die Effektivität dabei durch die gewählte Architektur des Exozyklus sowie durch Grad und Regiochemie der Polyfunktionalisierung steuerbar ist. So wurden neuartige Singulett-Sauerstoff-Generatoren zugänglich, die durch hohe Stabilitäten und Quantenausbeuten bestechen. Diese Systeme sind dabei sowohl für die Hygienetechnik (z. B. die Entkeimung von Luft) als auch für die medizinische Technologie (v. a. für die PDT) interessant, da sie, gegenüber den Pheophorbiden z. B., eine weitaus höhere Stabilität, eine bessere Löslichkeit und eine gesteigerte Effizienz besitzen. Da auch entsprechende Metallkomplexe (z. B. des Indium(III)) und kopplungsfähige amino-funktionalisierte Derivate zugänglich sind, ohne dass sich die photophysikalischen Eigenschaften signifikant ändern, konnten neuartige Photosensibilisierungskonzepte untersucht werden. Z. B. konnte anhand des Konjugats aus Pyropheophorbid a und dem mono-exozyklischen Amino-Zykloketo-Porphyrin gezeigt werden, dass der Förster Resonanz-Energietransfer in derartige Konzeptionen für die PDT implementiert werden kann. Damit werden Anregungswellenlängen steuerbar. Ferner legen die Versuche mit der Indium(III)-Verbindung nahe, dass Kombinationstherapieformen erschlossen werden können. Die Einbringung eines radioaktiven Metallzentrums (z. B. In-111) kann dabei für die Tumordetektion verwendet werden, während der Ligand als aktiver Sensibilisator eine in situ-Therapierung mittels PDT ermöglicht

The focus of this thesis was settled around the hypothetical structure of a graphene–porphyrin hybrid system, which was named “graphyrin”.
The first part is concerned with the exploration and preparation of functionalized hexaarylbenzenes (HABs) and hexa-peri-hexabenzocorones (HBCs). In that respect, literature known techniques, namely the Knoevenagel–Diels–Alder cascade and the metal-catalyzed [2+2+2]cyclotrimerization approach were investigated in order to prepare scaffolds with the so-called “common” geometry pattern. Utilizing a monomode microwave reactor, improved protocols were developed that outperform literature known reaction procedures in feasibility, yield and reaction time.
Furthermore, a novel rational pathway towards highly functionalized hexaarylbenzenes was developed; namely the “functionalization of para-nitroaniline” (FpNA). The novel approach allows for the preparation and investigation of HABs with up to five different substituents and thus geometries of yet unexplored nature, while the formation of regioisomers are completely avoided. In detail, the preparation of 26 substitution patterns, starting from para-nitroaniline, can be carried out in a cascade of facile bromination, Sandmeyer-halogenation, and Suzuki coupling reactions. Preliminary emission experiments in solution and in the solid state showed strong blue to white fluorescence of hexaarylbenzenes, bearing an intrinsic quadrupole moment. These findings are likely to be explained by a toroidal delocalization of pi-electrons, which can be observed for in close proximity parallel standing aryl rings, as it is true for an HAB.
The second part of the thesis covers the synthetic strategy, adding hexaarylbenzenes and hexa-peri-hexabenzocorones to the periphery of porphyrins, in order to extend the pi-conjugated carbon framework, which is closely related to graphyrin. Thus, a feasible and high yielding strategy was realized, starting from a tetratolanylporphyrin, which was obtained in high yields in a microwave-assisted reaction. The Diels–Alder reaction of such porphyrins with tetraphenylcyclopentadienones resulted in a statistical distribution of all possible hexaarylbenzene-porphyrin scaffolds. These could be separated by column chromatography and analyzed in detail. It was found that all conjugates show an extraordinary solid–state fluorescence of intensities, completely unknown to porphyrins. Additionally, the statistical product mixtures were successfully introduced as calibration mixtures for mass spectrometry.
The oxidation of HAB-porphyrins to the respective HBC–porphyrin conjugates was achieved with modified Scholl conditons, using FeCl3 as Lewis-acid and oxidant. It was found that additional amounts of trifluoroacetic acid facilitated the solubilization of the HAB–porphyrin scaffolds and resulted in higher conversions. Following this methodology, the largest PAH-substituted porphyrin, which is known in literature, could be synthesized in high yields. Accompanied with this novel compound class, peculiar gas phase behavior in the mass spectrometer could be detected. Therefore, a series of meso-unsubstituted HBC–porphyrin conjugates, namely A, trans-A2, A3 and A4 were synthesized in order to understand their tendency to form large supramolecular clusters. Analysis of the whole series revealed that the intermolecular cluster formation could be ascribed to the pi-pi-interactions of the HBC moieties. These findings were further confirmed by X-ray diffraction analysis of several HBC–porphyrin conjugates that showed the repetitive intermolecular HBC–HBC packing motif as the dominant solid state interaction. Photophysically, an almost instant energy transfer on the picosecond timescale from the HBC to the central porphyrin could be observed, which makes these conjugates interesting candidates as light–harvesters.
Furthermore, the conceptual design of HBC–porphyrin scaffolds was inverted and HBC–centered bisporphyrin conjugates, substituted in ortho, meta and para position were synthesized. These conjugates showed a peculiar deviation of the spectroscopic properties, depending on the position of the porphyrins towards each other. In detail, a split in the Soret band could be observed for the para-conjugate, whereas the ortho conjugate showed no splitting and the meta-conjugate an intermediate shoulder. These observations are indicative for an exciton coupling, which is pronounced in the para-conjugate due to a sufficient communication ability over the pi-system of the HBC. This ability is decreased for the meta and ortho system, which is reflected in the low or no splitting of the Soret band.
The family of HBC–porphyrins was further extended by alkynyl-bridged conjugates, which were investigated in single-molecule-conductance studies at the University of Oxford. It turned out that t-butyl substituted HBCs are no sufficient anchor groups for the utilization as single molecule transistors.
The third part of the thesis is dedicated to the field of tetraaryltetrabenzoporphyrins (TATBPs). Therefore, the usability of different masked-isoindoles was evalutated. In that respect, bicyclo[2.2.2]octadiene fused pyrrole was determined as isoindole synthon of choice and a general condensation protocol towards TATBPs was developed. The new protocol, which is essentially a modified Lindsey protocol, outperforms every literature known procedure towards TATBPs in feasibility and yield.
By means of a gamma-ortho cyclization, the exploration of TATBPs as fragment of graphyrin was evaluated. Therefore, literature known procedures towards strained PAHs were explored. Collision–induced dissociation experiments (CID), carried out in the mass spectrometer showed that the fjord-region cyclodehydrofluorination worked effectively towards the eightfold gamma-ortho cyclization. In that respect a fundamental fragment of graphyrin could be prepared and analyzed by mass spectrometry. Wet-chemically, the palladium-catalyzed intramolecular direct arylation of ortho-chlorinated TATBPs could be successfully applied to the closure of up to four C–C bonds. A closure beyond four bonds however, could not be achieved and the respective halogen atoms were found to be replaced by hydrogen atoms. With regard to the largest synthesized graphyrin fragment, an HBC substituted tetrabenzoporphyrin was synthesized. Unfortunately, it was found that a gamma-ortho cyclization could not be achieved under oxidative or reductive conditions.
The last part of the thesis describes one of the smallest and most fundamental fragments of graphyrin, namely [18]annulene. A novel synthetic three-step procedure was developed, which is superior over yet published procedures towards [18]annulene in terms of feasibility and yield. Hereby, the intermediate compound hexadehydro[18]annulene, which is prone to rapid decomposition in the solid state, could be crystallized in a benzene matrix and analyzed by X-ray diffraction. The benzene molecules served as effective protectants towards topological polymerization in the solid state. Furthermore, a novel phenomenon – the “annulene–hole effect” – leading to very tight pi-pi-interactions in the solid state was gained from the respective crystal structure of [18]annulene. This effect could potentially lead to a novel design of semi-conducting materials that rely on effective charge-carrier mobilities in the solid state.
Finally, all insights that were gained from each research segment en route to graphyrin resulted in important and fundamental information about graphene–porphyrin hybrid systems; however, even more important, each field itself developed as highly interesting and flourishing research topic, that allows to continue the exploration of fragments of graphyrin in a manifold manner, without an end in sight.Im Fokus dieser Arbeit lag die hypothetische Struktur eines Graphen–Porphyrin–Hybrid Systems, das „Graphyrin“ benannt wurde.
Der erste Teil der Arbeit beschäftigte sich mit der Herstellung funktionalisierter Hexaarylbenzole (HABs) und Hexa-peri-hexabenzocoronene (HBCs). Diesbezüglich wurden die literaturbekannten Protokolle, nämlich die Knoevenagel–Diels–Alder–Kaskade und die Metall–katalysierte [2+2+2]Cyclotrimerisierung untersucht, um zu Gerüsten mit sogenannten „gewöhnlichen“ Substituitionsmustern zu gelangen. Durch Zuhilfenahme eines Monomode–Mikrowellenreaktors konnten verbesserte Protokolle entwickelt werden, die literaturbekannte Synthesen in Handhabbarkeit, Ausbeute und Reaktionszeit deutlich überragten. Desweiteren konnte eine komplett neuartige Methode zu hochfunktionalisierten Hexaarylbenzolen entwickelt werden: nämlich die „Funktionalisierung von para-Nitroanilin“ (FpNA). Die neue Methode erlaubt die Herstellung von HABs mit bis zu fünf verschiedenen Substituenten und somit Geometrien von unerforschter Natur. Die Generierung von Regioisomeren kann dabei komplett vermieden werden. Die Synthese von 26 Substitutionsmustern konnte durch eine Abfolge von einfachen Bromierungen, Sandmeyer–Halogenierungen und Suzuki–Kopplungen, beginnend mit para-Nitroanilin, realisiert werden. Vorläufige Emissionsexperimente sowohl in Lösung als auch im Feststoff zeigten eine starke blaue bis hin zu einer weißen Fluoreszenz einiger Hexaarylbenzole mit intrinsischem Quadrupolmoment. Diese Beobachtungen lassen sich möglicherweise auf die torusförmige Delokalisierung von Pi-Elektronen, welche für einander nahestehende, parallel angeordnete Aryle, wie sie in HABs zu finden sind, möglich ist, zurückführen.
Der zweite Abschnitt der Arbeit umfasst die Synthesestrategie HABs und HBCs in die Peripherie von Porphyrinen zu integrieren, um das Pi-konjugierte Kohlenstoffgerüst, welches einen weiteren Schritt in Richtung Graphyrin darstellt, zu erweitern. Dahingehend wurde ausgehend von einem Tetratolanylporphyrin, welches in hohen Ausbeuten in einer mikrowellenassistierten Reaktion erhalten wurde, eine praktikable Strategie mit hohen Ausbeuten entwickelt. Die Diels–Alder Reaktion solcher Porphyrine mit Tetraphenylcyclopentadienonen resultierte in einer statistischen Verteilung aller möglichen Hexaarylbenzol–Porphyrin Architekturen. Diese konnten mittels Säulenchromatographie von einander getrennt und charakterisiert werden. Bis dato unbekannt für Porphyrine zeigten alle Konjugate eine aussergewöhnlich hohe Intensität der Feststofffluoreszenz. Desweiteren konnten die statistischen Produktmischungen erfolgreich als Kalibrierungsreagenzien für die Massenspektrometrie eingesetzt werden. Die Oxidation der HAB–Porphyrine zu den entsprechenden HBC–Porphyrin–Konjugaten wurde mittels einer modifizierten Scholl–Reaktion, in der FeCl3 sowohl als Lewissäure als auch als Oxidationsmittel fungiert, erreicht. Hierbei stellte sich heraus, dass zusätzliche Mengen an Trifluoressigsäure die Löslichkeit der HAB–Porphyrin Systeme verbesserte und in einer besseren Umsetzung resultierte. Aufgrund dieser Methode konnte das bisher größte literaturbekannte PAH–substituierte Porphyrin in hohen Ausbeuten synthetisiert werden. Einhergehend mit dieser neuen Substanzklasse wurde ein einzigartiges Verhalten in der Gasphase in massenspektrometrischen Untersuchungen festgestellt. Dazu wurde eine Serie an meso-unsubstituierten HBC–Porphyrin–Konjugaten, A, trans-A2, A3 und A4, hergestellt, um deren Tendenz zur Bildung großer supramolekularer Architekturen zu untersuchen. Die Analyse dieser Reihe zeigte, dass die Ausbildung intermolekularer Architekturen auf die Pi-Pi-Wechselwirkungen der HBC–Einheiten zurückzuführen ist. Diese Erkenntnisse wurden außerdem mittels Röntgenstrukturanalyse einiger HBC–Porphyrin–Konjugate, welche das repetitive intermolekulare HBC–HBC–Packungsverhalten als dominierende Wechselwirkung im Feststoff aufzeigte, bestätigt. Photophysikalische Untersuchungen legten einen nahezu sofortigen Energietransfer im Pikosekunden–Bereich vom HBC zum zentralen Porphyrinkern offen. Dies lässt auf eine mögliche Anwendung der Konjugate als interessante Kandidaten für Lichtsammelkomplexe schließen. Desweiteren wurde das konzeptionelle Design von HBC–Porphyrin Gerüsten invertiert und HBC-zentrierte Bisporphyrin–Konjugate, substituiert in ortho, meta und para Stellung zueinander, synthetisiert. Diese Konjugate zeigten aussergewöhnliche spektroskopische Eigenschaften abhängig von der geometrischen Anordnung der Porphyrine zueinander. Im Detail konnte für das para-System eine Aufspaltung der Soret Bande festgestellt werden, wohingegen das ortho-Konjugat dies nicht zeigte und das meta-System als Intermediat dazwischen eine Schulter aufwies. Diese Beobachtungen sind ein Indiz für eine Exzitonkopplung, welche verstärkt im para-Konjugat aufgrund einer hinreichend guten Kommunkation über das Pi-System des HBC auftritt. Diese Fähigkeit findet in verminderter Form im meta und ortho-Konjugat statt, welche in der geringen bis hin zu keiner Aufspaltung der Soret Bande reflektiert wird. Die Familie der HBC–Porphyrine wurde um alkinylverbrückte Konjugate erweitert, die in Einzelmolekül-Leitfähigkeitsstudien an der Universität von Oxford untersucht wurden. Dabei stellte sich heraus, dass t-Butyl-substituierte HBCs keine geeignete Ankergruppe für Einzelmolekültransistoren darstellen.
Der dritte Teil der Arbeit widmet sich dem Feld der Tetraaryltetrabenzoporphyrine (TATBPs). Eine Evaluierung der Verwendbarkeit unterschiedlich maskierter Isoindole in der Synthese war daher unumgänglich. Aus dieser Studie ging das Bicyclo[2.2.2]octadien-annellierte Pyrrol als Isonindol-Synthon der Wahl hervor und ein generelles Protokoll für die Kondensation der TATBPs wurde entwickelt. Dieses Protokoll, welches grundlegend aus einer Modifikation der Lindesy Bedingungen besteht, überbietet alle bislang literaturbekannten Strategien für TATBPs in Handhabbarkeit und Ausbeuten. Mittels einer Gamma-ortho Cyclisierung wurden TATBPs als mögliche Fragmente des Graphyrins evaluiert. Hierfür wurden literaturbekannte Strategien für gespannte PAHs untersucht. Kollisionsinduzierte Dissoziation (CID) im Massenspektrometer zeigte, dass die Cyclodehydrofluorierung in der Fjord-Region effektiv zu einer achtfachen Gamma-ortho Cyclisierung führt. Somit konnte ein fundamentales Fragment des Graphyrins mittels Massenspektrometrie hergestellt und analysiert werden. In der nasschemischen Synthese stellte sich die palladiumkatalysierte intramolekulare direkte Arylierung von ortho chlorierten TATBPs als vielversprechendste Methode zur Schließung von bis zu vier C–C Bindungen heraus. Bindungsbildungen über vier Bindungen hinaus konnten allerdings nicht erzielt werden, da die verbleibenden Halogenatome während der Reaktion durch Wasserstoffatome ersetzt wurden. Im Bezug auf das größte synthetisierte Graphyrin Fragment wurde ein HBC–substituiertes Tetrabenzoporphyrin hergestellt. Unglücklicherweise zeigte es sich inert gegenüber einer Gamma-ortho Cyclisierung sowohl unter oxidativen als auch reduktiven Bedingungen.
Im letzten Teil der Arbeit wurde das wohl kleinste und fundamentalste Fragment von Graphyrin – [18]Annulen – untersucht. Eine neuartige dreistufige Synthesemethode, welche alle bisher publizierten Strategien für das [18]Annulen in Handhabbarkeit und Ausbeute übersteigt, konnte entwickelt werden. Dabei wurde erreicht, das Zwischenprodukt Hexadehydro[18]annulen, welches dafür bekannt ist sich im festen Zustand schnell zu zersetzen, in einer Matrix aus Benzol zu kristallisieren und mittels Röntgenstrukturanalyse zu charakterisieren. Die Benzol–Moleküle fungieren hierbei als effektiver Schutz gegenüber einer topologischen Polymerisation im Festkörper. Besonders aus der Kristallstrukturanalyse des [18]Annulens hervorzuheben ist der „Annulen-Loch“–Effekt, welcher zu einer sehr dichten Pi-Pi-Wechselwirkung im Festkörper führt. Dieser Effekt kann möglicherweise zu einem neuartigen Design von Halbleiter-Materialien mit hohen Ladungsträgermobilitäten im Festkörper dienen.
Die zahlreichen Einblicke jedes einzelnen Forschungsschwerpunktes auf dem Weg zu Graphyrin resultierten in fundamentalen und essentiellen Ergebnissen über Graphen–Porphyrin–Hybrid Systeme. Jedes Feld für sich – als Auszug aus dem Graphyrin – stellte sich jedoch auch als vielversprechendes, ertragreiches und eigentständiges Forschungsgebiet heraus, welches die weiterführende Untersuchung jenes Gesamtziels in unerschöpflicher und vielfacher Weise verspricht
Entwicklung und Untersuchung von neuartigen Porphyrin(oid)-Konjugat-Systemen
Within the first part of this thesis novel water-soluble donor-acceptor arrays built up by porphyrins and pyrene or perylene derivatives were designed via a copper(I)-catalysed 1,3-dipolar azide-alkyne cycloaddition reaction. For this purpose, suitable precursors, which were available by well-established synthetic literature procedures, were combined with newly developed complementary “click” counterparts. Thus, also the substance library of ortho-benzylic functionalised tetraarylporphyrins was further extended by valuable building blocks. To provide water-solubility to the synthesised porphyrin-pyrene and porphyrin-perylene conjugates, pyrene and perylene precursors with a second generation Newkome-dendron were employed in the “click” reaction. The dendron’s peripheral carboxylic groups were meant to bring solubility of the conjugates in aqueous media. Former approaches with comparable systems had shown that water-soluble conjugates are accessible by the acidic hydrolysis of the dendron’s ester groups to yield the corresponding carboxylic acids employing formic acid. However, often a mixture of metalloporphyrins and the corresponding free base porphyrins was obtained with zinc(II) porphyrins. In the course of this thesis an alternative route using trifluoroacetic acid for the abstraction of the peripheral esters of the dendrons was established avoiding the formation of a product mixture and yielding exclusively water-soluble free base porphyrins. In addition to the synthesis of these novel conjugates, they were characterised in details with respect to their spectroscopic and electrochemical properties employing NMR spectroscopy, UV/Vis absorption spectroscopy, steady-state and time-resolved fluorescence spectroscopy, femtosecond transient absorption spectroscopy as well as cyclic voltammetry, differential pulse voltammetry, and square wave voltammetry. A change of the photophysical and redox properties of the conjugates relative to the individual building blocks is expected to evolve in the case of intramolecular interaction processes such as energy or electron transfer between the porphyrins and the covalently linked pyrenes or perylenes. The analyses clearly revealed the existence of mutual interactions in the conjugate systems and, thus, evidenced that the covalent attachment of pyrenes and perylenes as some kind of “accessory” chromophores enriches the porphyrins’ photophysical properties yielding promising artificial molecular systems applicable for example for photoenergy conversion or photoelectronic operations. The task of the second part of this thesis was the development of novel photo-triggered drug release systems based on photosensitive pyropheophorbide building blocks. Starting from pyropheophorbide a, a pyropheophorbide-dimer was synthesised following strategies well known from peptide chemistry. Thereby, the beta amino acid (S)-3-amino-3-phenylpropionic acid was incorporated as indirectly photo-cleavable scission unit. Irradiation of the pyropheophorbide-based conjugate with visible light should generate singlet oxygen via sensitisation of molecular oxygen, which, in turn, was expected to induce a cascade of transformations finally resulting in the cleavage of the bond between the amino group and the benzyl substituent of the beta amino acid moiety and, thus, the release of the drug entities possessing photosensitising properties. The pyropheophorbide-dimer was investigated in the group of Prof. Dr. Beate Röder at the Humboldt University of Berlin with respect to its general behaviour and in particular to its photophysical and photosensitising properties to prove the concept of the employment of a singlet oxygen sensitive scission unit for the controlled light-triggered release of the pyropheophorbides as exemplary drug molecules. Although the experiments revealed that the benzylic amide does not seem to be sensitive towards the cleavage by singlet oxygen, valuable information was obtained for the synthesis of comparable, modified compounds acting as promising simple model systems for the design of high-order multiple-drug carrier architectures provided with a large number of biological active drug moieties that allow for the defined release of the individual drugs triggered by the application of light and, thus, without the need of any additional (chemical) agent.Im Rahmen des ersten Teils dieser Arbeit konnten verschiedene neuartige wasserlösliche Donor-Akzeptor-Systeme bestehend aus Porphyrinen und Pyren- beziehungsweise Perylen-Derivaten über eine Kupfer(I)salz katalysierte 1,3-dipolare Cykloadditionsreaktion erfolgreich aufgebaut werden. Für die Darstellung der Konjugat-Systeme wurden geeignete bereits literaturbekannte Bausteine mit innerhalb dieser Arbeit neu entwickelten „Click“-Gegenstücken kombiniert. Dadurch konnte zusätzlich die Substanz-Bibliothek von ortho-funktionalisierten Tetraarylporphyrinen noch um wertvolle Bausteine erweitert werden. Um wasserlösliche Porphyrin-Pyren- und Porphyrin-Perylen-Konjugate zu erhalten, wurden Pyren- und Perylen-Systeme in welche zuvor ein Newkome-Dendron zweiter Generation eingebaut wurde als Ausgangsverbindungen für die „Click“-Reaktion eingesetzt. Dabei sollten die Carbonsäurefunktionen der entschützten Dendronen die Löslichkeit der Konjugate in wässrigem Medium vermitteln. Bereits in vorangegangenen Arbeiten mit vergleichbaren Systemen wurden wasserlösliche Verbindungen durch eine säurekatalysierte Hydrolyse der Ester-Gruppen der Dendronen zu den entsprechenden Carbonsäuren unter Verwendung von Ameisensäure erhalten. Unter diesen Bedingungen wurde beim Einsatz von Zink(II)-Porphyrinen allerdings häufig die Bildung einer Mischung aus noch metalliertem und bereits demetalliertem Porphyrin beobachtet. Hier konnte nun eine alternative Verfahrensweise entwickelt werden mit Hilfe derer die Entstehung eines Produktgemisches vermieden wird. Setzt man für die Entschützungsreaktion Trifluoressigsäure ein, so findet eine quantitative Demetallierung statt und man erhält ausschließlich die freie Base als wasserlösliches Produkt. Neben der Darstellung dieser neuartigen wasserlöslichen Donor-Akzeptor-Systeme war ein weiterer Aspekt dieser Arbeit deren eingehende Untersuchung in Hinblick auf ihre spektroskopischen sowie elektrochemischen Eigenschaften. Zur Charakterisierung wurden dabei NMR-, UV/Vis-Absorptions-, stationäre und zeitaufgelöste Fluoreszenz-, differentielle Transienten-Absorptionsspektroskopie sowie Differential-Puls-, Square-Wave- und zyklische Voltammetrie herangezogen. Im Falle von intramolekularen Wechselwirkungen zwischen den einzelnen Komponenten der Konjugat-Systeme, wie z.B. Übertragung von Energie oder Elektronen, sollten sich diese in einer Veränderung der photophysikalischen sowie elektrochemischen Eigenschaften gegenüber den isolierten Einzelbausteinen widerspiegeln. Die Untersuchungen haben deutlich gezeigt, dass Wechselwirkungsprozesse innerhalb der hergestellten Konjugate auftreten und belegen damit, dass durch die kovalente Anknüpfung von Pyrenen oder Perylenen an Porphyrine als zusätzliche chromophore Einheit die photophysikalischen Eigenschaften der Porphyrine noch erweitert und optimiert werden können. Auf diese Art und Weise ist die Darstellung von synthetischen molekularen Strukturen möglich mit potentieller Anwendbarkeit in künstlichen Photosynthese-Systemen z.B. zur Erzeugung von elektrischer Energie oder in photoelektronischen Prozessen. Ziel des zweiten Teils dieser Arbeit war die Entwicklung einer neuen Art von Verbindungen, die auf die Bestrahlung mit sichtbarem oder infrarotem Licht hin photosensible Wirkstoffmoleküle, in diesem Fall auf Pyropheophorbiden basierende Einheiten, freisetzen. Unter Verwendung von gut ausgearbeiteten bereits aus der Peptid-Chemie bekannten Synthese-Strategien konnte ausgehend von Pyropheophorbid a ein Pyropheophorbid-Dimer erfolgreich hergestellt werden. Dabei wurde die beta-Aminosäure beta-Phenylalanin als eine Art indirekte „Bruchstelle“ in die Systeme eingebaut. Bei Bestrahlung des Dimers mit sichtbarem Licht wurde erwartet, dass die Pyropheophorbid-Einheiten zunächst Singulett-Sauerstoff generieren, welcher dann anschließend zu einer Spaltung der aktivierten C-N-Bindung innerhalb der Aminosäure-Einheit führen und somit die gezielte Freisetzung der photosensiblen Wirkstoffmoleküle bewirken sollte. Hierzu wurde in der Arbeitsgruppe von Prof. Dr. Beate Röder an der Humboldt-Universität zu Berlin das hergestellte Dimer-System auf seine photophysikalischen Eigenschaften hin untersucht, vor allem in Hinblick auf sein Verhalten gegenüber Singulett-Sauerstoff, um so die Idee eine gegenüber Singulett-Sauerstoff labile „Bruchstelle“ für die gezielte und kontrollierte Freisetzung der Pyropheophorbid-Einheiten (als eine Art Model-Wirkstoffsystem) auf ihre praktische Anwendbarkeit hin zu überprüfen. Obwohl die Ergebnisse der Messungen schlussendlich vermuten lassen, dass die benzylische Amid-Funktion des Dimer-Systems gegenüber Singulett-Sauerstoff stabil zu sein scheint und somit in dieser Hinsicht offensichtlich keine geeignete „Bruchstelle“ darstellt, konnten dennoch wertvolle Informationen für die zukünftige Entwicklung vielversprechender, entsprechend modifizierter Model-Systeme gewonnen werden
Pyrrolhaltige Hexaarylbenzole für die Porphyrinoid-Synthese
This work focuses on newly discovered pyrrole substituted hexaarylbenzenes, prepaired from their appropriate pyrrole containing acetylene derivatives. These building blocks represent relevant precursors for a novel strategy synthesizing carbon rich porphycenes.Diese Arbeit beschäftigt sich mit neu entdeckten pyrrolsubstituierten Hexaarylbenzolen, die aus ihren entsprechenden pyrrolhaltigen Acetylenderivaten hergestellt werden. Diese Bausteine stellen wichtige Vorstufen für einen neuartigen Weg zur Synthese kohlenstoffreicher Porphycene dar
