Design and Development of COX-II Inhibitors: Current Scenario and Future Perspective

Published by American Chemical Society.Innate inflammation beyond a threshold is a significant problem involved in cardiovascular diseases, cancer, and many other chronic conditions. Cyclooxygenase (COX) enzymes are key inflammatory markers as they catalyze prostaglandins production and are crucial for inflammation processes. While COX-I is constitutively expressed and is generally involved in “housekeeping” roles, the expression of the COX-II isoform is induced by the stimulation of different inflammatory cytokines and also promotes the further generation of pro-inflammatory cytokines and chemokines, which affect the prognosis of various diseases. Hence, COX-II is considered an important therapeutic target for drug development against inflammation-related illnesses. Several selective COX-II inhibitors with safe gastric safety profiles features that do not cause gastrointestinal complications associated with classic anti-inflammatory drugs have been developed. Nevertheless, there is mounting evidence of cardiovascular side effects from COX-II inhibitors that resulted in the withdrawal of market-approved anti-COX-II drugs. This necessitates the development of COX-II inhibitors that not only exhibit inhibit potency but also are free of side effects. Probing the scaffold diversity of known inhibitors is vital to achieving this goal. A systematic review and discussion on the scaffold diversity of COX inhibitors are still limited. To address this gap, herein we present an overview of chemical structures and inhibitory activity of different scaffolds of known COX-II inhibitors. The insights from this article could be helpful in seeding the development of next-generation COX-II inhibitors.Peer reviewe
Isolation and Identification of Crude Triacontanol from Rice Bran Wax
In present investigation crude triacontanol was isolated and identified from rice bran wax. Triacontanol was isolated by saponification and extraction method. The obtained mixture is crude Triacontanol. It was analyzed by Gas Chromatography (GC) and melting point method. The intent of NPP-2000 is to eliminate unmet contraceptive needs by providing high quality reproductive healthcare. In particular, the NPP-2000 aims to address flaws in healthcare infrastructure and to achieve a total fertility rate of 2.1 births per woman by 2010. Unfortunately, the implementation difficulties of past years persist in the era of NPP-2000. Indian families are subject to poorly-trained healthcare personnel and insufficient medical supplies, among other setbacks. and qualitative studies, the following analysis exposes widespread variation in the quality of family planning practice. Effect of Chitosan on Okra (Abelmoschus esculentus (L.) Moench) Seed Germination
Present paper deals with the effect of chitosan at different concentration on seed germination of okra. Investigating Self-Efficacy as a Mediator of Peer Violence and Dating Violence Perpetration Among High-Risk Adolescents
Background: Dating violence is a significant public health problem. This study explored the potential mediation of self-efficacy of nonviolent conflict negotiation in the association between peer violence and dating violence among youth in a high-risk community.
Methods: This study used cross-sectional data from the Youth Violence Survey: Linkages among Different Forms of Violence study funded by CDC. Data were collected in in 2004 and analyzed in 2016. The sample comprised 4,131 public school students in the seventh, ninth, eleventh, and twelfth year in an urban school district with high crime and poverty rates who completed a self-report questionnaire following parental consent and student assent. The analytic sample was restricted to participants who dated in the past year (n= 2,888). A mediation analysis was conducted using both the Causal Steps method and the Sobel Test to determine if self-efficacy in nonviolent conflict negotiation partially mediated the association between peer violence perpetration and dating violence perpetration.
Results: Results supported the study hypothesis that self-efficacy partially mediated the relationship between peer violence perpetration and dating violence perpetration. Even after controlling for dating violence victimization, peer violence victimization, and peer violence perpetration, self-efficacy contributed significantly to the model explaining dating violence perpetration and lowered the dating violence perpetration variation for which peer violence perpetration accounted (total effect: β= .930 and p\u3c.001, direct effect: β= .841 and p\u3c.001) . Self-efficacy decreased the odds by .674 of participants who had perpetrated peer violence from perpetrating dating violence (p\u3c.001). The significance of the mediation effect was confirmed by the Sobel Test (z= 3.917, 95% CI).
Conclusion: Self-efficacy in nonviolent conflict negotiation could be an effective intervening factor for dating violence perpetration, contributing to stopping the cycle of violence among youth in high-risk communities

Social Media Analytics - Digital Footprints
In this work we describe an approach to distinguish real and impostor/compromised accounts on social media. Compromising a user's social media account is not only a breach of security, but can also lead to dissemination of misinformation at a fast pace on social media. There have been several such high profile attacks recently, including on Twitter feeds of AP, CBS, and Delta Airlines. A fake account for the Prime Minister's Office (PMO) in India was used to spread malicious rumors last year. Our approach builds a digital social footprint of users using both the content of their tweets and the structure of their social network. We analyze the real time content of users' tweets and compare it with information about them from reliable sources on the web (newspapers / news channels etc.)to compute a similarity metric. We also compute a metric based on the social network analysis of the users -- who connects to them, who they are connected with, and how central they are in their network. We have shown how these metrics combined judiciously can detect fake accounts for not just well known people such as President Obama, but also for lesser known people and organizations. We also show promising initial results on how this approach can be used to detect an account which has been hacked
